Study on epidemiology and pathogenic spectrum of HFMD cases in Xiamen, 2013-2016
手足口病由肠道病毒感染引起,是一种高发于0-5岁儿童的急性传染病,其在世界范围内出现多次大规模爆发流行,仅仅在中国大陆每年就有几百万例病例。手足口病的流行给公共卫生带来了威胁。目前仅有靶向EV71的手足口病单价疫苗上市,但由于肠道病毒病原的多样性,EV71单价疫苗可能无法有效控制疫情。因此对于手足口病病原谱的监测及主要病原分子进化的研究仍然十分重要。为此,本研究在前期建立的肠道病毒诊断分型方法的基础上,进一步完善检测体系,对厦门地区手足口病肠道病毒病原谱进行了全面的监测,并对主要病原的分子进化情况进行了分析: 1、首先对厦门地区手足口病流行情况和肠道病毒病原谱的组成及变化趋势进行了分析。结果...HFMD is a common infectious disease in young children caused by HEVs. 重组蛋白PTD-HSP27进入细胞保护视网膜神经节细胞抗氧化损伤
【目的】观察重组蛋白PTD-HSP27穿透视网膜视神经节细胞RGC-5的能力及其效率,测定重组蛋白PTD-HSP27对氯化钴(CoCl2)诱导的视网膜神经节细胞(RGC-5)细胞损伤的保护作用并初步探讨其机制。【方法】用FITC标记重组蛋白PTD-HSP27,荧光显微镜观察PTD-HSP27-FITC的穿透细胞膜的能力,用紫外分光光度计检测PTD-HSP27-FITC的穿膜效率,随后建立RGC-5细胞损伤模型。实验分为正常对照组、CoCl2损伤组、PTD-HSP27+CoCl2处理组,分别进行相应处理后,通过Annexin V-FITC/PI双染法检测各组细胞凋亡比例;Western Blotting检测各组细胞Bcl-2、Bax和Caspase-3等凋亡相关蛋白的表达水平。【结果】荧光显微镜下观察到重组蛋白PTD-HSP27-FITC能够有效通过细胞膜进入RGC-5细胞内,其进入细胞的效率为(47.29±2.33)%。CoCl2浓度依赖性地抑制RGC-5细胞存活率。比较正常对照组、CoCl2损伤组和PTD-HSP27+CoCl2处理组的检测结果示:PTD-HSP27能有效抑制CoCl2诱导的RGC-5细胞早期凋亡(P<0.01);PTD-HSP27能有效提高Bcl-2/Bax的表达比例,并抑制Caspase-3的激活(P<0.01)。【结论】重组蛋白PTD-HSP27能有效地穿过细胞膜进入RGC-5细胞内,且对CoCl2诱导的RGC-5细胞氧化应激损伤有保护作用,机制可能是通过调控Bcl-2/Bax的蛋白比例抑制细胞的早期凋亡
Development and Application of a Novel Neutralization Test for Echovirus 25
目的:建立一种新型的快速、高通量的埃可病毒25型(ECHO25)中和抗体检测方法,并初步评价其在ECHO25中和抗体筛选和血清流行病学调查中的应用价值。方法:应用免疫荧光方法筛选ECHO25高亲和性抗体并将其作为检测单抗,结合酶联免疫斑点检测技术(ELISPOT)建立ECHO25中和抗体检测方法;使用不同效价的血清评价该方法的准确性;采用所建立的中和方法对ECHO25单克隆抗体、临床血清样品进行检测。结果:建立了快速检测ECHO25中和抗体的Nt-ELISPOT方法,以ECHO25单克隆抗体5B9作为检测抗体;相比经典的中和实验方法 Nt-CPE,该方法可显著缩短检测时间(从5~7 d缩短至1 d以内),检测结果具有较好的一致性;采用所建立的Nt-ELISPOT方法首次筛选获得3株对ECHO25具有较好中和能力的单克隆抗体;临床血清样品检测结果显示厦门地区可能存在ECHO25的流行。结论:该方法可以应用于中和抗体筛选和血清学的临床辅助诊断,为ECHO25的防治研究提供支持。Objective: To establish a rapid and high-throughput neutralization test for echovirus 25(ECHO25),and evaluate its application in neutralizing antibody screening and seroepidemiological surveys. Methods: Immuno-fluorescence assay was applied to screen a high affinity antibody, which was used as the detection antibody forECHO25, and a rapid neutralization test was established based on enzyme- linked immunospot assay(Nt-ELISPOT). The accuracy of this method was evaluated by detecting serum samples with different titer. Monoclonalantibodies against ECHO25 and clinical serum samples were detected via the established neutralization test. Results: A rapid method to detect neutralizing antibody against ECHO25 was established and an anti-ECHO25 anti-body, 5B9, was used as the detection antibody. The detection period could be shortened significantly comparedwith the classical neutralization test(Nt- CPE)(from five to seven days to less than one day), and the Nt-ELISPOT had good consistency with the Nt- CPE. Meanwhile, three neutralizing antibodies for ECHO25 werescreened firstly by this method. The detection results of clinical serum samples showed that infection of ECHO25 might be popular in Xiamen. Conclusion: This method can be used in neutralizing antibody screening and seroepi-demiological surveys, and it may provide support for the control of ECHO25.国家自然科学基金(81371817,81401669
