    Extending an industrial root controller : implementation and applications of a fast open sensor interface

    An overview is given of the design and implementation of a platform for fast external sensor integration in an industrial robot system called ABB S4CPlus. As an application and motivating example, the implementation of force-controlled grinding and deburring within the AUTOFETT-project is discussed. Experiences from industrial usage of the fully developed prototype confirms the appropriateness of the design choices, thus also confirming the fact that control and software need to be tightly integrated. The new sensor can be used for the prototyping and development of a wide variety of new application

    Glioma stem cells but not bulk glioma cells upregulate IL-6 secretion in microglia/brain macrophages via toll-like receptor 4 signaling

    Peripheral macrophages and resident microglia constitute the dominant glioma-infiltrating cells. The tumor induces an immunosuppressive and tumor-supportive phenotype in these glioma-associated microglia/brain macrophages (GAMs). A subpopulation of glioma cells acts as glioma stem cells (GSCs). We explored the interaction between GSCs and GAMs. Using CD133 as a marker of stemness, we enriched for or deprived the mouse glioma cell line GL261 of GSCs by fluorescence-activated cell sorting (FACS). Over the same period of time, 100 CD133(+ )GSCs had the capacity to form a tumor of comparable size to the ones formed by 10,000 CD133(-)GL261 cells. In IL-6(-/-)mice, only tumors formed by CD133(+ )cells were smaller compared with wild type. After stimulation of primary cultured microglia with medium from CD133-enriched GL261 glioma cells, we observed an selective upregulation in microglial IL-6 secretion dependent on Toll-like receptor (TLR) 4. Our results show that GSCs, but not the bulk glioma cells, initiate microglial IL-6 secretion via TLR4 signaling and that IL-6 regulates glioma growth by supporting GSCs. Using human glioma tissue, we could confirm the finding that GAMs are the major source of IL-6 in the tumor context

    Anisotropic, non-monotonic behavior of the superconducting critical current in thin YBa2Cu3O7-d films on vicinal SrTiO3 surfaces

    The critical current density of epitaxial YBCO films grown on vicinal SrTiO3 substrates was investigated by electrical transport measurements along and across the steps of the SrTiO3 surface for a range of temperatures of 10 K to 85 K and in applied magnetic fields varying from 0 to 14 T. For vicinal angles of 4 and 8 degrees, we found evidence of enhanced pinning in the longitudinal direction at low magnetic fields for a wide region of temperatures and attribute this phenomenon to anti-phase boundaries in the YBCO film. The transverse Jc data showed a peak in the Jc(H) curve at low magnetic fields, which was explained on the basis of magnetic interaction between Abrikosov and Abrikosov-Josephson vortices. The in-plane Jc anisotropy observed for vicinal angles of 0.4 degrees was reversed with respect to the 8 degree and 4 degree samples. This phenomenon was interpreted on the basis of strain induced in the YBCO film by the stepped substrate's surface.Comment: accepted for publication in Phys. Rev.

    Ghrelin increases intake of rewarding food in rodents

    We investigated whether ghrelin action at the level of the ventral tegmental area (VTA), a key node in the mesolimbic reward system, is important for the rewarding and motivational aspects of the consumption of rewarding/palatable food. Mice with a disrupted gene encoding the ghrelin receptor (GHS-R1A) and rats treated peripherally with a GHS-R1A antagonist both show suppressed intake of rewarding food in a free choice (chow/rewarding food) paradigm. Moreover, accumbal dopamine release induced by rewarding food was absent in GHS-R1A knockout mice. Acute bilateral intra-VTA administration of ghrelin increased 1-hour consumption of rewarding food but not standard chow. In comparison with sham rats, VTA-lesioned rats had normal intracerebroventricular ghrelin-induced chow intake, although both intake of and time spent exploring rewarding food was decreased. Finally, the ability of rewarding food to condition a place preference was suppressed by the GHS-R1A antagonist in rats. Our data support the hypothesis that central ghrelin signaling at the level of the VTA is important for the incentive value of rewarding food

    Neurofibromatosis 1 - mutant microglia exhibit sexually-dimorphic cyclic AMP-dependent purinergic defects

    As critical regulators of brain homeostasis, microglia are influenced by numerous factors, including sex and genetic mutations. To study the impact of these factors on microglia biology, we employed genetically engineered mice that model Neurofibromatosis type 1 (NF1), a disorder characterized by clinically relevant sexually dimorphic differences. While microglia phagocytic activity was reduced in both male and female heterozygous Nf1 mutant (Nf1+/-) mice, purinergic control of phagocytosis was only affected in male Nf1+/- mice. ATP-induced P2Y-mediated membrane currents and P2RY12-dependent laser lesion-induced accumulation of microglial processes were also only impaired in male, but not female Nf1+/-, microglia. These defects resulted from Nf1+/- male-specific defects in cyclic AMP regulation, rather than from changes in purinergic receptor expression. Cyclic AMP elevation by phosphodiesterase blockade restored the male Nf1+/- microglia defects in P2Y-dependent membrane currents and process motility. Taken together, these data establish a sex-by-genotype interaction important to microglia function in the adult mouse brain
