Differentiation of marrow stromal cells into photoreceptors in the rat eye
Retinal degenerations and dystrophies are the major causes of genetically inherited blindness that are characterized by the apoptotic death of the photoreceptor cell layer of the retina. To date, no treatment exists for these diseases and only recently have they been considered as candidates for gene and stem cell therapies. Here we report the ability of adult CD90+ marrow stromal cells (MSCs) to be induced by activin A, taurine, and EGF into cells (20-32%) expressing photoreceptor-specific markers rhodopsin, opsin, and recoverin in vitro. CD90+ cells were either transduced with recombinant adeno-associated virus expressing green fluorescent protein (GFP) or bromodeoxyuridine (BrdU) labeled and then injected into the subretinal space of adult Royal College of Surgeons rats. Fundus photography and angiography showed no adverse effects of CD90+ MSC transplantation. GFP-expressing cells or BrdU-positive cells covered ∼30% of the entire retinal area. By 2 weeks after injection, CD90+ MSCs integrated into the host retina, forming structures similar to the photoreceptor layer and expressed a photoreceptor-specific marker. No teratoma formation was observed in the recipient retina. The subretinally delivered CD90+ MSCs did not stain for proliferating cell nuclear antigen, indicating that they primarily undergo differentiation rather than proliferation. In addition, we established that transplanted cells can attract synaptic vesicles and hence are potentially capable of signal transduction. This study demonstrates for the first time the partial differentiation of adult CD90+ MSCs into photoreceptors in vitro and in vivo. Our results establish a proof of concept for CD90+ MSC differentiation with autologous transplantation, which may provide a promising therapeutic strategy for the treatment of some forms of genetically inherited retinal degenerations
Submanifolds in five-dimensional pseudo-Euclidean spaces and four-dimensional FRW universes
Equations for submanifolds, which correspond to embeddings of the
four-dimensional FRW universes in five-dimensional pseudo-Euclidean spaces, are
presented in convenient form in general case. Several specific examples are
considered.Comment: 7 pages, LaTeX, the mathematical part of this paper is based on the

The Physical Principles of Quantum Mechanics. A critical review
The standard presentation of the principles of quantum mechanics is
critically reviewed both from the experimental/operational point and with
respect to the request of mathematical consistency and logical economy. A
simpler and more physically motivated formulation is discussed. The existence
of non commuting observables, which characterizes quantum mechanics with
respect to classical mechanics, is related to operationally testable
complementarity relations, rather than to uncertainty relations. The drawbacks
of Dirac argument for canonical quantization are avoided by a more geometrical
approach.Comment: Bibliography and section 2.1 slightly improve
QCD Strings as Constrained Grassmannian Sigma Model:
We present calculations for the effective action of string world sheet in R3
and R4 utilizing its correspondence with the constrained Grassmannian sigma
model. Minimal surfaces describe the dynamics of open strings while harmonic
surfaces describe that of closed strings. The one-loop effective action for
these are calculated with instanton and anti-instanton background, reprsenting
N-string interactions at the tree level. The effective action is found to be
the partition function of a classical modified Coulomb gas in the confining
phase, with a dynamically generated mass gap.Comment: 22 pages, Preprint: SFU HEP-116-9
Regulatory T cells are paramount effectors in progesterone regulation of embryo implantation and fetal growth
Published: June 8, 2023Progesterone (P4) is essential for embryo implantation, but the extent to which the pro-gestational effects of P4 depend on the maternal immune compartment is unknown. Here, we investigate whether regulatory T cells (Treg cells) act to mediate luteal phase P4 effects on uterine receptivity in mice. P4 antagonist RU486 administered to mice on days 0.5 and 2.5 post coitum (dpc) to model luteal phase P4 deficiency caused fewer CD4+Foxp3+ Treg cells and impaired Treg functional competence, along with dysfunctional uterine vascular remodeling and perturbed placental development in mid-gestation. These effects were linked with fetal loss and fetal growth restriction, accompanied by a Th1/CD8-skewed T cell profile. Adoptive transfer at implantation of Treg cells - but not T conventional (Tconv) cells - alleviated fetal loss and fetal growth restriction by mitigating adverse effects of reduced P4 signaling on uterine blood vessel remodeling and placental structure, and restoring maternal T cell imbalance. These findings demonstrate an essential role for Treg cells in mediating P4 effects at implantation, and indicate that Treg cells are a sensitive and critical effector mechanism through which P4 drives uterine receptivity to support robust placental development and fetal growth.Ella S. Tensor-scalar gravity and binary-pulsar experiments
