Mesenchymal-epithelial interaction mechanism has been applied in tissue engineering and regenerative medicine researches recently. Although many studies have been attempted to reconstruct HF using mesenchymal-epithelial mechanism, the regeneration of the complete HF in vitro is still challenging. In this study, we extracted dermal papillae (DP) cells and outer root sheath (ORS) cells from human scalp and used them to develop into human hair microfollicles (hHMF). Cultured DP cells were used as mesenchymal cells and ORS cells were used as epithelial cells to induce mesenchymal-epithelial interactions and de novo hHMF were developed by incubation in 3D hanging drop (HD) culture for 5 days. We extracted DP and ORS cells with a simple and effective method from the scalp of young patients and characterized cells by attaching specific antibodies through immunofluorescence staining. DP cells and ORS cells were cultured according to specific timeline that we established. Spontaneous localization of the single cells was observed after 5 days of HD incubation. The morphology of hHMF resembled HF in that hHMF has uni-directional structure and the DP spheroid resides in the hair bulb-like area. Also, immunofluorescence analysis of hHMF showed the expression of specific DP and ORS cell markers in the specific site of structure. This efficient hHMF development technique provides a potential to demonstrate the mechanism of HF development in vitro. Finally, an advanced culture system will allow screening of hair-related drugs in patients with hair loss or hair implantation with hHMF. [์˜๋ฌธ]Recently, a nerve conduit, which regenerates a missing nerve tissue by bridging the two nerve stumps by the tube, has been introduced as a promising alternative to autografts and allografts. Among a series of parameters that control the quality of the conduit, attention has been paid to permeability, tube dimension, mechanical property, and biodegradation rate, as well as to biocompatibility and neuron cell affinity. The objective of this study is to evaluate properties of poly(lactic-co-glycolic acid) (PLGA)- or poly(DL-lactic acid) (PLLA)-coated collagen conduits for nerve regeneration compared to those of grutaraldehyde(GA)-crosslinked collagen conduits, which has been widely used as collagen-based conduits. The coating was performed by spraying PLGA or PLLA solution over the only outer phase of the collagen conduit. The coated collagen conduit is expected to have bilayers that the outside of the coated collagen conduit improves physical and chemical properties due to coating of PLGA or PLLA and the inside of the coated collagen conduit has appropriate environment for nerve regeneration due to untreated collagen. First, spray-coating with PLGA or PLLA could control pore size and mechanical properties of porous collagen film with varying the concentration of polymer solution. In addition, the PLGA- or PLLA-coated collagen films degraded slowly enough to maintain a stable support structure for the entire regeneration process and showed mechanical property similar to nerve tissues. Moreover, the PLGA- or PLLA-coated collagen conduits showed proper swelling degree which could maintain the lumen space of the conduit constant. In the evaluation of biological properties for nerve regeneration, the PLGA- or PLLA-coated collagen conduits showed non-cytotoxicity similar to the untreated collagen film, and had high affinity and seeding ability to PC12 cells inducing proliferation and neurite outgrowth of the cells. Finally, the PLGA-coated collagen conduit resulted in excellent nerve regeneration 12 weeks after implantation in the rabbit, showing complete degradation of the conduit. However, the GA-crosslinked collagen conduit showed unmatched mechanical properties with nerve tissues, the decrease of hydrophilicity, moderate cytotoxicity, and low PC12 cell affinity. Therefore, the PLGA- or PLLA-coated collagen conduits consist of more effective materials for nerve regeneration, which improve adverse effects of the GA-crosslinked collagen conduits. Furthermore, spray-coating method will be able to be used in order to control pore size and mechanical properties of a fiber-shaped scaffold and strengthen limited region of the scaffold.ope
