    Synaptic Remodeling Depends on Signaling between Serotonin Receptors and the Extracellular Matrix

    Rewiring of synaptic circuitry pertinent to memory formation has been associated with morphological changes in dendritic spines and with extracellular matrix (ECM) remodeling. Here, we mechanistically link these processes by uncovering a signaling pathway involving the serotonin 5-HT7 receptor (5-HT7R), matrix metalloproteinase 9 (MMP-9), the hyaluronan receptor CD44, and the small GTPase Cdc42. We highlight a physical interaction between 5-HT7R and CD44 (identified as an MMP-9 substrate in neurons) and find that 5-HT7R stimulation increases local MMP-9 activity, triggering dendritic spine remodeling, synaptic pruning, and impairment of long-term potentiation (LTP). The underlying molecular machinery involves 5-HT7R-mediated activation of MMP-9, which leads to CD44 cleavage followed by Cdc42 activation. One important physiological consequence of this interaction includes an increase in neuronal outgrowth and elongation of dendritic spines, which might have a positive effect on complex neuronal processes (e.g., reversal learning and neuronal regeneration)

    Vectors and vector-borne diseases of horses

    Most diseases of horses with zoonotic importance are transmitted by arthropods. The vectors belong to two very distantly related groups, the chelicerate Ixodidae (Acari = ticks) and the hexapod Diptera (true flies). Almost all relevant species are predestined for transmitting pathogens by their blood-sucking habits. Especially species of Diptera, one of the megadiverse orders of holometabolan insects (ca. 150.000 spp.), affect the health status and performance of horses during the grazing period in summer. The severity of pathological effect depends on the pathogen, but also on the group of vectors and the intensity of the infection or infestation. Dipteran species but also blood-sucking representatives of Acari (Ixodidae) can damage their hosts by sucking blood, causing myiasis, allergy, paralysis and intoxication, and also transmit various bacterial, viral, parasitic, spirochetal and rickettsial diseases to animals and also humans. The aim of this review was to provide extensive information on the infectious diseases transmitted by members of the two arthropod lineages (Ixodidae, Diptera) and a systematic overview of the vectors. For each taxon, usually on the ordinal, family, and genus level a short characterisation is given, allowing non-entomologists easy identification. Additionally, the biology of the relevant species (or genera) is outlined briefly

    Knowledge, attitudes, beliefs and behaviours of older adults about pneumococcal immunization, a Public Health Agency of Canada/Canadian Institutes of Health Research Influenza Research Network (PCIRN) investigation

    Background: Fewer Canadian seniors are vaccinated against pneumococcal disease than receive the influenza vaccine annually. Improved understanding of factors influencing pneumococcal vaccination among older adults is needed to improve vaccine uptake. Methods: A self-administered survey measuring knowledge, attitudes, beliefs and behaviours about pneumococcal vaccination was administered to a cohort of seniors participating in a clinical trial of seasonal influenza vaccines at eight centers across Canada. Eligible participants were ambulatory adults 65 years of age or older, in good health or with stable health conditions, previously given influenza vaccine. The primary outcome was self-reported receipt of pneumococcal vaccination. Multi-variable logistic regression was used to determine factors significantly associated with pneumococcal vaccine receipt. Results: A total of 863 participants completed questionnaires (response rate 92%); 58% indicated they had received the pneumococcal vaccine. Being offered the vaccine by a health care provider had the strongest relationship with vaccine receipt (AOR 23.4 (95% CI 13.4-40.7)). Other variables that remained significantly associated with vaccine receipt in the multivariable model included having heard of the vaccine (AOR 10.1(95% CI 4.7-21.7)), and strongly agreeing that it is important for adults > 65 to be vaccinated against pneumococcus (AOR 3.3 (95% CI 1.2-9.2)). Participants who were < 70 years of age were less likely to be vaccinated. Conclusions: These results indicate healthcare recommendation significantly influenced vaccine uptake in this population of older adults.
