    低聚倍半硅氧烷基铁络合物阻燃剂的合成及改性环氧树脂

    采用Kabachnik-Fields反应合成出含有邻苯二酚功能团、磷杂菲基团和笼型倍半硅氧烷结构基元的新型配体POSS-DOPO-Catechol(PDC),再与Fe3+配位络合形成低聚倍半硅氧烷基铁络合物Fe(PDC)3,进一步加入双酚A型环氧树脂(EP)进行固化制备出EP/Fe(PDC)3复合材料。利用红外、质谱和核磁共振表征配体及络合物的化学结构;通过差示扫描量热分析、热重分析、氧指数测定、微形量热、动态力学实验等方法研究铁络合物Fe(PDC)3的引入对环氧树脂固化温度、热性能、阻燃性能、力学性能及介电性能的影响。结果表明,添加Fe(PDC)3可以降低环氧树脂的固化温度、热释放速率及总热释放量,提高残炭量、氧指数,这是由于铁络合物可以有效促进材料在高温下形成稳定性良好的致密炭层,起到隔氧隔热作用,从而在燃烧过程中阻止材料进一步燃烧。国家自然科学基金资助项目(51573150);;\n福建省自然科学基金计划资助项目(2018J01079);;\n福建省技术重大研发平台项目(2014H2006);;\n福建省科技重大项目(2013HZ0005-1

    茵陈蒿汤抗游离脂肪酸对HepG2细胞脂毒性作用的效应中药研究

    目的:探讨对游离脂肪酸(FFA)诱导人肝癌细胞株(HepG2)细胞脂肪沉积和对肿瘤坏死因子(TNF-α)分泌有显著抑制作用的茵陈蒿汤的效应中药或组合。方法:采用FFA诱导HepG2细胞脂肪沉积和TNF-α分泌的体外细胞模型和药物血清技术,运用数学模型"均匀设计法",根据复方中的3味中药(茵陈、栀子、大黄),选用U7(76)表进行组方设计所得6种中药组合进行干预,以对甘油三酯(TG)及TNF-α的抑制效应作为考察指标,筛选效应中药或组合,并重新区间分组验证。结果:茵陈和栀子在高剂量组合时有显著降低细胞TG及TNF-α含量的效应,与全方无显著性差异,其中单一茵陈也可显著降低细胞TG及TNF-α含量。结论:茵陈及其与栀子的组合是茵陈蒿汤抑制FFA诱导HepG2细胞脂肪沉积和TNF-α分泌作用的效应中药

    为探讨丝裂霉素C实验性血管外漏致皮肤溃疡的防护措施,我们以0.1Mg丝裂霉素C皮下注射bAlb/C小鼠后腿部,制成丝裂霉素C血管外漏性皮肤溃疡模型。在这模型上经8种不同药物或物理措施单独处理后10天所产生的皮肤溃疡面积与对照组(生理盐水封闭)相比较(经T检验后),差异有显著性的组分别是:外漏后及时用0.1Ml99%二甲基亚砜局部封闭或加用它在外漏表皮处外涂(P<0.001),0.1Ml5%碳酸氢钠(P<0.001),0.1Ml1%普鲁卡因(内含0.4%地塞米松)(P<0.001),0.1Ml90%二甲基亚砜(内含10%维生素E)外涂(P<0.01);外漏后半小时用0.1Ml99%二甲基亚砜封闭所产生的皮肤溃疡面积与对照组相比虽有显著差异(P<0.05),但溃疡防护能力较差。冰水冷敷或0.1Ml10%硫代硫酸钠(及时封闭)组与对照组相比差异无显著性(P>0.05)。本研究为临床上丝裂霉素C血管外漏的防护提供了药物选择的依据,文中还讨论了二甲基亚砜对丝裂霉素C外漏性皮肤溃疡防护的机理。in order to make an investigation on skin ulcer induced by Mitomycin C extravasation, the preventive measures were undergone by the Following ex-periment.A subcutaneous ulcerative model induced by 0.1mg Mitomycin C(MMC)extravasation in the leg of BALA/C mice was established.On the model subject, 8 pharmacologic adjuvants or physical modulation were adopted.It is preFerential to choose local injection with 0.1ml 99% dimethyl sulFoxide(DMSO),or with additional smearing DMSO on the aFFected skin.10 days later, the ulcerative areas with DMSO treatment were signiFicantly decreased in contrast with 0.1 ml 0.9% NaCI treatment (P0.05) .The mechanisms of prevention of MMC extravascular ulcer by DMSO or 5%NaHCO3 were brieFly discusse

