    A comparison of analytical and numerical model predictions of shallow soil temperature variation with experimental measurements

The Publisher's final version can be found by following the DOI link.In several fields of enquiry such as geothermal energy, geology and agriculture, it is of interest to study the thermal behaviour of shallow soils. For this, several analytical and numerical methodologies have been proposed to analyse the temperature variation of the soil in the short and long term. In this paper, a comparative study of different models (sinusoidal, semi-infinite and finite difference method) is conducted to estimate the shallow soil temperature variation in the short and long term. The models were compared with hourly experimental measured data of soil temperature in Leicester, UK, at depths between 0.75 and 2.75 m. The results show that the sinusoidal model is not appropriate to evaluate the short-term temperature variations, such as hourly or daily fluctuations. Likewise, this model is highly affected by the undisturbed ground temperature and can lead to very high errors. Regarding the semi-infinite model, it is accurate enough to predict the short-term temperature variation. However, it is useless to predict the long-term variation at depths greater than 1 m. The finite difference method (FDM) considering the air temperature as a boundary condition for the soil surface is the most accurate approach for estimating both short and long-term temperature variations while the FDM with heat flux as boundary condition is the least accurate approach due to the uncertainty of the assumed parameters. The ranges of errors for the sinusoidal, semi-infinite and FDM are found to be from 76.09 to 142.13%, 12.11 to 104.88% and 1.82 to 28.14% respectively

    The Skn7, Ssk1 and Rim15 proteins are response regulators involved in osmotic, oxidative and nutritional stress in fungi. In order to verify the involvement of these genes in Trichoderma atroviride IMI206040’s growth, conidiation, direct antagonism against plant pathogens (Rhizoctonia solani and Sclerotinia sclerotiorum), production of volatile organic compounds (VOCs) with fungistatic effect, and interaction with plants (growth promotion), single mutants were generated, and the phenotypic patterns were analysed in comparison to the wild-type (wt) strain. The mutants were submitted to osmotic, oxidative, membrane and cell wall stress conditions in vitro. The Δskn7 and Δrim15 mutants did not show either significant differences at morphological level, or marked decreases in mycelial growth and conidiation in relation to wt, whereas Δssk1 had altered phenotypes in most conditions tested. The plant-growth promotion of Arabidopsis thaliana seedlings induced by VOCs was not quantitatively modified by any of the mutants in relation to the wt strain, although possible differences in secondary root hairs was noticed for Δrim15. The fungistatic activity was significantly altered for Δssk1 and Δrim15. Overall, the Δssk1 strain showed remarkable morphological differences, with decrease in mycelial growth and conidiation, being also affected in the antagonistic capacity against plant pathogens. The impacts demonstrated by the deletion of ssk1 suggest this gene has a relevant participation in the signalling response to different stresses in T. atroviride and in the interactive metabolism with phytopathogens and plants. On the other hand, unlike other fungal models, Skn7 did not appear to have a critical participation in the above-mentioned processes; Rim15 seemed to confirm its involvement in modulating cellular responses to nutritional status, although with a possible cross-talk with other cellular processes. Our results suggest that Ssk1 likely plays a key regulatory role, not only in basic metabolisms of T. atroviride, but also in biocontrol-related characteristics

    Are shallow boreholes a suitable option for inter-seasonal ground heat storage for the small housing sector?

    open accessIn recent years, various researchers have studied the performance of Solar Assisted Ground Source Heat Pump (SAGSHP) systems using borehole heat exchangers. However, the research conducted has been limited to conventional boreholes (30m to 150m depth), which are expensive and not suitable for the small housing sector. This paper reports an experimental analysis of a shallow SAGSHP system with inter-seasonal storage. The system, installed in Leicester UK, consists of seven photovoltaic-thermal (PVT) collectors connected in series with an array of 16 shallow boreholes (1.5 meters depth). Data regarding the energy fluxes involved in the soil-based thermal store have been monitored and analysed for one year. The results show that the shallow soil is able to serve as a storage medium to cover the heating demands of a near zero energy domestic building. However, it was noticed that in addition to the solar heat captured and stored in the soil, the system covers part of the heating demand from heat extracted from the soil surrounding the thermal store. During winter, the lowest temperature reached by the soil so far is 2 °C. Hence, no freezing problems have occurred in the soil. An analysis of the temperature variation of the ground storage under the system operation is also shown

