    Immigration Legislation and Issues in the 114th Congress

    The House and the Senate have considered immigration measures on a variety of issues in the 114th Congress. The Consolidated Appropriations Act, 2016 (P.L. 114-113) extends four immigration programs through September 30, 2016: the EB-5 immigrant investor Regional Center Pilot Program, the E-Verify employment eligibility verification system, the Conrad State program for foreign medical graduates, and the special immigrant religious worker program. P.L. 114-113 also contains provisions on the Visa Waiver Program and certain nonimmigrant visa categories. Other enacted immigration-related measures include the Border Jobs for Veterans Act of 2015 (P.L. 114-68) on border security personnel, the Adoptive Family Relief Act (P.L. 114-70) on intercountry adoption, and the National Defense Authorization Act for Fiscal Year 2016 (P.L. 114-92) on the Afghan special immigrant visa program. The House has passed several other immigration-related bills. Among them are the Northern Border Security Review Act (H.R. 455), the Preclearance Authorization Act of 2015 (H.R. 998), the Border Security Technology Accountability Act of 2015 (H.R. 1634), the Enforce the Law for Sanctuary Cities Act (H.R. 3009), and the American SAFE Act of 2015 (H.R. 4038). H.R. 998 has also been reported by the Senate Homeland Security and Governmental Affairs Committee with an amendment in the nature of a substitute. In addition, various bills on border security, interior enforcement, visa security, and asylum, among other issues, have been considered by a House or Senate committee. Border security- related measures have been reported or ordered to be reported by the House Homeland Security Committee (H.R. 399, H.R. 3583, H.R. 3586), or the Senate Homeland Security and Governmental Affairs Committee (S. 750, S. 1808, S. 1864,S. 1873). Interior enforcement provisions are included in bills ordered to be reported by the House Judiciary Committee (H.R. 1147, H.R. 1148, H.R. 1153) or reported by the House Appropriations Committee (H.R. 3128). S. 1635, as reported by the Senate Foreign Relations Committee, also contains interior enforcement- related provisions. Several of these interior enforcement bills also contain key provisions on other immigration issues. Among the other issues addressed in these bills are employment eligibility verification (H.R. 1147); visa security and naturalization (H.R. 1148); and expedited removal, asylum, parole, and unaccompanied alien children (H.R. 1153). H.R. 1149, as ordered to be reported by the House Judiciary Committee, also addresses unaccompanied alien children. This report discusses these and other immigration-related issues that have received legislative action or are of significant congressional interest in the 114th Congress. Department of Homeland Security appropriations are addressed in CRS Report R44053, Department of Homeland Security Appropriations: FY2016, and, for the most part, are not covered here

    The Nutrients L-Glutamine and Glucose Have Unique Roles in B Lymphocyte Growth and Proliferation Responses

    Thesis advisor: Welkin JohnsonThesis advisor: Thomas ChilesB cell activation is an energetically demanding process during which B lymphocytes undergo reprogramming and shift from a resting state to a highly proliferative, metabolically active state. Little is known about the metabolic reprogramming process or the role extracellular nutrients play in the activation response. Here we demonstrate that there are distinct requirements for the nutrients L-glutamine and glucose during activation. We show that cells activated in glucose-depleted conditions are still able to undergo growth and signaling events. In contrast, we show that extracellular L-glutamine is essential for all but the earliest activation events, and cells cultured in L-glutamine-deprived conditions are unable to enter the cell cycle. Consistently, we show that extracellular supplementation of the cell-permeable derivative of α-ketoglutarate (α-KG), a glutaminolytic product, is able to rescue cell activation in the absence of glutamine. We also show the induction of the high affinity amino acid transporter ASCT2 is required for glutamine uptake following B cell receptor (BCR) crosslinking. Specifically, we found that halting glutamine uptake or processing by inhibiting ASCT2 or the glutaminolytic enzyme glutaminase causes activation defects that parallel those observed in glutamine deprived conditions, indicating a requirement for glutaminolysis during the very early stages of activation. We found that -KG does not contribute to epigenetic remodeling, but is necessary for mammalian target of rapamycin complex 1 (mTORC1) activation. In turn, mTORC1 activity is required for upregulation of the glucose transporter Glut1 during the initial 24 hours of activation, as well as increased glucose uptake. These findings indicate a distinct metabolic profile that begins with glutamine uptake, and acts through mTORC1 signaling to later promote glucose uptake. Finally, we show that nutrients contribute to functional differentiation events during B cell activation. Glucose is required to support biogenesis of the endoplasmic reticulum as well as differentiation into plasma-like cells, while glutamine is required to support differentiation into IL-10 secreting regulatory B cell subsets. The requirement for glutamine for in vitro B10 cell differentiation is the first reported link between nutrient signaling and regulatory B cell development, and is a novel finding in the field.Thesis (PhD) — Boston College, 2016.Submitted to: Boston College.     Identification of a Widespread Palmitoylethanolamide Contamination in Standard Laboratory Glassware

