    Preparation,identification and application of monoclonal antibody against urease subuint B of Helicobacter pylori

    目的制备抗幽门螺杆菌(HP)的单克隆抗体(MAb),并对该HP MAb进行分析鉴定,建立一种检测患者由于HP感染而在血清中产生HP抗体的竞争ElISA检测方法。方法用灭活的HP免疫bAlb/C小鼠,通过杂交瘤技术制备HP MAb。我们采用HP混合蛋白包括毒素相关蛋白A(CAgA)、空泡毒素A(VACA)和尿素酶以及灭活的HP菌体筛选阳性杂交瘤细胞株,用ElISA和WESTErn blOT等技术对所获得的HP MAb进行鉴定。利用辣根过氧化物酶(HrP)标记所筛选的HP MAb来建立一个可检测患者血清中HP抗体的竞争ElISA。结果通过大规模的杂交瘤筛选,我们选择了1株将其命名为C3 HP MAb,其抗体亚型为Igg2A,腹水效价可达1x107。WESTErn blOT法、ElISA和质谱检测结果显示,该C3 HP MAb能特异地识别HP的尿素酶b亚单位。用这个C3 HP MAb,我们建立了一种可检测患者血清中HP抗体的竞争ElISA。结论成功获得一种可以特异识别HP尿素酶b亚单位的MAb,建立了一种可检测患者血清中HP抗体的竞争ElISA。Objective To prepare and characterize the monoclonal antibody(mAb) against Helicobacter pylori(Hp)and establish a competitive ELISA used for detection of Hp antibodies in the sera of Hp-infected patients.Methods BALB /c mice were immunized with inactivated Hp to generate Hp mAb using the hybridoma technology.Hp mixed proteins including cytotoxin-associated gene A(CagA),vacuolating cytotoxin A(VacA) and urease,as well as inactivated Hp were applied to screen positive hybridoma.Selected Hp mAb was analyzed and characterized with ELISA and Western blotting,and then labeled with horseradish peroxidase( HRP) for establishing a competitive ELISA to detect Hp antibodies in the sera of Hp-infected patients.Results One Hp mAb named as C3 was selected after screening large amount of hybridoma,and the C3 Hp mAb was special for IgG2 a subtype with the affinity titer of 1 × 107.Western blotting,ELISA and mass spectrum analysis indicated that the C3 Hp mAb could recognize Hp urease subunit B specifically.Using the C3 Hp mAb,we developed a competitive ELISA which could be used to detect Hp antibodies in the sera of Hp-infected patients.Conclusion We successfully obtained one mAb that could specifically recognize Hp urease subunit B and developed a competitive ELISA using the Hp mAb.福建省科技重点项目(2011Y0050); 厦门市科技计划项目(3502Z20123009); 国家基础科学人才培养基金(J1310027

    针对利用光学探测设备对海底目标探测与搜索的使命,自治水下机器人(AUV)需具备更精确的航行控制性能,为此提出一种不依赖模型的改进型PID控制算法,通过对增设的左右水平推进器进行控制,以实现AUV低速时水平面的精确航行。将整个控制器分为两层:内层为偏航距离PID控制器,将输出量转化为所需偏转角;外层为航向PID控制器,将内层计算结果转化为航向偏差,对其进行PID计算,输出为偏转所需推力和推力矩值。通过对设定探测路线进行路径跟随,以反映水平面航行控制精度。通过湖上试验,得出精确的路径跟随航迹,实现了航向角偏差均值为0.09°、航向距离偏差均方差为0.29 m的航行稳定控制,验证了该控制方法的可行性..

    Abstract: In the 21st century computer technologies develop at a tremendous pace. Teachers teaching Chinese as a second language through smart searching engines and databases on Internet may collect, edit and upload contents to automatically generate teaching materials, thus combining methodology and curriculum with computer technologies to manage teaching on line. This study focuses on how an e-learning platform Moodle is being applied in our Intermediate Chinese Speaking and Listening Course, particularly on how it manages and supervises pace of study of students, and how it coordinates with teachers' classroom teaching, in order to examine its effectiveness. This paper also addresses the feedback from students on their perspectives of comprehensive computer technologies being employed in teaching Chinese as a second language, and the reflections of teachers towards teaching with Moodle

