    BRCA1 Zinc RING Finger Domain Disruption Alters Caspase Response in Ovarian Surface Epithelial Cells

    BACKGROUND: The frequently occurring 185delAG mutation occurs in the amino-terminal zinc RING domain of the breast and ovarian cancer susceptibility gene, BRCA1. We sought to determine differential cell viability and apoptotic response of human ovarian surface epithelial cells with and without the 185delAG mutation. RESULTS: BRCA1wt and BRCA1+ cells were treated with staurosporine. Cell proliferation assays showed BRCA1wt cells grew to a greater extent compared to BRCA1+ cells. Trypan blue exclusion assays confirmed this observation. Western immunoblot analysis revealed that caspase 3 levels were higher after staurosporine treatment in BRCA1+ cells than in wild type cells, while full length DNA Fragmentation Factor 45 levels were lower in BRCA1+ cells. While there was no significant difference in levels of excision repair cross complementing protein1 (ERCC1) with BRCA1 status, BRCA1+ cells demonstrated cleavage of polyribose ADP polymerase (PARP) before wild type cells. CONCLUSIONS: Disruption of the BRCA1 RING domain caused altered cell viability and caspase-dependent apoptotic response after chemotoxic stress

    We build a database of genus 2 curves defined over Q\mathbb Q which contains all curves with minimal absolute height h5h \leq 5, all curves with moduli height h20\mathfrak h \leq 20, and all curves with extra automorphisms in standard form y2=f(x2)y^2=f(x^2) defined over Q\mathbb Q with height h101h \leq 101. For each isomorphism class in the database, an equation over its minimal field of definition is provided, the automorphism group of the curve, Clebsch and Igusa invariants. The distribution of rational points in the moduli space M2\mathcal M_2 for which the field of moduli is a field of definition is discussed and some open problems are presented

    Equity of Inpatient Health Care in Rural Tanzania:\ud A Population- and Facility-Based Survey

    To explore the equity of utilization of inpatient health care at rural Tanzanian health centers through the use of a short wealth questionnaire.Methods: Patients admitted to four rural health centers in the Kigoma Region of Tanzania from May 2008 to May 2009 were surveyed about their illness, asset ownership and demographics. Principal component analysis was used to compare the wealth of the inpatients to the wealth of the region’s general population, using data from a previous population-based survey. Among inpatients, 15.3% were characterized as the most poor, 19.6% were characterized as very poor, 16.5% were characterized as poor, 18.9% were characterized as less poor, and 29.7% were characterized as the least poor. The wealth distribution of all inpatients (p < 0.0001), obstetric inpatients (p < 0.0001), other inpatients (p < 0.0001), and fee-exempt inpatients (p < 0.001) were significantly different than the wealth distribution in the community population, with poorer patients underrepresented among inpatients. The wealth distribution of pediatric inpatients (p = 0.2242) did not significantly differ from the population at large. The findings indicated that while current Tanzanian health financing policies may have improved access to health care for children under five, additional policies are needed to further close the equity gap, especially for obstetric inpatients.\u

    The virtual observatory service TheoSSA: Establishing a database of synthetic stellar flux standards. II. NLTE spectral analysis of the OB-type subdwarf Feige 110

    In the framework of the Virtual Observatory (VO), the German Astrophysical Virtual Observatory (GAVO) developed the registered service TheoSSA (Theoretical Stellar Spectra Access). It provides easy access to stellar spectral energy distributions (SEDs) and is intended to ingest SEDs calculated by any model-atmosphere code, generally for all effective temperature, surface gravities, and elemental compositions. We will establish a database of SEDs of flux standards that are easily accessible via TheoSSA's web interface. The OB-type subdwarf Feige 110 is a standard star for flux calibration. State-of-the-art non-local thermodynamic equilibrium (NLTE) stellar-atmosphere models that consider opacities of species up to trans-iron elements will be used to provide a reliable synthetic spectrum to compare with observations. In case of Feige 110, we demonstrate that the model reproduces not only its overall continuum shape from the far-ultraviolet (FUV) to the optical wavelength range but also the numerous metal lines exhibited in its FUV spectrum. We present a state-of-the-art spectral analysis of Feige 110. We determined Teff=47250±2000KT_\mathrm{eff} = 47\,250 \pm 2000\,\mathrm{K}, logg=6.00±0.20\log g = 6.00 \pm 0.20 and the abundances of He, N, P, S, Ti, V, Cr, Mn, Fe, Co, Ni, Zn, and Ge. Ti, V, Mn, Co, Zn, and Ge were identified for the first time in this star. Upper abundance limits were derived for C, O, Si, Ca, and Sc. The TheoSSA database of theoretical SEDs of stellar flux standards guarantees that the flux calibration of astronomical data and cross-calibration between different instruments can be based on models and SEDs calculated with state-of-the-art model-atmosphere codes.Comment: 19 pages, 7 figure

    Metal abundances in PG1159 stars from Chandra and FUSE spectroscopy

    We investigate FUSE spectra of three PG1159 stars and do not find any evidence for iron lines. From a comparison with NLTE models we conclude a deficiency of 1-1.5 dex. We speculate that iron was transformed into heavier elements. A soft X-ray Chandra spectrum of the unique H- and He-deficient star H1504+65 is analyzed. We find high neon and magnesium abundances and confirm that H1504+65 is the bare core of either a C-O or a O-Ne-Mg white dwarf.Comment: To be published in: Proceedings 13th European Workshop on White Dwarfs, NATO Science Series, 4 pages, 1 figur

    Ovarian Epithelial-Stromal Interactions: Role of Interleukins 1 and 6

    Ovarian epithelial cancer is the most lethal gynecologic malignancy. The high mortality is attributed to the fact that most cases typically present in late stage when ovarian cancer (OC) has already spread beyond the ovary. Ovarian epithelial cancer cells are shed into intraperitoneal ascites and easily disseminate throughout the peritoneal cavity with preferential metastasis to the omentum, peritoneum, and local organs. Understanding how ovarian epithelial cells interact with and modulate their microenvironment can provide insight into the molecular mechanism(s) involved with malignant transformation and progression which may eventually identify novel diagnostic, prognostic, and therapeutic targets. The objective of this paper is to provide a brief consideration of ovarian surface epithelial-stromal interactions in regard to normal physiological function and tumor progression as influenced by two potentially key interleukins, interleukins-1 (IL-1) and -6 (IL-6), present in the microenvironment. Lastly, we will consider the clinical implications of IL-1 and IL-6 for OC patients
