Muscle Tension Induced after Learning Enhances Long-Term Narrative and Visual Memory in Healthy Older Adults
Arousing events are better remembered than mundane events. Indeed, manipulation of arousal, such as by muscle tension, can influence memory even when it occurs shortly after learning. Indeed, our founding study showed this approach can raise delayed memory performance in older adults to a level comparable to that of unaided young adults. Yet, systematic studies, especially those investigating different modalities or types of memory, have not been done. This study investigated the effects of a brief bout of isometric exercise via handgrip on narrative and visuospatial episodic memory in healthy elders. Forty-seven participants completed the Logical Memory subtest of the Wechsler Memory Scales III (LM) and the Benton Visual Retention Test (BVRT), followed alternately by no treatment and by moderately squeezing a sand-filled latex ball for 1-min (counterbalanced order and test forms). Isometric exercise significantly increased both positive and negative affect ratings. Retention was tested 2 weeks later. Delayed recall and recognition of LM was enhanced by arousal relative to control, as was recognition of the BVRT. The results extend past findings that muscle tension induced after learning modulates memory consolidation, extending findings in elders to suggest that a simple form of isometric exercise can have practical effects, such as aiding memory for stories and images
Screening of organically based fungicides for apple scab (Venturia inaequalis) control and a histopathological study of the mode of action of a resistance inducer.
A range of possible substitutes for copper-based fungicides for control of apple scab (Venturia inaequalis) in organic growing were tested in laboratory and growth chamber experiments in the Danish project StopScab (2002-2004). Eighteen crude plant extracts, 19 commercial plant-based products and 6 miscellaneous compounds were tested for their ability to reduce scab symptoms on apple seedlings. Most of the compounds were also tested for their effect on conidium germination on glass slides. Fourteen of the crude plant extracts, 13 of the commercial plant products and 5 of the miscellaneous compounds showed promising control efficacies when used either preventively or curatively in the plant assay. A histopathological study was carried out on the mode of action of the resistance inducer, acibenzolar-S-methyl (ASM), which reduced scab severity and sporulation on apple seedlings in several plant assays when applied as preventive treatment. The effect of the inducer on key pre- and post-penetration events of V. inaequalis was studied and compared to these events in water-treated control leaves. The histopathological study showed that the inducer had its strongest effect on post-penetration events indicated by delayed infection and reduced stroma development. In addition, a small but significant inhibition of conidial germination and a stimulation of germ tube length were observed. This investigation provides new histopathological evidence for the mode of action of ASM against V. inaequalis and serves as a model for evaluation of the mechanisms by which the organically based fungicides reduce infection of V. inaequalis
THE ROLE OF THE BACTERIAL ENDOSYMBIONT, \u3ci\u3eARSENOPHONUS\u3c/i\u3e, IN THE SOYBEAN APHID, \u3ci\u3eAPHIS GLYCINES\u3c/i\u3e
Bacterial endosymbionts can have profound impacts on their host’s ecology. Notably, endosymbionts can protect their hosts against natural enemies and influence host plant interactions. The endosymbiont Candidatus Arsenophonus infects a wide taxonomic range of arthropod hosts, and is suspected of an uncharacterized mutualistic role in hemipterous insects. In the soybean aphid, Aphis glycines, an introduced pest of soybeans in the United States, Arsenophonus is the sole facultative endosymbiont. The focus of this dissertation is to characterize the role of Arsenophonus in the aphid, with an overall emphasis on its impact on aphid management strategies.
I first used diagnostic PCR to determine Arsenophonus infection frequency and strain diversity for native and introduced soybean aphids. I found that Arsenophonus infection is a uniform strain that is highly prevalent in soybean aphid. I then determined if Arsenophonus was a defense symbiont by curing two genotypes of soybean aphid of their natural Arsenophonus infection, resulting in infected and uninfected isolines within the same genetic background. I subjected these isolines to assays with three parasitoid species and a common aphid fungal pathogen, Pandora neoaphidis. I did not find differences in parasitism or fungal infections within the treatments. These results indicate that, although Arsenophonus is widespread, the symbiont should not interfere with biological control efforts.
