    Expression and functional analysis of recombinant α-bungarotoxin (P22-A31)

    α-银环蛇毒素在神经科学研究以及在临床和制药上有重要作用,为获得大量的α-银环蛇毒素,我们用已构建的pGEX-BgTX(P22-A31)质粒在大肠杆菌BL21(DE3)中表达,并用谷胱甘肽Sepharose FF纯化GST-α-银环蛇毒素融合蛋白,再用凝血酶切掉融合标签谷胱苷肽转移酶(Glutathione S-transferase,GST),得到了较纯的重组α-银环蛇毒素同工毒素,得率约为1.225mg/L。用重组α-银环蛇毒素制备多克隆抗体,经ELISA和Western杂交鉴定后可知重组α-银环蛇毒素与天然α-银环蛇毒素的抗原性一致。小鼠毒性试验表明,重组α-银环蛇毒素同工毒素的半致死剂量LD50为1.598mg/kg,约为天然α-银环蛇毒素的1/5。小鼠化学法镇痛试验显示,重组α-银环蛇毒素有一定的镇痛药效,1/4LD50剂量的镇痛百分率为55.2%,1/8LD50剂量的镇痛百分率为20.5%,在外周镇痛作用中呈一定的量效关系。α-bungarotoxin plays very important role in neuroscience research, clinical application and the pharmaceutical industry. In order to acquire quantitites of α-bungarotoxin, we expressed GST-α-bungarotoxin fusion protein using constructed plasmid pGEX-BgTX (P22-A31) in E.coli BL21 (DE3)cell, purified GST-α-bungarotoxin using glutathione-Sepharose FF, cleavaged Glutathione S-transferase(GST) tag by thrombin in the column, and finally, we obtained recombinant α-bungarotoxin with yields of about 1.225 mg/L. In order to compare the immunogenicity of recombinant protein and crude α-bungarotoxin, we prepared multi-antibody using recombinant α-bungarotoxin. The results of both ELISA and Western blot showed that recombinant α-bungarotoxin has the same antigenicity as natural α-bungarotoxin. In vivo toxicity tests showed that the LD_ 50 of recombinant α-bungarotoxin was 1.598 mg/kg, about 1/5 that of natural α-bungarotoxin. Analgesis percentages with doses of 1/4 LD_ 50 and 1/8 LD_ 50 were 55.2% and 20.5% respectively, indicating that recombinant α-bungarotoxin possesses analgesic efficiency.国家自然科学基金(No.30470938);; 教育部留学回国人员启动基金资助~

    探讨多壁碳纳米管对人肺上皮细胞A549核转录因子-κB(NF-κB)活性的影响及其活化机制.不同浓度的多壁碳纳米管作用于A549细胞后,用活性氧(ROS)敏感探针2′,7′-二氯荧光素二乙酸酯结合流式细胞仪检测细胞内氧化应激状态;用凝胶电泳迁移率改变这一分析技术检测A549细胞NF-κB DNA结合活性;用蛋白印迹检测A549细胞NF-κB p65蛋白和IκBα蛋白表达;用免疫荧光结合共聚焦显微镜观察A549细胞NF-κB p65蛋白的核转位情况.结果表明,多壁碳纳米管诱导A549细胞内ROS过量产生和NF-κB DNA结合活性;同时伴有p65蛋白核移位和IκBα蛋白胞浆降解.抗氧化剂N-乙酰半胱氨酸(NAC)可抑制多壁碳纳米管诱导的A549细胞内ROS产生、NF-κB DNA结合活性、p65蛋白核移位以及IκBα蛋白降解.结果表明,多壁碳纳米管可以通过诱导A549细胞氧化应激机制从而活化核转录因子NF-κB活性

    Molecular Cloning and Expression of an Isotoxin Gene,α-bungarotoxin,from Bungarus multicinctus

    α银环蛇毒素是存在于银环蛇蛇毒中的一种长链神经毒素,能与神经递质受体特异性结合,因此它不仅是神经信号传导机制研究的重要分子探针,更具有开发治疗某些神经性疾病新型药物的可能性。用SMART技术,反转录合成银环蛇毒腺cDNA,克隆并测定了α银环蛇毒素基因,得到14种mRNA序列;进而将其中一种新的α银环蛇毒素基因亚克隆到原核表达载体pQE30a和pGEX4T1中,在大肠杆菌中通过IPTG诱导表达,其中带有GST融合蛋白的重组质粒αBgTX(P22A31)/pGEX4T1在BL21菌株中得到高效表达,表达量占菌总蛋白的30%,可溶形式存在的融合蛋白大于25%。Snake venom contains a number of small proteins,enzymes and other components,which displays a broad spectrum of biological activities.With the ability of specifically binding on acetylcholine acceptor,α-bungarotoxins are not only useful molecular probes in investigating the mechanism of neural signal transmission,but also potential pharmic preparations for neural disease treatment.In current research,cDNAs of Bungarus multicinutus venom gland were synthesized using SMART~TM cDNA amplification kit and then,α-bungarotoxin genes were cloned and sequenced.Total of 20 clones were sequenced representing 14 isotoxin mRNAs of α-bungarotoxins.Among those clones,a novel isotoxin gene was subcloned into two expression plasmids,α-BgTX/pQE30a and α-BgTX/pGEX-4T-1,and transformed into E.coli.After inducing with IPTG,fused protein of GST-α-BgTX was successfully expressed at level of 30% gross proteins of bacteria.More than 25% of fused protein was in the soluble fraction and the rest in inclusion body.国家自然科学基金(编号:30470938);; 教育部骨干教师资助计划项目(编号:GG180210024031740);; 教育部留学回国人员启动基金资助~

    兰州重离子加速器(HIRFL)是一个回旋加速器组合系统.它的注入器是K=69的扇聚焦回旋加速器.在十多年运行过程中,曾做过两次较大的改进,使加速的束流种类及流强都有了显著的改善.但由于SFC的引出效率比较低,只有30%左右,一方面损失了大量束流,另一方面许多束流损失在引出静电偏转板上,造成了大量出气,破坏了真空,难以维持长期大束流运行.文章重新对SFC引出系统进行了物理设计研究,在真实磁场的基础上做了大量计算工作,得到了一个新的引出系统方案