Some recently discovered nonperturbative strong-field effects in
tensor-scalar theories of gravitation are interpreted as a scalar analog of
ferromagnetism: "spontaneous scalarization". This phenomenon leads to very
significant deviations from general relativity in conditions involving strong
gravitational fields, notably binary-pulsar experiments. Contrary to
solar-system experiments, these deviations do not necessarily vanish when the
weak-field scalar coupling tends to zero. We compute the scalar "form factors"
measuring these deviations, and notably a parameter entering the pulsar timing
observable gamma through scalar-field-induced variations of the inertia moment
of the pulsar. An exploratory investigation of the confrontation between
tensor-scalar theories and binary-pulsar experiments shows that nonperturbative
scalar field effects are already very tightly constrained by published data on
three binary-pulsar systems. We contrast the probing power of pulsar
experiments with that of solar-system ones by plotting the regions they exclude
in a generic two-dimensional plane of tensor-scalar theories.Comment: 35 pages, REVTeX 3.0, uses epsf.tex to include 9 Postscript figure
Tribological performance of an H-DLC coating prepared by PECVD
Carbon-based coatings are of wide interest due to their application in machine elements subjected to continuous contact where fluid lubricant films are not permitted. This paper describes the tribological performance under dry conditions of duplex layered H-DLC coating sequentially deposited by microwave excited plasma enhanced chemical vapour deposition on AISI 52100 steel. The architecture of the coating comprised Cr, WC, and DLC (a-C:H) with a total thickness of 2.8 μm and compressive residual stress very close to 1 GPa. Surface hardness was approximately 22 GPa and its reduced elastic modulus around 180 GPa. Scratch tests indicated a well adhered coating achieving a critical load of 80 N. The effect of normal load on the friction and wear behaviours were investigated with steel pins sliding against the actual coating under dry conditions at room temperature (20 ± 2°C) and 35-50% RH. The results show that coefficient of friction of the coating decreased from 0.21 to 0.13 values with the increase in the applied loads (10-50 N). Specific wear rates of the surface coating also decrease with the increase in the same range of applied loads. Maximum and minimum values were 14 × 10-8 and 5.5 × 10-8 mm-3/N m, respectively. Through Raman spectroscopy and electron microscopy it was confirmed the carbon-carbon contact, due to the tribolayer formation on the wear scars of the coating and pin. In order to further corroborate the experimental observations regarding the graphitisation behaviour, the existing mathematical relationships to determine the graphitisation temperature of the coating/steel contact as well as the flash temperature were used
Testing gravity to second post-Newtonian order: a field-theory approach
A new, field-theory-based framework for discussing and interpreting tests of
gravity, notably at the second post-Newtonian (2PN) level, is introduced.
Contrary to previous frameworks which attempted at parametrizing any
conceivable deviation from general relativity, we focus on the best motivated
class of models, in which gravity is mediated by a tensor field together with
one or several scalar fields. The 2PN approximation of these
"tensor-multi-scalar" theories is obtained thanks to a diagrammatic expansion
which allows us to compute the Lagrangian describing the motion of N bodies. In
contrast with previous studies which had to introduce many phenomenological
parameters, we find that the 2PN deviations from general relativity can be
fully described by only two new 2PN parameters, epsilon and zeta, beyond the
usual (Eddington) 1PN parameters beta and gamma. It follows from the basic
tenets of field theory, notably the absence of negative-energy excitations,
that (beta-1), epsilon and zeta (as well as any new parameter entering higher
post-Newtonian orders) must tend to zero with (gamma-1). It is also found that
epsilon and zeta do not enter the 2PN equations of motion of light. Therefore,
light-deflection or time-delay experiments cannot probe any theoretically
motivated 2PN deviation from general relativity, but they can give a clean
access to (gamma-1), which is of greatest significance as it measures the basic
coupling strength of matter to the scalar fields. Because of the importance of
self-gravity effects in neutron stars, binary-pulsar experiments are found to
constitute a unique testing ground for the 2PN structure of gravity. A
simplified analysis of four binary pulsars already leads to significant
constraints: |epsilon| < 7x10^-2, |zeta| < 6x10^-3.Comment: 63 pages, 11 figures.ps.tar.gz.uu, REVTeX 3.