    Introduction: Fatty acid ethanolamides (FAEs) are a family of lipid mediators that participate in a host of biological functions. Procedures for the quantitative analysis of FAEs include organic solvent extraction from biological matrices (e.g., blood), followed by purification and subsequent quantitation by liquid chromatography-mass spectrometry (LC/MS) or gas chromatography-mass spectrometry. During the validation process of a new method for LC/MS analysis of FAEs in biological samples, we observed unusually high levels of the FAE, palmitoylethanolamide (PEA), in blank samples that did not contain any biological material. Materials and Methods: We investigated a possible source of this PEA artifact via liquid chromatography coupled to tandem mass spectrometry, as well as accurate mass analysis. Results: We found that high levels of a contaminant indistinguishable from PEA is present in new 5.75″ glass Pasteur pipettes, which are routinely used by laboratories to carry out lipid extractions. This artifact might account for discrepancies found in the literature regarding PEA levels in human blood serum and other tissues. Conclusions: It is recommended to take into account this pitfall by analyzing potential contamination of the disposable glassware during the validation process of any method used for analysis of FAEs

    Synthesis, characterization and antibacterial activity of copper, nickel and bimetallic Cu–Ni nanoparticles for potential use in dental materials

    AbstractThe antibacterial effect is a desirable property in dental materials. Development of simple methods for the preparation of nanosized metal particles has attracted significant attention because of their future applications due to unusual size-dependent antibacterial properties. Copper (Cu), Nickel (Ni) and bimetallic Cu–Ni nanoparticles were prepared by a simple chemical method and their antibacterial activity was tested against the widely used standard human pathogens Staphylococcus aureus (gram-negative) and Escherichia coli (gram-positive). Additionally, these nanoparticles were tested against the dental pathogen Streptococcus mutans. Our results are promising for potential use in dental materials science

    Role of Dendritic Cells in Parasitic Infections

    Dendritic cells comprise a complex array of cell populations that play a leading role in immune defense. In an immature state, they have the capacity to sense and uptake different antigens. Upon capturing antigens, they become activated, mature, and migrate to lymph nodes where they present antigen-derived peptides to naïve T cells. Due to these excellent surveillance properties, dendritic cells play an important role against parasitic infections. Also, dendritic cells are an important source of IL-12, which is a fundamental proinflammatory cytokine in the control of intracellular parasites.     Predicting Earthquake-Induced Landslides by Using a Stochastic Modeling Approach: A Case Study of the 2001 El Salvador Coseismic Landslides