    Tablas, graficas, mapas, imágenesPara el desarrollo del proyecto se estudia la factibilidad de crear y ejecutar una empresa procesadora de hamburguesas a base de gluten de trigo, estas se caracterizan por ser un producto innovador dirigido a personas de diversas clases sociales, que pueden acceder a este novedoso producto por su bajo costo. Por esta razón se realiza un producto de buena calidad que presentan propiedades físicas y químicas muy particulares con relación a otros productos sustitutos, ofreciendo una nutrición rica y balanceada; estas cualidades hacen que el producto sea necesario para el consumidor, creando una demanda insatisfecha en el mercado; la cual se verifico mediante encuestas realizadas a los habitantes de esta zona y por consiguiente genera un nivel de oferta bajo del producto ya que la competencia es mínima. Teniendo en cuenta lo mencionado anteriormente se ha elegido desarrollar el proyecto en la localidad de Kennedy, ya que este sector cuenta con los servicios públicos necesarios y la planta procesadora posee buenas condiciones de infraestructura, además porque no existen normas o restricciones que impidan la ejecución del proyecto. Se establece por medio del estudio financiero que el procesamiento de las hamburguesas a base de gluten de trigo es viable, y que de igual forma la relación costo beneficio indica que por cada peso invertido en el desarrollo del proyecto se obtiene una ganancia adicional. Se determina que el presente proyecto se ubica en el sector secundario, en el subsector de industrias alimenticias diversas, con la actividad económica de fabricación de hamburguesas a base de gluten de trigo.No Abstrac

    ERBB4 confers metastatic capacity in Ewing sarcoma.

    Metastatic spread is the single-most powerful predictor of poor outcome in Ewing sarcoma (ES). Therefore targeting pathways that drive metastasis has tremendous potential to reduce the burden of disease in ES. We previously showed that activation of the ERBB4 tyrosine kinase suppresses anoikis, or detachment-induced cell death, and induces chemoresistance in ES cell lines in vitro. We now show that ERBB4 is transcriptionally overexpressed in ES cell lines derived from chemoresistant or metastatic ES tumours. ERBB4 activates the PI3K-Akt cascade and focal adhesion kinase (FAK), and both pathways contribute to ERBB4-mediated activation of the Rac1 GTPase in vitro and in vivo. ERBB4 augments tumour invasion and metastasis in vivo, and these effects are blocked by ERBB4 knockdown. ERBB4 expression correlates significantly with reduced disease-free survival, and increased expression is observed in metastatic compared to primary patient-matched ES biopsies. Our findings identify a novel ERBB4-PI3K-Akt-FAK-Rac1 pathway associated with aggressive disease in ES. These results predict that therapeutic targeting of ERBB4, alone or in combination with cytotoxic agents, may suppress the metastatic phenotype in ES

    The regionalization of framing issues for producers Livestock Group of Validation and Technology Transfer (GGAVATT) from San Jose Miahuatlán Veracruz, Mexico, under the current production of native grass prairie with mainly during the months of higher temperature have a poor performance and thus milk production declines which promotes a great use of concentrated feeds which causes an increase in production cost and less use to them. For this purpose we made a diagnosis of current conditions GGAVATT production, yielding information that helps us source and compared to determine precisely what the effect of applying the proposed alternative. We propose an alternative implementation of pastures with improved pastures needed to produce a greater amount of forage per unit area especially in the colder months.La regionalización de la problemática encuadrando a los productores del Grupo Ganadero de Validación y Transferencia de Tecnología (GGAVATT) de San José Miahuatlán Veracruz, México, bajo la actual situación de producción de praderas básicamente con gramas nativas que durante los meses de menor frío tienen un pobre desempeño y por ende la producción de leche se deteriora lo cual promueve una gran utilización de alimentos concentrados lo que provoca un incremento en el costo de producción y una menor utilidad para estos. Para dicho propósito se realizó un diagnóstico de las condiciones actuales de producción del GGAVATT, arrojando información que nos sirve de partida y de comparación para determinar con precisión cuál es el efecto de aplicar la alternativa propuesta. Se plantea como alternativa la implantación de praderas con pastos mejorados que permitan producir una mayor cantidad de forrajes por unidad de superficie sobre todo en los meses de menor temperatura