铜铝锰钛形状记忆合金的微观组织、超弹性和形状记忆效应
制备了5种掺杂Ti的Cu-Al-Mn基形状记忆合金,并对其显微组织、马氏体相变、超弹性和形状记忆效应进行研究.结果表明,Cu-Al-Mn-Ti形状记忆合金组织中存在两种L21相,包括L21-Cu2AlMn母相和L21-Cu2TiAl析出相.在变形过程中,应力诱发了L21-Cu2AlMn母相到马氏体相的相转变,由于存在弥散的L21-Cu2TiAl相,部分应力诱发马氏体相发生稳定化.因此,制备的5种成分的形状记忆合金(Cu-13.3Al-9.7Mn-4.3Ti、Cu-12.4Al-5.0Mn-4.3Ti、Cu-12.3Al-6.8Mn-4.2Ti、Cu-12.7Al-9.8Mn-2.1Ti和Cu-12.9Al-5.3Mn-2.8Ti)在室温下变形的同时具有超弹性和形状记忆效应.国家自然科学基金(51471138,51571168);;中央高校基本科研业务费专项(20720160078
组织政治氛围与组织绩效的关系研究
作为一种消极组织氛围,组织政治氛围(Organizational Political Climate)可能给组织结果带来诸多不利影响。加强组织政治氛围的研究,可为企业防治其消极影响提供理论指导。遗憾的是,目前组织政治氛围的研究尚处于起步阶段,与诸多组织层结果变量之间的关系尚缺乏理论分析与实证检验。为此,基于社会交换理论,本研究着重探讨组织政治氛围对组织绩效的影响,并考察了这一影响过程的中介机制与作用边界。通过问卷调研企业高层管理者,本文获得235家企业相关数据作为样本。实证检验结果表明:(1)组织政治氛围对组织绩效具有显著的负向影响——组织政治氛围越浓厚,企业的绩效表现越差;(2)知识分享在组织政治氛围与组织绩效之间起中介作用;(3)环境竞争性在知识分享与组织绩效的关系之间起调节作用——随着环境竞争性的提高,知识分享对组织绩效的正向影响会进一步加强。本研究丰富了组织政治氛围的相关文献,拓展了组织氛围、组织政治、知识分享等领域的研究,具有一定的理论价值,并为企业提供了相应的实践指导。教育部新世纪优秀人才支持计划(NCET-13-0892);;国家自然科学基金项目(71472116)资
Epidemiology and Evolution of the EV71-B5Subgenogroup in Asian-Pacific Region
EV71-B5亚型自2003年首次报道以来,在亚太地区频繁引发手足口病(Hand,foot and mouth disease,; HFMD)疫情,并可引起部分婴幼儿重症表现甚至死亡。为了阐明EV71-B5亚型在亚太地区的流行病学规律及进化特征,本文采用分子生物学软件(主要包; 括BioEdit、MEGA 5.0、Jmodeltest2.1.7、BEAST1.8.2、Tracer1.5; 、FigTree1.4.3、SPREAD1.0.7),对60条亚太地区的EV71-B5亚型序列进行进化分析及系统发育地理学分析。分析结果显示EV; 71-B5亚型可在同一地区(中国台湾、泰国等)频繁流行,最近共同祖先的起源时间为2000年9月(95%CI:1999年8月~2001年9月),平; 均进化速度为4.045*10~(-3)(95%CI:3.222*10~(-3); ~4.909*10~(-3));此外,迁移图谱表明EV71-B5亚型在亚太地区具有明显的迁移传播现象,中国厦门EV71-; B5亚型分离株(JN964686)推测来源于新加坡地区(BF=3.17)。这种现象提示我国需要加强对该亚型的监测力度,以免发生大范围暴发。Since its first identification,the EV71-B5 subgenogroup has been showed; to triggere hand,foot and mouth diseases in Asian-Pacific Region,and to; cause severe manifestations among some infants(even death).We wished to; understand the epidemiology,transmission,pathogenic origin and evolution; of the EV71-B5 subgenogroup.Molecular biology-based; softwares(BioEdit,MEGA3.0,Jmodeltest2.1.7,; BEAST1.8.2,Tracer1.5,FigTree1.4.3,SPREAD1.0.7)were used to make; phylogenetic trees and undertake phylogeographic analyses on 60; sequences of the EV71-B5 subgenotype.Results showed that EV71- B5; subgenotype could spread within the same district,and that its most; recent common ancestorwas inSeptember, 2000(95%confidence; interval(CI):August,1999 to September,2001).Median speed of evolution is; 4.045*10~(-3)(95%CI:3.222~4.909*10~(-3)).The EV71-B5 subgenotype has an; obvious rule of migration transmission,and we speculate that isolation; of the EV71-B5 subgenotype in southern China is derived from; Singapore(Bayes Factor=3.17).In conclusions,the EV71-B5 subgenotype; showsactive migration among the Asian-Pacific region,and our epidemic; surveillance should be strengthened.福建省自然科学基