    一株嗜盐古菌噬菌体的诱导及其性质的初步研究

    目前,已有15株嗜盐古菌噬菌体被报道。在已报道的15株嗜盐古菌噬菌体中,绝大多数分离于自然的盐湖、盐田等高盐的水体中:少数来自于溶源菌株的自发裂解。本研究首次报道了利用丝裂霉素-c从溶源性的嗜盐古菌Natrinema sp.F5中诱导分离了一株嗜盐古菌噬菌体--SNJ1,并对其性质进行了初步研究。噬菌斑的形态表明该噬菌体为温和噬菌体;电镜照片显示该噬菌体为头尾噬菌体。与已报道的嗜盐古菌噬菌体不同之处在于该噬菌体头部直径为67nm,尾部长约570nm,是目前报道的最大的嗜盐古菌噬菌体。研究还表明该噬菌体对NaCl具有很强的依赖性。NaCl浓度对该噬菌体活性保持、噬菌体对宿主的吸附以及噬菌斑的形成都有很大的影响。NaCl浓度低于23%时,3d时活性就有很大程度的下降: 当盐浓度为30%时最有利于噬菌体吸附,低于21%时噬菌体几乎不能吸附于宿主菌的表面: 当盐浓度低于19%时,该噬菌体在宿主菌的菌苔上不能形成噬菌斑

    目的探讨珍香胶囊对人宫颈癌的抗癌作用及其机制。方法建立人宫颈癌裸鼠移植瘤模型,分别应用TUNEL法和免疫组化SABC法观察不同浓度的珍香胶囊对裸鼠移植瘤凋亡指数(AI)增殖细胞核抗原阳性细胞指数(PI)的影响。结果经珍香胶囊治疗后,高浓度组和中浓度组AI分别为(2.83±1.52)%和(2.50±1.43)%均明显高于阴性对照组的(0.78±0.31)%(P<0.01,P<0.05),而低浓度组AI为(0.87±0.27)%与对照组比较,差异无显著性(P>0.05);高浓度组和中浓度组PI分别为(63.58±9.31)%和(68.83±9.04)%均低于阴性对照组(84.42±9.25)%(P<0.01),而低浓度组PI为(79.25±8.53)%与对照组比较,差异无显著性(P>0.05)。结论珍香胶囊通过抑制肿瘤细胞增殖、诱导肿瘤细胞凋亡而达到其抗人宫颈癌作用。Objective To investigate the antitumor effect and mechanisms of Zhenxiang capsule(ZXC)on human cervical carci- noma.Methods The impact of ZXC of various concentrations on the apoptosis index(AI)and proliferation cell nuclear antigen (PCNA)-positive nuclei cell index(PI)in nude mice with human cervical carcinoma were observed by using the TUNEL detection and SABC method.Results After treatment with ZXC the AI in rats of high and intermediate concentration groups was higher than that of negative controls(P<0.01 or P<0.05),PI in high and intermediate concentration groups was lower than that of negative group(P<0.01 ).Conclusion ZXC possesses antitumor effect on human cervical carcinoma through inhibiting proliferation and inducing apoptosis of tumor cells.广东省中医药局科研基金资助项目(课题编号102055

