    Effect of sorafenib and celecoxib combination therapy on proliferation of the human cholangiocarcinoma cell line SK-ChA-1 in vitro

    目的研究索拉非尼联合塞来昔布在体外对胆管癌细胞株Sk-CHA-1增殖的影响。方法体外培养人胆管癌细胞株Sk-CHA-1,通过MTT法检测索拉非尼单用或与塞来昔布联用时对胆管癌细胞株增殖的抑制作用,WESTErn blOT分析索拉非尼单用或与塞来昔布联用时对胆管癌细胞株内多聚AdP核糖聚合酶(PArP)蛋白表达的影响。结果索拉非尼抑制胆管癌细胞株Sk-CHA-1的增殖并诱导细胞凋亡。索拉非尼联合塞来昔布协同抑制胆管癌细胞株Sk-CHA-1的增殖。塞来昔布使索拉非尼诱导的胆管癌细胞株Sk-CHA-1的凋亡增加。结论索拉非尼联合塞来昔布能协同抑制胆管癌细胞株Sk-CHA-1的增殖,这与塞来昔布使索拉非尼诱导的细胞凋亡增加有关。Objective To investigate the effect of sorafenib and celecoxib combination therapy on proliferation of human cholangiocarcinoma(CC) cells,using the cultured SK-ChA-1 cell line.Methods Inhibition of SK-ChA-1 cell proliferation by sorafenib alone and in combination with celecoxib was studied in vitro using the MTT assay.The anti-neoplastic mechanisms of sorafenib alone and in combination with celecoxib were assessed by Western blot detection of changes in the caspase cleavage substrate poly ADP-ribose polymerase(PARP).Results SK-ChA-1 cells treated with sorafenib alone showed a dose-dependent growth inhibition and degradation of PARP.Combination treatment with sorafenib and celecoxib synergistically increased the growth inhibition effects and enhanced the degradation of PARP.Conclusion Combination treatment with sorafenib and celecoxib results in a synergistic anti-proliferative effect in the human CC cell line SK-ChA-1;the addition of celecoxib enhances sorafenib-induced apoptosis.国家自然科学基金资助项目(81072014

    适于HIV-1 p24抗原检测试剂的单抗制备与筛选

    目的制备抗HIV-1p24单克隆抗体(单抗,McAb),并利用双抗体夹心ELISA法建立HIV-1p24抗原检测试剂。方法以基因工程原核重组抗原HIV-1p24蛋白免疫BALB/c小鼠,利用常规杂交瘤技术和间接ELISA法制备单克隆抗体,单抗经纯化和HRP标记后,利用双抗体夹心ELISA筛选检测p24蛋白的最佳配伍单抗以期建立HIV-1p24抗原检测试剂。结果成功筛选到16株稳定分泌抗HIV-1p24单抗的杂交瘤细胞株,并从中筛选出最佳单抗配伍组合“16G12+2F2b-HRP”,该组合对p24蛋白的检测灵敏度为20pg/mL,对感染性克隆pNL4-3表达的HIV病毒培养上清检测呈阳性,对100份HIV阴性人血清无非特异性反应,显示出良好的检测灵敏度和特异性。结论本研究初步成功地建立了HIV-1p24抗原的检测试剂,并为最终建立HIV第四代诊断试剂(HIV Ag/Ab Assay)奠定了坚实的基础

The compound crystallised in the space group C2/c with a = 32.624(2)A˚ , b = 9.4814(4)A˚,c=33.395(2)A˚,b=109.339(5)◦ and V = 9746.8(9)A˚ 3. The crystal structure is a mixed-valence nonanuclear Co(II/III) oligomer, which contains a square-pyramidal CoII 5 core formed through m4-Cl and m3-alkoxo bridges and the pentanuclear CoII 5 core is surrounded by four peripheral Co(III) ions at the square-pyramidal base plane through m2-alkoxo bridges. Magnetic properties of the compound can be considered as the magnetism of the CoII 5 core, and the superexchange interactions between Co(II) ions within the core were calculated with three Co(II)–Co(II) superexchange parameters by using MAGPACK software package to give J1 = + 11.1 cm-1, J2 = -6.50 cm-1, J3 = -3.25 cm-1, D = + 6.0 cm-1 and g = 2.30.NNSF 20671079 20721001 MOE 107068 MSTC NCET-08 -0470 2007CB81530

    森林火灾是山东森林地区严重的环境问题之一。本研究采用山东2001—2010年MOD14A1每日1 km温度异常/火L3级产品与地形、植被、天气、人为和可访问性数据,分析评估了火灾发生原因;收集了林火发生/未发生相关的15个解释变量的空间数据,利用二项Logistic回归模型估计了解释变量的函数与林火存在的概率。结果表明,高火险区域主要集中在黄河三角洲、鲁西北平原,包括德州、菏泽、济宁、枣庄南部、临沂东南部;中火险主要在聊城、滨州、济南北部、淄博北部、潍坊东部、泰安、日照和青岛大部分地区(包括蒙山林区、沂山林区、五莲山林区、徂徕山林区、尼山林区、泰莱林区);低火险主要集中在济南南部、淄博南部、莱..

    多晶钙钛矿薄膜的故意掺杂及其在太阳电池中的应用

    作为光吸收层的多晶钙钛矿薄膜的形貌和光电子学特性对钙钛矿太阳电池的性能具有重要影响。文献报道了溶剂工程、组分工程等多种调控多晶钙钛矿薄膜形貌和光电子学特性的方法。通过溶液化学方法故意引入外来杂质元素可以对多晶钙钛矿薄膜实现连续定量的外掺杂,并有望改善多晶钙钛矿薄膜的光电子学特性。一般地,不同金属原子半径和电负性等的差别,会导致前驱体形核和结晶动力学的变化,因而在前驱体溶液中引入不同的金属阳离子,钙钛矿薄膜的结构、形貌和光电子学特性会发生相应的变化。另外,掺杂会引起晶格结合能的变化,从而对钙钛矿材料和器件的稳定性产生一定程度的影响。我们通过在前驱体溶液中引入外来金属离子实现了钙钛矿形貌和光电子特性的连续调控,这体现在晶格常数(图1a,b)、紫外可见光吸收谱(图1c)、稳态和瞬态PL谱(图1d,e)、晶粒粒径、电子能级和带隙等随掺杂浓度的变化方面。经过掺杂浓度的优化,所制备的钙钛矿太阳电池的器件效率得到了显著提升(图1f)