Kinetic-resonance fluorescence determination of trace bisphenol A residue in water based on rhodamine 6G
目的建立环境水样中双酚A(bisphenol A,BPA)的罗丹明6G-动力学共振荧光测定法。方法向水样品中依次加入0.1 mol/L硫酸溶液0.6 ml,1.0×10-4 mol/L罗丹明6G溶液0.4 ml,0.1 mol/L溴酸钾溶液0.5 ml,70℃水浴加热12 min,流水冷却3 min,在524.2 nm处测定共振荧光强度。结果在0.02~1.40μg/ml线性范围内,该方法所得BPA的回归方程为ΔF=1 708.4c+33.2,r=0.999 8。该方法的检出限为0.014μg/ml,回收率为97.5%~106.5%,RSD为3.9%~5.0%。结论该方法简单、方便、灵敏,适用于饮用水中BPA的测定
Comparison of brain functional stability in patients with bipolar disorder in depressive and euthymic mood states
目的 分析处于双相情感障碍(BD)缓解期的患者(EP)和BD抑郁期的患者(DP)脑功能稳定性的特点。方法 选取2009年2月至2016年6月于首都医科大学附属北京安定医院就诊的56例BD患者为研究组,并根据患者所处的情绪状态分为EP组(n=30)和DP组(n=26)。招募与研究组患者匹配的71名健康对照者作为健康对照组。采用17项汉密尔顿抑郁量表(HAMD-17)比较EP组、DP组、健康对照组之间的抑郁程度。采集所有受试者的静息态功能性磁共振成像数据,采用DPABI软件对数据进行预处理和功能稳定性指标的计算并比较3组功能稳定性差异,变换不同窗宽、步长和窗的类型进行重复验证。采用Pearson相关分析患者右侧小脑后叶的功能稳定性与HAMD-17总分之间的相关性。结果 EP组、DP组、健康对照组受试者HAMD-17总分比较[2.00(0,4.00)分比14.00(10.75,19.50)分比0(0,1.00)分],差异有统计学意义(P<0.05);EP组患者右侧小脑后叶的功能稳定性低于DP组和健康对照组,差异有统计学意义(P0.05)。研究组、EP组和DP组右侧小脑后叶的功能稳定性与HAMD-17总分无相关性(P>0.05)。结论 EP、DP患者表现出不同的脑功能活动模式,仅EP患者表现出右侧小脑后叶的功能稳定性降低。</p
Radially Aligned Electrospun Fibers with Continuous Gradient of SDF1 alpha for the Guidance of Neural Stem Cells
Repair of spinal cord injury will require enhanced recruitment of endogenous neural stem cells (NSCs) from the central canal region to the lesion site to reestablish neural connectivity. The strategy toward this goal is to provide directional cues, e.g., alignment topography and biological gradients from the rostral and caudal ends toward the center. This study demonstrates a facile method for fabrication of continuous gradients of stromal-cell-derived factor-1 alpha (SDF1 alpha) embedded in the radially aligned electrospun collagen/poly (e-caprolactone) mats. Gradients can be readily produced in a controllable and reproducible fashion by adjusting the collection time and collector size during electrospinning. To get a long-term gradient, the SDF1 alpha is fused with a unique peptide of collagen-binding domain (CBD), which can bind to collagen specifi cally. Aligned CBD-SDF1 alpha gradients show stable, sustained, and gradual release during 7 d. Further, the effect of aligned CBD-SDF1 alpha gradients on the guidance of NSCs is investigated. It is found that the CBD-SDF1 alpha gradient scaffolds direct and enhance NSC migration from the periphery to the center along the aligned electrospun fi bers. Taken together, the tubular conduits based on radially aligned electrospun fi bers with continuous SDF1 alpha gradient show great potential for guiding nerve regeneration