    线粒体是细胞能量代谢、生物合成和细胞死亡的调控中心,其功能异常会导致许多疾病的产生。线粒体蛋白质组学研究为解析线粒体的生理功能、探索线粒体相关疾病的发病机制及促进线粒体为靶向的药物研发提供重要理论依据。本文对线粒体蛋白质组学的研究方法,尤其是近年的技术发展以及线粒体蛋白质组学的性质及其应用进行总结,并对其未来的发展前景和挑战进行展望。Mitochondria play a central role in the regulation of cellular energy metabolism,bio-synthesis and cell death.Dysfunction of mitochondria can lead to many diseases.Mitochondrial proteomics provides important theoretical foundation for a systematic understanding of the biological functions of mitochondria,studying the mechanisms of mitochondria-related diseases,and promoting the research and development of mitochondria-targeting drugs.The methodologies,recent technology development,and characteristics and applications of mitochondrial proteomics were reviewed and the challenges and prospects were also discussed.国家自然科学基金(Nos.91313302;90913015;21225523)资助项目~

    线粒体是真核细胞能量代谢和信号转导的调控中枢,虽然各种灵敏、特异的线粒体荧光标记技术已经被广泛应用于线粒体研究,但仍缺乏单线粒体水平的荧光染色性能评估方法。基于超高灵敏流式检测技术(High sensitivity flow cytometry,HSFCM)能对单个线粒体进行高灵敏、高通量、多参数定量分析的独特优势,本研究发展了一种单线粒体水平的荧光标记高通量评估方法。将携带靶向线粒体绿色荧光蛋白基因的p Ac GFP1-Mito质粒转染至人宫颈癌He La细胞中,用G418筛选出稳定转染细胞,分别从瞬时转染和稳定转染的细胞中提取线粒体。此外,从未转染质粒的正常He La细胞中提取线粒体,分别进行Mito Tracker Green标记以及SYTO 62线粒体DNA染色,应用实验室自行研制的超高灵敏流式检测装置在单线粒体水平对这4种线粒体标记方法的荧光亮度、标记效率和稳定性进行评估。实验结果表明,稳定转染细胞中单个线粒体的绿色荧光蛋白(GFP)荧光亮度为瞬时转染的17.7倍,比Mito Tracker Green标记的线粒体亮度高约两个数量级,且标记稳定性好。本研究为线粒体标记方法的选择提供了一种先进的分析方法。Mitochondria play a central role in the regulation of energy metabolism and signal transduction in eukaryotic cells. Although many fluorescent labeling strategies have been developed for mitochondrial studies,the methods that enable labeling efficiency assessment at the single-mitochondrion level are still lacking. By employing the unique advantages of high sensitivity flow cytometry( HSFCM) in the sensitive,rapid,and quantitative multiparameter analysis of individual mitochondria,here we examined the performance of several different mitochondrial labeling strategies from the perspectives of brightness,labeling ratio,and stability.Mitochondria isolated from He La cells transfected with p Ac GFP1-Mito plasmid upon transient or stable transfections,and mitochondria directly labeled with Mito Tracker Green or SYTO 62 were analyzed by a laboratory-built three-channel HSFCM. Upon the quantitative measurement of fluorescence brightness,it was found that the fluorescence intensity of green fluorescent protein( GFP) in mitochondria isolated from cells with stable transfection was about 17. 7-fold higher than the transient transfection ones,and was approximately two orders of magnitude brighter than mitochondria labeled with Mito Tracker Green. On the other hand,the fluorescence signal of SYTO 62 labeling decreased upon washing,indicating its rapid dissociation rate. The strong fluorescence intensity and good labeling stability make stable transfection an efficient method to label mitochondria. The experimental results demonstrates that HSFCM provides a powerful analytical tool to assess the performance of mitochondrial fluorescence labeling via high throughput single mitochondria analysis.国家自然科学基金项目(Nos.91313302,90913015,21225523)资助~