    In January and February 2001, El Salvador was hit by two strong earthquakes that triggered thousands of landslides, causing 1259 fatalities and extensive damage. The analysis of aerial and SPOT-4 satellite images allowed us to map 6491 coseismic landslides, mainly debris slides and flows that occurred in volcanic epiclastites and pyroclastites. Four different multivariate adaptive regression splines (MARS) models were produced using different predictors and landslide inventories which contain slope failures triggered by an extreme rainfall event in 2009 and those induced by the earthquakes of 2001. In a predictive analysis, three validation scenarios were employed: the first and the second included 25% and 95% of the landslides, respectively, while the third was based on a k-fold spatial cross-validation. The results of our analysis revealed that: (i) the MARS algorithm provides reliable predictions of coseismic landslides; (ii) a better ability to predict coseismic slope failures was observed when including susceptibility to rainfall-triggered landslides as an independent variable; (iii) the best accuracy is achieved by models trained with both preparatory and trigger variables; (iv) an incomplete inventory of coseismic slope failures built just after the earthquake event can be used to identify potential locations of yet unreported landslides

    Ceramic and Composite Polishing Systems for Milled Lithium Disilicate Restorative Materials: A 2D and 3D Comparative In Vitro Study.

    Purpose: This study aims to evaluate the effectiveness of two ceramic and two composite polishing systems for a novel chairside computer-aided design/computer-aided manufacturing (CAD/CAM) lithium disilicate ceramic with three-dimensional and two-dimensional microscopy images. This ceramic material can be used for implant-supported or tooth-borne single-unit prostheses. Materials and Methods: Sixty flat samples of novel chairside CAD/CAM reinforced lithium disilicate ceramic (Amber Mill, Hass Bio) were divided into five groups (n = 15/group) and treated as follows: Group 1 (NoP), no polished treatment; group 2 (CeDi), polished with ceramic Dialite LD (Brasseler USA); group 3, (CeOp) polished with ceramic OptraFine (Ivoclar Vivadent); group 4, (CoDi) polished with composite DiaComp (Brasseler USA), and group 5 (CoAs), polished with composite Astropol (Ivoclar Vivadent). The polished ceramic surface topography was observed and measured with three-dimensional and two-dimensional images. Results: All polishing systems significantly reduced the surface roughness compared with the non-polished control group (Sa 1.15 μm). Group 2 (CeDi) provided the smoothest surface arithmetical mean eight with 0.32 μm, followed by group 3 (CeOp) with 0.34 μm. Group 5 (CoAs) with 0.52 μm provided the smoothest surface among the composite polishing kits. Group 4 (CoDi) with 0.66 μm provided the least smooth surface among all polishing systems tested. Conclusions: Despite the effectiveness of ceramic polishing systems being superior to composite polishing systems of the CAD/CAM lithium disilicate restorative material, both polishing systems significantly improved the smoothness

    Computational Analysis of Hybrid Norwood Circulation with Distal Aortic Arch Obstruction and Reverse Blalock-Taussig Shunt

    BACKGROUND: The hemodynamics characteristics of the hybrid Norwood (HN) procedure differ from those of the conventional Norwood and are not fully understood. We present a multi-scale model of HN circulation to understand local hemodynamics and effects of aortic arch stenosis and a reverse Blalock-Taussig shunt (RBTS) on coronary and carotid perfusion. METHODS: Four 3-dimensional models of four HN anatomic variants were developed, with and without 90% distal preductal arch stenosis and with and without a 4-mm RBTS. A lumped parameter model of the circulation was coupled to a local 3-dimensional computational fluid dynamics model. Outputs from the lumped parameter model provided waveform boundary conditions for the computational fluid dynamics model. RESULTS: A 90% distal arch stenosis reduced pressure and net flow-rate through the coronary and carotid arteries by 30%. Addition of the RBTS completely restored pressure and flow rate to baseline in these vessels. Zones of flow stagnation, flow reversal, and recirculation in the presence of stenosis were rendered more orderly by addition of the RBTS. In the absence of stenosis, presence of the shunt resulted in extensive zones of disturbed flow within the RBTS and arch. CONCLUSIONS: We found that a 4-mm × 21-mm RBTS completely compensated for the effects of a 90% discrete stenosis of the distal aortic arch in the HN. Placed preventatively, the RBTS and arch displayed zones with thrombogenic potential showing recirculation and stagnation that persist for a substantial fraction of the cardiac cycle, indicating that anticoagulation should be considered with a prophylactic RBTS