    Amyloid-b peptide on sialyl-LewisX-selectin-mediated membrane tether mechanics at the cerebral endothelial cell surface

    Increased deposition of amyloid-b peptide (Ab) at the cerebral endothelial cell (CEC) surface has been implicated in enhancement of transmigration of monocytes across the brain blood barrier (BBB) in Alzheimer’s disease (AD). In this study, quantitative immunofluorescence microscopy (QIM) and atomic force microscopy (AFM) with cantilevers biofunctionalized by sialyl-Lewisx (sLex) were employed to investigate Ab-altered mechanics of membrane tethers formed by bonding between sLex and p-selectin at the CEC surface, the initial mechanical step governing the transmigration of monocytes. QIM results indicated the ability for Ab to increase p-selectin expression at the cell surface and promote actin polymerization in both bEND3 cells (immortalized mouse CECs) and human primary CECs. AFM data also showed the ability for Ab to increase cell stiffness and adhesion probability in bEND3 cells. On the contrary, Ab lowered the overall force of membrane tether formation (Fmtf), and produced a bimodal population of Fmtf, suggesting subcellular mechanical alterations in membrane tethering. The lower Fmtf population was similar to the results obtained from cells treated with an F-actin-disrupting drug, latrunculin A. Indeed, AFM results also showed that both Ab and latrunculin A decreased membrane stiffness, suggesting a lower membrane-cytoskeleton adhesion, a factor resulting in lower Fmtf. In addition, these cerebral endothelial alterations induced by Ab were abrogated by lovastatin, consistent with its anti-inflammatory effects. In sum, these results demonstrated the ability for Ab to enhance p-selectin expression at the CEC surface and induce cytoskeleton reorganization, which in turn, resulted in changes in membrane-cytoskeleton adhesion and membrane tethering, mechanical factors important in transmigration of monocytes through the BBB.This work was supported by Alzheimer Association Grant NIRG-06-24448; NIH Grant 1P01 AG18357, R21NS052385, 5R21AG032579 and in part by 1P01HL095486 and AHA 0835676N; ‘‘Bolashak’’ scholarship and Ministry of Education and Science of the Republic of Kazakhstan 1029/GF2.     Detection of Babesia spp. in High Altitude Cattle in Ecuador, Possible Evidence of the Adaptation of Vectors and Diseases to New Climatic Conditions

    Background: Babesia species are intraerythrocytic protozoa, distributed in tropical and subtropical areas of the world, causing anemic diseases in many animals, including cattle. This disease, called babesisosis, is transmitted from one animal to another through ticks (Tick Borne-Disease or TBD). On the other hand, Ecuador has a tropical climate that allows the development of the vector Rhipicephalus microplus, and therefore favors the transmission of Babesia spp. in cattle. Methods and principal findings: We determined the presence of Babesia spp. by PCR using 18s ribosomal gene as target (18s PCR) in 20 farms in the area of El Carmen (zone below 300 m above sea level) and 1 farm in Quito (2469 m.a.s.l.). In addition, we analyzed parameters such as age, sex, and packed cell volume (PCV) as explanatory variable associated with the disease. Results: The 18s PCR test showed that 18.94% (14.77% Babesia bovis and 4.17% Babesia bigemina) and 20.28% (14.69% B. bovis and 5.59% B. bigemina) of the cattle were positive for Babesia spp in farms sampled in El Carmen and in Quito, respectively. Age influenced the presence of animals positive for Babesia spp., but sex and PCV did not. The phylogenetic analysis of sequences showed 4 isolates of B. bovis and 3 isolates of B. bigemina in the 2 study zones, with similarities between 99.73 and 100% with other sequences. One B. bovis isolate was similar in the zone of El Carmen and Quito. Conclusion and significance: This work is the first molecular characterization of B. bigemina and B. bovis in Ecuador, and it is also the first evidence of Babesia spp. in cattle in the area of Quito at an altitude of 2469 m.a.s.l., being the highest altitude reported for animals with babesiosis and for the tick R. microplus. Climatic factors as well as mobility of tick-carrying animals without any control allow the presence of Babesiosis outbreaks in new geographical areas