    目的:探讨中医专业人员进行五行成员生克关系判断时的大脑加工特征。方法:采用关系判断研究范式,将五行成员组成相生和相克关系词对,应用事件相关电位(ERP)和功能性磁共振成像(fMRI)技术,记录并分析被试者进行五行生克关系判断任务时的反应时、正确率、所诱发的脑电成分及脑区的激活情况。结果:行为学结果显示,相生关系判断的反应时小于相克关系判断(P&lt;0.01);ERP结果显示,2种条件均诱发200 ms事件相关电位正波(P200)、300 ms事件相关电位正波(P300)和晚期正成分(LPC)。平均波幅统计结果均为相生条件大于相克条件(P&lt;0.05)。fMRI实验显示,相克条件激活右脑顶下小叶区域;相克条件比相生条件额外激活右脑顶上小叶和额叶的中央前回区域;相生条件比相克条件额外激活右脑额叶的额内侧回、中央旁小叶、岛叶和左脑楔叶区域。进一步的感兴趣区(ROI)分析结果显示,楔叶(BA18)处和顶上小叶(BA7/8)处均表现为相生条件激活更为显著。结论:五行生克关系的大脑加工过程与分类关系的加工相似。相较于相克关系,相生关系加工占用更多的大脑认知资源。提示五行生克关系判断的策略是基于分类关系加工的认知策略,词对关系是否符合相生关系是分类策略加工的核心。</p

    针对低速状态下自治水下机器人(AUV)翼舵力不足的问题,提出了一种基于垂向推进方式的AUV低速近底稳定航行控制方案。基于航速因素,对比分析了翼舵和垂向推进器的推力特性,设计艏艉槽道垂向推进器布置方案。在此基础上通过改进PID(比例-积分-微分)控制策略,优化参数完成垂向运动控制.最后利用某型AUV进行湖上试验,对比翼舵推进方式,得出垂向推进器驱动方式具有更好的低速状态下近底定高航行操控性,实现了控制航行体高度均方差在0.1m、纵倾均方差在0.5&deg;以下的稳定航行

    目的:利用事件相关电位(ERP)和功能性磁共振成像(fMRI)技术揭示阴阳转化思维不对称性及其认知神经机制。方法:采用ERP和fMRI技术分别记录30名中医学专业研究生完成一项医学阴阳转化的判断任务过程中的反应时、正确率、脑电波信号和血氧水平依赖信号,并进行统计分析。结果:中医研究生判断阳转阴的反应速度快于判断阴转阳。ERP结果表明阴阳转化在认知加工后期的不对称性,与阴转阳在注意资源分配、工作记忆过程、语义分析与情境整合中存在优势相关。fMRI结果发现右侧脑岛和右侧额中回对阴阳转化认知的响应方式存在不对称,提示情绪加工和奖赏机制可能对维持阴转阳任务的工作记忆密切相关。而额中回或参与消极信息的抑制反向促进阴转阳任务的语义整合分析。结论:阴阳转化存在认知加工不对称性,大脑右半球对阴阳转化认知转化存在不同的加工模式。</p

    Concrete and Abstract Cognition of Yin-yang Analogical Reasoning in Traditional Chinese Medicine: An Event Related Potential Study

    目的:探索中医阴阳类比推理的大脑神经电生理活动的时间进程。方法:采用事件相关电位(ERP)技术对34名中医学专业研究生判断具体和抽象的中医阴阳类比过程的脑电波进行记录、比较。结果:行为学上抽象中医阴阳类比表现出了反应时优势。ERP研究结果发现,阴阳类比的抽象、具体和非阴阳目标域诱发的400 ms事件相关电位负波(N400)依次减小,提示其二元对立统一关系提取与语义整合难度依次增大。在阴阳类比的关系映射阶段,阴阳类比诱发的晚期正成分(LPC)大于非阴阳类比,但具体和抽象阴阳类比之间差异无统计学意义。结论:中医阴阳思维的核心是二元对立统一的关系提取与整合。中医学概念受阴阳思维的影响,表现出关系类比的具体和抽象类比映射趋同效应,可能与抽象概念的隐喻具象化信念和具体概念的关系类比抽象化信念双重强化有关。</p