I next examined the influence of Arsenophonus on the ability of soybean aphid “biotypes” to colonize resistant Rag plants. I cured three additional soybean aphid biotypes. All isolines were subjected to growth rate assays on resistant Rag versus susceptible soybean. My results indicate that Arsenophonus infected soybean aphids have an increased population growth compared to uninfected aphids regardless of soybean plant type
Finally, I induced soybean plants with jasmonic acid (JA) or salicylic acid (SA) to determine the effective plant defense against soybean aphid feeding. I also used Arsenophonus infected and uninfected aphids to determine any interaction between Arsenophonus and plant defense. I found SA treatment decreased soybean aphid population growth for one experiment, but had no effect when replicated. JA treatment had no effect, and there were no interactions between Arsenophonus infection and plant treatments
Evaluating a Hospital-Based Volunteer Doula Program in Vermont
Background: Continuous labor support is a feature of high quality maternal healthcare, yet access to doulas is not universal. Hospital-based doula programs can minimize barriers to continuous labor support, and evaluation of hospital-based doula programs is needed. This project evaluated a pilot volunteer doula program at an academic medical center.
Methods: Sign-in process monitored volunteer activity. Likert scale surveys measured participant (RN and doula) perspectives. Likert scale surveys with a comment section measured patient experience. Survey results and sign-in sheets were analyzed using descriptive statistics. Patient comments were assessed for themes using NVivo software.
Results: Volunteer doulas (VD) (n=74) supported 92 patients within 6 months (approximately 8.2% of total births during the period). Among RNs and VDs, average perception of support given to the patient was rated 4/5. Forty-two surveys were given to patients who received volunteer doula support, and 23 responses were received (54% response rate). All patient respondents (n=23) rated feeling supported during labor as “most important” (5/5). Ninety one percent of respondents “strongly agree” (5/5) they felt supported by the VD, and that the VD helped them to have a positive experience. Comment analysis revealed three themes: describing the doula, expressing gratitude for the program, and explaining how the doula helped.
Conclusions: The volunteer program increased access to continuous labor support, and patient response suggests positive experiences. Perspectives of nurses and volunteers during the pilot stage explores buy-in and guides improvements. Given these findings, the impact of this program on labor and delivery care was positive

Sensitivity of Antarctic Urospora penicilliformis (Ulotrichales, Chlorophyta) to ultraviolet radiation is life stage dependent
The sensitivity of different life stages of the eulittoral green alga Urospora penicilliformis (Roth) Aresch. to ultraviolet radiation (UVR) was examinedin the laboratory. Gametophytic filaments and propagules (zoospores and gametes) released from filaments were separately exposed to different fluence of radiation treatments consisting of PAR (P = 400700 nm), PAR + ultraviolet A (UVA) (PA, UVA = 320400 nm), and PAR + UVA + ultraviolet B (UVB) (PAB, UVB = 280320 nm). Photophysiological indices (ETRmax, Ek, and a) derived from rapid light curves were measured in controls, while photosynthetic efficiency and amount of DNA lesions in terms of cyclobutane pyrimidine dimers (CPDs) were measured after exposure to radiation treatments and after recovery in low PAR; pigments of propagules were quantified after exposure treatment only. The photosynthetic conversion efficiency (a) and photosynthetic capacity (rETRmax) were higher in gametophytes compared with the propagules. The propagules were slightly more sensitive to UVB-induced DNA damage; however, both life stages of the eulittoral inhabiting turf alga were not severely affected by the negative impacts of UVR. Exposure to a maximum of 8 h UVR caused mild effects on the photochemical efficiency of PSII and induced minimal DNA lesions in both the gametophytes and propagules. Pigment concentrations were not significantly different between PAR-exposed and PAR + UVRexposed propagules. Our data showed that U. penicilliformis from the Antarctic is ratherinsensitive to the applied UVR. This amphi-equatorial species possesses different protective mechanisms that can cope with high UVR in coldtemperatewaters of both hemispheres and in polar regions under conditions of increasing UVR as a consequence of further reduction of stratosphericozone
