    The Neural Encoding of Cocaine-Induced Devaluation in The Ventral Pallidum

    Cocaine experience affects motivation structures such as the nucleus accumbens (NAc) and its major output target, the ventral pallidum (VP). Previous studies demonstrated that both NAc activity and hedonic responses change reliably as a taste cue comes to predict cocaine availability. Here we extended this investigation to examine drug-experience induced changes in hedonic encoding in the VP. VP activity was first characterized in adult male Sprague–Dawley rats in response to intraoral infusions of palatable saccharin and unpalatable quinine solutions. Next, rats received 7 daily pairings of saccharin that predicted either a cocaine (20 mg/kg, ip) or saline injection. Finally, the responses to saccharin and quinine were again assessed. Of 109 units recorded in 11 rats that received saccharin–cocaine pairings, 71% of responsive units significantly reduced firing rate during saccharin infusions and 64% increased firing rate during quinine exposure. However, as saccharin came to predict cocaine, and elicited aversive taste reactivity, VP responses changed to resemble quinine. After conditioning, 70% of saccharin-responsive units increased firing rate. Most units that encoded the palatable taste (predominantly reduced firing rate) were located in the anterior VP, while most units that were responsive to aversive tastes were located in the posterior VP. This study reveals an anatomical complexity to the nature of hedonic encoding in the VP

    Serum and synovial fluid lipidomic profiles predict obesity-associated osteoarthritis, synovitis, and wound repair

    High-fat diet-induced obesity is a major risk factor for osteoarthritis (OA) and diminished wound healing. The objective of this study was to determine the associations among serum and synovial fluid lipid levels with OA, synovitis, adipokine levels, and wound healing in a pre-clinical obese mouse model of OA. Male C57BL/6 J mice were fed either a low-fat (10% kcal) or one of three high-fat (HF, 60% kcal) diets rich in saturated fatty acids (SFAs), ω-6 or ω-3 polyunsaturated FAs (PUFAs). OA was induced by destabilization of the medial meniscus. Mice also received an ear punch for evaluating wound healing. Serum and synovial fluid were collected for lipidomic and adipokine analyses. We demonstrated that the serum levels of ω-3 PUFAs were negatively correlated with OA and wound size, but positively correlated with adiponectin levels. In contrast, most ω-6 PUFAs exhibited positive correlations with OA, impaired healing, and inflammatory adipokines. Interestingly, levels of pentadecylic acid (C15:0, an odd-chain SFA) and palmitoleic acid were inversely correlated with joint degradation. This study extends our understanding of the links of FAs with OA, synovitis and wound healing, and reports newly identified serum and synovial fluid FAs as predictive biomarkers of OA in obesity

    Stress and coping skills are among the most essential components of the mental health counseling field. The use of coping skills (e.g., meditation, physical activities, appropriate uses of leisure) has been identified as an effective strategy for stress management. Adventure therapy has emerged as a modality that can positively augment other therapeutic approaches by improving coping skills and assisting clients in managing stress. As with all therapies, a positive working alliance has been found to be important toward achieving clinical outcomes. This study explored how adventure therapy enhanced learned coping strategies for stress and improved therapeutic alliance. Outcomes from this exploratory research highlighted the potential of adventure therapy to decrease stress, increase coping skills, and build therapeutic rapport with the therapist

    Characterization of the Hamamatsu R11410-10 3-Inch Photomultiplier Tube for Liquid Xenon Dark Matter Direct Detection Experiments

    To satisfy the requirements of the next generation of dark matter detectors based on the dual phase TPC, Hamamatsu, in close collaboration with UCLA, has developed the R11410-10 photomultipler tube. In this work, we present the detailed tests performed on this device. High QE (>30%) accompanied by a low dark count rate (50 Hz at 0.3 PE) and high gain (10^7) with good single PE resolution have been observed. A comprehensive screening measurement campaign is ongoing while the manufacturer quotes a radioactivity of 20 mBq/PMT. These characteristics show the R11410-10 to be particularly suitable for the forthcoming zero background liquid xenon detectors.Comment: 19 pages, 18 figure

    Expected Sensitivity to Galactic/Solar Axions and Bosonic Super-WIMPs based on the Axio-electric Effect in Liquid Xenon Dark Matter Detectors

