    COP1 and GSK3β Cooperate to Promote c-Jun Degradation and Inhibit Breast Cancer Cell Tumorigenesis

    近年来乳腺癌已成为全球妇女健康威胁最大的疾病之一。乳腺癌治疗的发展依赖于乳腺癌形成机制相关研究的进展。侵袭性乳腺癌细胞系及恶性乳腺癌肿瘤中常能检测到c-Jun蛋白表达的异常上升。其过表达对乳腺癌细胞的存活、增殖、转移和侵袭有重要促进作用。我们的研究证明在侵袭性乳腺癌细胞中,c-Jun蛋白过表达的主要原因是c-Jun缺乏泛素化修饰导致蛋白稳定性上升。在已知的c-Jun的特异性E3泛素化连接酶中,我们鉴定发现在侵袭性弱的乳腺癌细胞系中,组成型光形态建成1(constitutivelyphotomorphogenic1,COP1)介导了c-Jun的多泛素化,如果沉默COP1的表达,c-Jun的泛素化...Breast cancer is one of the most serious problems that threat woman health.     The Relationships Between Divergent Thinking, Mathematical Problem Finding, and Mathematical Achievement

    本研究先藉由文獻分析法瞭解擴散性思考、數學問題發現與數學學業成就三者的內涵,繼而依據它們的內涵並佐以學習、問題解決的角度,建立起三者間關係的假設,並透過實證調查研究法來驗證這些假設。本研究之研究對象為台北縣市五所國中的318位國三學生,研究工具為「新編創造思考測驗」、「數學問題發現測驗」、「第一次數學科基本學力測驗」,並以次數統計、集群分析、相關分析、變異數分析、逐步迴歸分析進行資料分析。本研究主要的研究結果如下:一、學生能夠發現各種思考產物類型與數學類型的問題。其中,關係性問題與發現性問題最多人提出,而單位性、類別性與驗證性問題則較少人提出。二、學生的數學問題發現型態有個別差異。三、擴散性思考與數學問題發現間為顯著中低度相關。四、擴散性思考與數學學業成就多為顯著中低度相關。五、數學問題發現與數學學業成就間為顯著中低度相關。六、能問大量且層次高數學問題的學生其數學學業成就比較不會問數學問題的學生為佳。七、擴散性思考之流暢力、數學學業成就、擴散性思考之變通力可以有效預測數學問題發現之問題數。八、擴散性思考之流暢力、數學學業成就、擴散性思考之變通力可以有效預測數學問題發現之問題獨特性。九、數學學業成就與擴散性思考之流暢力可以有效預測數學問題發現之問題品質。十、數學問題發現之問題品質、數學問題發現之問題數可以有效預測數學學業成就。本研究最後針對數學教育以及未來研究提出若干具體建議。First, this study probed into the contents of divergent thinking, mathematical problem finding, and mathematical achievement by literature review. Then the researcher made hypotheses of the relationships between divergent thinking, mathematical problem finding, and mathematical achievement based on the contents of them and the views of learning and problem solving, and designed survey research to examine these hypotheses. The subjects were 318 9th grade students from five junior high schools in Taipei county and Taipei city. The data- collection instruments included:(a) New Creativity Test; (b) Mathematical Problem Finding Test; (c) Basic Educational Indicator Tests of Mathematics. After utilizing frequency, cluster analysis, correlation analysis, ANOVA, and stepwise regression, the main results of this investigation are:(a) Students can find problems of all kinds of intellectual products and mathematics. Among them, problems of relations and problems to find were found most and problems of units and classes and problems to prove were found least ; (b) There are individual differences between mathematical problem finding styles; (c) The correlations between divergent thinking and mathematical problem finding are significantly positive; (d) Most of the correlations between divergent thinking and mathematical achievement are significantly positive; (e) The correlations between mathematical problem finding and mathematical achievement are significantly positive; (f) Students who can finds many high-level problems have higher mathematical achievement than those who can not; (g) Fluency of divergent thinking, mathematical achievement, and flexibility of divergent thinking can be used to predict the number of problems of mathematical problem finding effectively; (h) Fluency of divergent thinking, mathematical achievement, and flexibility of divergent thinking can be used to predict the rarity of problems of mathematical problem finding effectively; (i) Mathematical achievement and fluency of divergent thinking can be used to predict the quality of problems of mathematical problem finding effectively; (j) The quality of problems and the number of problems can be used to predict mathematical achievement effectively. Finally, the researcher brings up some suggestions on mathematical education and the future research

