    HDAC3 is essential for Human Leukemic Cell Growth and the Expression of β-catenin, MYC, and WT1

    Therapy of acute myeloid leukemia (AML) is unsatisfactory. Histone deacetylase inhibitors (HDACi) are active against leukemic cells in vitro and in vivo. Clinical data suggest further testing of such epigenetic drugs and to identify mechanisms and markers for their efficacy. Primary and permanent AML cells were screened for viability, replication stress/DNA damage, and regrowth capacities after single exposures to the clinically used pan-HDACi panobinostat (LBH589), the class I HDACi entinostat/romidepsin (MS-275/FK228), the HDAC3 inhibitor RGFP966, the HDAC6 inhibitor marbostat-100, the non-steroidal anti-inflammatory drug (NSAID) indomethacin, and the replication stress inducer hydroxyurea (HU). Immunoblotting was used to test if HDACi modulate the leukemia-associated transcription factors beta-catenin, Wilms tumor (WT1), and myelocytomatosis oncogene (MYC). RNAi was used to delineate how these factors interact. We show that LBH589, MS-275, FK228, RGFP966, and HU induce apoptosis, replication stress/DNA damage, and apoptotic fragmentation of beta-catenin. Indomethacin destabilizes beta-catenin and potentiates anti-proliferative effects of HDACi. HDACi attenuate WT1 and MYC caspase-dependently and -independently. Genetic experiments reveal a cross-regulation between MYC and WT1 and a regulation of beta-catenin by WT1. In conclusion, reduced levels of beta-catenin, MYC, and WT1 are molecular markers for the efficacy of HDACi. HDAC3 inhibition induces apoptosis and disrupts tumor-associated protein expression

    Making Meaning Together: An Exploratory Study of Therapeutic Conversation between Helping Professionals and Homeless Shelter Residents

    This exploratory study examined the nature of therapeutic conversation between helping professionals and homeless persons as an intervention to optimize health. Meaningful conversation occurred in relationships where there was a sense of connection and the presence of rapport. Emergent facilitators of therapeutic conversation included respectful engagement, casual nature of conversation, alternative settings for therapeutic conversation, effective listening, and establishing trust. Barriers included prejudging homeless persons, fear of punishment and authority, and academic and professional intimidation. Central to the study findings was the acknowledgement of the client\u27s personhood. Acknowledgement of personhood is a critical element in engagement between homeless persons and helping professionals

    Vat photopolymerization of cemented carbide specimen

    Numerous studies show that vat photopolymerization enables near-net-shape printing of ceramics and plastics with complex geometries. In this study, vat photopolymerization was investigated for cemented carbide specimens. Custom-developed photosensitive WC-12 Co (wt%) slurries were used for printing green bodies. The samples were examined for defects using quantitative microstructure analysis. A thermogravimetric analysis was performed to develop a debinding program for the green bodies. After sintering, the microstructure and surface roughness were evaluated. As mechanical parameters, Vickers hardness and Palmqvist fracture toughness were considered. A linear shrinkage of 26–27% was determined. The remaining porosity fraction was 9.0%. No free graphite formation, and almost no η-phase formation occurred. WC grain growth was observed. 76% of the WC grains measured were in the suitable size range for metal cutting tool applications. A hardness of 1157 HV10 and a Palmqvist fracture toughness of 12 MPa m\sqrt{m} was achieved. The achieved microstructure exhibits a high porosity fraction and local cracks. As a result, vat photopolymerization can become an alternative forming method for cemented carbide components if the amount of residual porosity and defects can be reduced

    Inhibition of FLT3 and PDGFR tyrosine kinase activity by bis(benzo[b]furan-2-yl)methanones

    A series of bis(benzo[b]furan-2-yl)methanones was synthesized and tested for inhibition of FLT3 and PDGFR autophosphorylation. Mostly, C-5 substitution leads to PDGFR selectivity, which was strongest in the case of the 5,5'-dimethoxy deriv. The 5,5'-diamino and the 6,6'-dihydroxy compds. are more active at FLT3. At both kinases, the potency of the best inhibitors approaches IC50 values of .apprx.0.5 mM. Mol. modeling studies suggest that the bisbenzofuranylmethanones are able to fit into the same binding site as their indolyl analogs which have been suggested to form a bidentate hydrogen bridge with the backbone in the hinge regions. The loss of one H bond by the NH-O exchange might be partially compensated by, for example, the weak interaction of one furanyl oxygen with FLT3 Cys-828

    This 156 page publication (NRAES-133) was originally published by the Natural Resource, Agriculture, and Engineering Service (NRAES, previously known as the Northeast Regional Agricultural Engineering Service), a multi-university program in the Northeast US disbanded in 2011. Plant and Life Sciences Publishing (PALS) was subsequently formed to manage the NRAES catalog. Ceasing operations in 2018, PALS was a program of the Department of Horticulture in the College of Agriculture and Life Sciences (CALS) at Cornell University. PALS assisted university faculty in publishing, marketing and distributing books for small farmers, gardeners, land owners, workshops, college courses, and consumers.Plasticulture is the use of plastics in agriculture.
