Metabolic engineering of apple by overexpression of the MdMyb10 gene
Flavonoids are low-molecular-weight phenolic compounds that are widely distributed in the plant kingdom. They have different roles in plant resistance to biotic and abiotic stresses. The transcription factor gene MdMyb10 (Gene Bank: DQ267896) was introduced into two apple (Malus domestica Borkh.) cultivars i.e. ‘Holsteiner Cox (HC)’ and ‘Gala’ via Agrobacterium-mediated transformation. The regenerated shoots were selected on kanamycin containing media. The presence of additional MdMyb10 gene in putative shoots was confirmed by PCR, RT-PCR and Southern blotting. Expression level of introduced MdMyb10 gene was analyzed by quantitative real time PCR. The results confirmed a dramatic increase in overexpression of MdMyb10 in the transgenic plants, up to 1261 and 847-folds for cultivars Holsteiner Cox and Gala, respectively compared to non-transformed negative control plants. HPLC-MS was used to determine the levels of different flavonoid compounds in both non-transgenic and transgenic plants. In MdMyb10 ‘HC’ transgenic plants, some of the polyphenols analyzed were enhanced while others were reduced in comparison to their levels in the non-transgenic plants. On the other hand, all of the analyzed polyphenol classes were induced in MdMyb10 ‘Gala’ transgenic plants in comparison to their levels in the non-transgenic plants. In the present study, the flavonoid pathway was successfully modified in apple by overexpressing the MdMyb10 transcription factor to validate the hypothesis of increased effect on plant disease resistance.Islamic Development Ban
Density Dependent Hadron Field Theory
A fully covariant approach to a density dependent hadron field theory is
presented. The relation between in--medium NN interactions and
field--theoretical meson--nucleon vertices is discussed. The medium dependence
of nuclear interactions is described by a functional dependence of the
meson--nucleon vertices on the baryon field operators. As a consequence, the
Euler--Lagrange equations lead to baryon rearrangement self--energies which are
not obtained when only a parametric dependence of the vertices on the density
is assumed. It is shown that the approach is energy--momentum conserving and
thermodynamically consistent. Solutions of the field equations are studied in
the mean--field approximation. Descriptions of the medium dependence in terms
of the baryon scalar and vector density are investigated. Applications to
infinite nuclear matter and finite nuclei are discussed. Density dependent
coupling constants obtained from Dirac--Brueckner calculations with the Bonn
NN-potentials are used. Results from Hartree calculations for energy spectra,
binding energies and charge density distributions of , and
are presented. Comparisons to data strongly support the importance
of rearrangement in a relativistic density dependent field theory. Most
striking is the simultanuous improvement of charge radii, charge densities and
binding energies. The results indicate the appearance of a new "Coester line"

Emergence of quasi-metallic state in disordered 2D electron gas due to strong interactions
The interrelation between disorder and interactions in two dimensional
electron liquid is studied beyond weak coupling perturbation theory. Strong
repulsion significantly reduces the electronic density of states on the Fermi
level. This makes the electron liquid more rigid and strongly suppresses
elastic scattering off impurities. As a result the weak localization, although
ultimately present at zero temperature and infinite sample size, is
unobservable at experimentally accessible temperature at high enough densities.
Therefore practically there exists a well defined metallic state. We study
diffusion of electrons in this state and find that the diffusion pole is
significantly modified due to "mixture" with static photons similar to the
Anderson - Higgs mechanism in superconductivity. As a result several effects
stemming from the long range nature of diffusion like the Aronov - Altshuler
logarithmic corrections to conductivity are less pronounced.Comment: to appear in Phys. Rev.
Lubricated revolute joints in rigid multibody systems
The main purpose of this work is to present a general methodology for modeling lubricated revolute joints in constrained rigid multibody systems. In the dynamic analysis of journal-bearings, the hydrodynamic forces, which include both squeeze and wedge effects, generated by the lubricant fluid, oppose the journal motion. The hydrodynamic forces are obtained by integrating the pressure distribution evaluated with the aid of Reynolds’ equation, written for the dynamic regime. The hydrodynamic forces built up by the lubricant fluid are evaluated from the system state variables and included into the equations of motion of the multibody system. Numerical examples are presented in order to demonstrate the use of the methodologies and procedures described in this work.Fundação para a Ciência e a Tecnologia (FCT
The effect of autonomy, training opportunities, age and salaries on job satisfaction in the South East Asian retail petroleum industry
South East Asian petroleum retailers are under considerable pressure to improve service quality by reducing turnover. An empirical methodology from this industry determined the extent to which job characteristics, training opportunities, age and salary influenced the level of job satisfaction, an indicator of turnover. Responses are reported on a random sample of 165 site employees (a 68% response rate) of a Singaporean retail petroleum firm. A restricted multivariate regression model of autonomy and training opportunities explained the majority (35.4%) of the variability of job satisfaction. Age did not moderate these relationships, except for employees >21 years of age, who reported enhanced job satisfaction with additional salary. Human Capital theory, Life Cycle theory and Job Enrichment theory are invoked and explored in the context of these findings in the South East Asian retail petroleum industry. In the South East Asian retail petroleum industry, jobs providing employees with the opportunity to undertake a variety of tasks that enhanced the experienced meaningfulness of work are likely to promote job satisfaction, reduce turnover and increase the quality of service
Oncoplastic Breast Consortium consensus conference on nipple-sparing mastectomy
Purpose Indications for nipple-sparing mastectomy (NSM) have broadened to include the risk reducing setting and locally
advanced tumors, which resulted in a dramatic increase in the use of NSM. The Oncoplastic Breast Consortium consensus
conference on NSM and immediate reconstruction was held to address a variety of questions in clinical practice and research
based on published evidence and expert panel opinion.
Methods The panel consisted of 44 breast surgeons from 14 countries across four continents with a background in gynecology, general or reconstructive surgery and a practice dedicated to breast cancer, as well as a patient advocate. Panelists
presented evidence summaries relating to each topic for debate during the in-person consensus conference. The iterative
process in question development, voting, and wording of the recommendations followed the modified Delphi methodology.
Results Consensus recommendations were reached in 35, majority recommendations in 24, and no recommendations in the
remaining 12 questions. The panel acknowledged the need for standardization of various aspects of NSM and immediate
reconstruction. It endorsed several oncological contraindications to the preservation of the skin and nipple. Furthermore, it
recommended inclusion of patients in prospective registries and routine assessment of patient-reported outcomes. Considerable heterogeneity in breast reconstruction practice became obvious during the conference.
Conclusions In case of conflicting or missing evidence to guide treatment, the consensus conference revealed substantial
disagreement in expert panel opinion, which, among others, supports the need for a randomized trial to evaluate the safest
and most efficacious reconstruction techniques
Nucleosomes in gene regulation: theoretical approaches
This work reviews current theoretical approaches of biophysics and
bioinformatics for the description of nucleosome arrangements in chromatin and
transcription factor binding to nucleosomal organized DNA. The role of
nucleosomes in gene regulation is discussed from molecular-mechanistic and
biological point of view. In addition to classical problems of this field,
actual questions of epigenetic regulation are discussed. The authors selected
for discussion what seem to be the most interesting concepts and hypotheses.
Mathematical approaches are described in a simplified language to attract
attention to the most important directions of this field