    研究降级路网条件下的组合出行交通平衡问题。首先,基于出行者对降级路网的不同风险态度,将出行者分为三类:守时型,冒险型和平均型。其次,考虑小汽车出行和小汽车换乘地铁出行两种出行模式,分析降级路网条件下不同用户类的组合出行行为,建立相应的多用户变分不等式交通分配模型,讨论模型解的性质。最后,设计模型的求解算法,并通过算例说明模型和算法的有效性。算例结果表明:不同出行时间预算的出行者在降级路网中的路径选择行为存在较大差异,路网降级程度对出行者出行选择行为影响显著;随着道路通行能力的降低,出行者更倾向于选择组合出行模式。To study the effect of the degradable road network on travel decision and traffic assignment, firstly all travelers were categorized into three classes: risk-averse travelers, risk-prone travelers and risk-neutral travelers. Secondly, two travel modes including car trip and car-subway trip were considered in a multi-modal networks. A stochastic traffic equilibrium model was proposed by using the variational inequality theory, and the properties of the solution were discussed. Finally, an example was used to testify the validity of the model and the algorithm. The results show that the travel behavior varies greatly with different travel time budget; with the decrease of traffic capacity, travelers prefer to choose the combined modes.Accepted versio

    A one-step dipstick assay for the on-site detection of nucleic acid

    National Science and High Technology Project [2011AA02A101]; National High Technology Research and Development Program 863 [SQ2010AA0222525001]; Fujian Nature Science Project [2009J06020]; Xiamen Science and Technology Project [3502Z20111048]Objectives: We have developed a one-step nucleic acid dipstick assay (NADA) for visually detecting polymerase chain reaction (PCR) products within 3 min. "One-step" means that there were no additional procedures between amplification and detection. Methods: This method was achieved through the use of asymmetric PCR and specially designed probes with appropriate melting temperature values. We initially combined one-step NADA with asymmetric capillary convective PCR (ACCPCR), an easy and rapid nucleic acid amplification technique, to construct an on-site nucleic acid diagnostic platform. Results: We developed a diagnostic assay for the hepatitis B virus based on the ACCPCR-NADA platform to verify its feasibility. It exhibited an analytical sensitivity of three copies per test and a broad detection spectrum including genotype A-I. It also showed 97.9% sensitivity and 100% specificity based on the results observed using 67 serum samples with the Roche COBAS AmpliPrep/COBAS TaqMan (COBAS) system as the standard for comparison. Conclusion: The results provide evidence for the feasibility of using an ACCPCR-NADA platform in practical applications, especially in on-site test.     由中国科学院计算机网络信息中心、中国科学院植物研究所、深圳市中国科学院仙湖植物园"三方两地"共同合作研究建设的"达尔文树"——分子数据分析应用环境(DarwinTree——Molecular Data Analysis and Application Environment),从中国陆地植物发育系统框架的研究出发,逐步推动解决生命之树构建过程中存在的技术难题,探索利用基因和基因组信息构建生命之树的策略和方法,研究和开发DNA序列信息自动采集和生命之树自动生成技术(Automatic Reconstruction of The Tree of Life),建立生命之树信息平台及其利用体系,为最终在我国建立具有国际影响的,能很好地兼容物种分类、地理分布、形态性状、化石信息以及DNA信息的物种库(Species Bank)创造条件。DarwinTree旨在为科研人员提供数据和分析并举的工作平台,该平台将承担数据汇集和面向实际科研工作应用的双重作用。本文发布的数据集包括:(1)DarwinTree基础数据集:来自国际公共序列数据的标记处理得到的分子标记数据及其与任意阶元物种分类名称对应的统计数据集;(2)DarwinTree自测序数据集:面向中国陆地植物研究的补充测序序列数据;(3)DarwinTree中国维管植物进化数据集:已构建的中国维管植物属系统发育树的数据(Generic tree of Chinese vascular plants)