    We present systematic case studies to investigate the sensitivity of axion searches by liquid xenon detectors, using the axio-electric effect (analogue of the photoelectric effect) on xenon atoms. Liquid xenon is widely considered to be one of the best target media for detection of WIMPs (Weakly Interacting Massive Particles which may form the galactic dark matter) using nuclear recoils. Since these detectors also provide an extremely low radioactivity environment for electron recoils, very weakly-interacting low-mass particles (< 100 keV/c^2), such as the hypothetical axion, could be detected as well - in this case using the axio-electric effect. Future ton-scale liquid Xe detectors will be limited in sensitivity only by irreducible neutrino background (pp-chain solar neutrino and the double beta decay of 136Xe) in the mass range between 1 and 100 keV/c^2. Assuming one ton-year of exposure, galactic axions (as non-relativistic dark matter) could be detected if the axio-electric coupling g_Ae is greater than 10^-14 at 1 keV/c^2 (or $10^-13 at 100 keV/c^2). Below a few keV/c^2, and independent of the mass, a solar axion search would be sensitive to a coupling g_Ae ~ 10^-12. This limit will set a stringent upper bound on axion mass for the DFSV and KSVZ models for the mass ranges m_A < 0.1 eV/c^2 and < 10 eV/c^2, respectively. Vector-boson dark matter could also be detected for a coupling constant alpha'/alpha > 10^-33 (for mass 1 keV/c^2) or > 10^-27 (for mass 100 keV/c^2).Comment: 17 pages, 10 figure

    Microphotonic Harsh Environment Sensors for Clean Fuel and Power Generation

    Track IV: Materials for Energy ApplicationsIncludes audio file (19 min.)Low cost, reliable, in-situ sensors are highly desired for advanced process control and lifecycle management in various power and fuel systems. Many energy generation processes involve harsh conditions throughout the operation that requires monitoring to assist in attaining and maintaining the goals of high efficiency and high environmental performance. General measurements of interest include temperature, strain, pressure, gas compositions, and trace contaminants/pollutants. Unfortunately, most existing sensors are incapable of operating directly in the harsh environment of typical power and fuel systems involving high temperature and high pressure with presence of particulates and corrosive atmosphere. Funded by DoE/NETL, our group has been developing various novel microphotonic sensors for monitoring physical and chemical parameters under hostile conditions. The demonstrated sensors include the miniaturized inline fiber Fabry-Perot interferometer (FPI) fabricated by one-step femtosecond laser micromaching, the long period fiber grating (LPFG) fabricated by CO2 laser irradiations, the fiber inline core-cladding mode interferometers (CMMI), and the LPFG coupled CMMI sensors. These sensors can be directly used for the measurements of various physical parameters such as temperature, pressure and strain in a high temperature (tested up to 1100 degree C) harsh environment are presented. In addition, when coated with a thin layer of gas sensitive thin film (e.g., doped crystalline ceramic nanofilm), they can be used for measurements of various hot gases such as hydrogen, carbon dioxide, carbon monoxide, and hydrogen sulfide in high temperatures. With demonstrated advantages of small size, lightweight, immunity to electromagnetic interference, resistance to chemical corrosion, high sensitivity, remote operation capability, robustness and dependable performance in a hostile environment, these microphotonic sensor may find broad applications for process control and optimization in various fuel/power systems such as coal gasification, advanced engines, oil/gas extraction, fuel cell operation, coal/geothermal/wind/nuclear-based power generations, etc. We hope that this presentation will convey our interest in teaming up with UM researchers and industry partners to collectively explore future opportunities

    A Game of Attribute Decomposition for Software Architecture Design

    Attribute-driven software architecture design aims to provide decision support by taking into account the quality attributes of softwares. A central question in this process is: What architecture design best fulfills the desirable software requirements? To answer this question, a system designer needs to make tradeoffs among several potentially conflicting quality attributes. Such decisions are normally ad-hoc and rely heavily on experiences. We propose a mathematical approach to tackle this problem. Game theory naturally provides the basic language: Players represent requirements, and strategies involve setting up coalitions among the players. In this way we propose a novel model, called decomposition game, for attribute-driven design. We present its solution concept based on the notion of cohesion and expansion-freedom and prove that a solution always exists. We then investigate the computational complexity of obtaining a solution. The game model and the algorithms may serve as a general framework for providing useful guidance for software architecture design.
