    使用电化学方法对2种不同的醇溶型无机锌车间底漆进行了研究,结合浸泡以及盐雾的实验结果,发现2种车间底漆中锌含量对阴极保护时效有影响,但不是决定车间底漆防锈能力的唯一因素,锌含量与防锈颜料对于车间底漆的防锈性能有协同效果。其中,锌粉在早期提供阴极保护的作用,而防锈颜料在后期对基底提供防锈保护。对于锌粉含量相对较高的车间底漆,锌粉并不是从一开始就全部参加阴极保护作用,而是一部分具有激活潜力的锌粉在另一部分锌粉率先反应之后才由\"未激活状态\"转变为\"激活状态\"进而提供阴极保护的作用

    Preliminary Study on Regulation of XIOsPR10 Gene Expression in Rice

    为探索水稻Pr10蛋白在水稻抗细菌性条斑病中的作用,构建了XIOSPr10基因的植物过量表达载体P1301-XIOSPr10及rnAI表达载体PdS1301-XIOSPr10,通过农杆菌介导分别转化水稻愈伤组织,获得了相应的再生植株。经guS检测和PCr分析,证实XIOSPr10基因以及rnAI片段分别整合到水稻再生植株基因组中;半定量rT-PCr分析显示,过量表达植株中XIOSPr10基因的表达量高于对照,而rnAI转基因植株中XIO-SPr10基因的表达被抑制。In order to study the function of rice XIOsPR10 gene which is related to the plant disease resistant reaction,we constructed plant over expression and RNAi expression vector and integrated into the genome of rice via Agrobacterium tumefaciens EHA105,respectively.The transgenic cultivars were identified by PCR and GUS gene expression detection.The analysis result showed that the transcription level of XIOsPR10 of over-expression transgenic rice was higher than that of control.In construct,XIOsPR10 gene expression was blocked in RNAi transgenic rice.科技部863专题(2007AA10Z132);国家重大科技专项(2008ZX08001-001;2009ZX08009-045B);教育部重点项目(01102

    Expression of pin1 gene in lung cancer and its significance

    背景与目的:Pin是人类高度保守的特异性磷酸化肽基脯氨酰顺及异构酶,它作用于脯氨酸所形成的肽键,并且仅使磷酸化pSer/Thr-Pro发生异构化,这一磷酸化后调控机制能诱导磷酸蛋白的构像变化,使其发挥功能。近来的研究显示这种新的调控机制在许多生理过程中起重要的调节作用,一旦失调便导致一系列人类疾病。如癌症患者体内Pin1表达异常增高,并调控多种癌基因的信号通路。本研究探讨pin1基因在非小细胞肺癌(NSCLC)中的表达及其意义。方法:利用免疫组织化学方法、定量PCR方法分别检测肺癌组织和正常肺组织中Pin1基因在翻译水平和mRNA水平上的表达差异。结果:在蛋白质水平上,Pin1的表达在正常/癌两种不同的肺组织中,表达量差异有显著性;而在mRNA水平上,Pin1的表达量在正常/癌两种不同的肺组织中,差异无显著性。结论:Pin1在NSCLC组织中的表达,可能是在翻译水平受到了调控;我们的研究进一步证实:Pin1在NSCLC组织中存在蛋白水平的过量表达,这对利用Pin1作为NSCLC组织检测标志物提供了理论依据。Background and purpose:Pin1 is a highly conservative enzyme that isomerizes only the phosphorylated Ser/Thr-Pro bonds in certain proteins, thereby inducing conformational changes. Recent results indicate that such conformational changes following phosphorylation are a novel signaling mechanism pivotal in regulating many cellular functions. Overexpression of Pin1 is prevalently found in human cancers.We studied expression of pin1 in non-small cell lung cancer and its implication in clinical. Methods:We detected the differential expression level of Pin1 protein in lung cancer specimen and normal lung tissues by immunohistochemical staining. Real-time quantitative PCR was also applied to detect the mRNA differential expression level in lung cancer tissues and normal lung tissues. Results:Pin1 protein is overexpressed in lung cancer at the protein level. On the other hand, the expression level of Pin1 mRNA in lung cancer tissue has no significant change compared with normal tissue. Conclusion:It demonstrated that the Pin1 expression in lung cancer might be regulated by translation mechanism.Our data implicate that Pin1 may serve as a valuable molecular marker for human cancer

    酪氨酸激酶抑制药GW2974诱导乳腺癌细胞BT474耐药的代谢机制研究

    目的通过比较酪氨酸激酶抑制药GW2974给药后母本细胞株BT474与耐药株r BT474之间代谢相关因子mRNA表达的差异,探讨r BT474可能的耐药机制。方法收集对数生长期r BT474和BT474细胞,接种于96孔板,加入含有GW2974的培养基,分别于孵育0,12,24和48 h后,通过噻唑蓝法检测BT474与r BT474细胞的增殖情况。用逆转录实时定量-聚合酶链反应检测12种相关代谢因子葡萄糖转运体4(GLUT4)、2,6-二磷酸果糖激酶(PFK-2)、丙酮酸激酶2(PKM2)、乳酸脱氢酶(LDHA)、丙酮酸羧化酶(PC)、6-磷酸果糖激酶(G-6-PD)、脂肪酸合成酶(FASN)、脂酰肉碱转移酶1 (CPT1A)、葡萄糖调节蛋白75(GRP75)、电压依赖性阴离子通道蛋白1 (VDAC1)、葡萄糖调节蛋白78(GRP78)和钙网蛋白(CALR)的mRNA在BT474细胞和r BT474细胞中给药前后的表达水平。结果给予GW2974后,母本细胞BT474中糖脂代谢相关因子GLUT4、PFK-2、PKM2、LDHA、PC、G-6-PD、FASN和CPT1A的mRNA表达水平均明显下调,线粒体应激因子GRP75、VDAC1与内质网应激因子CALR表达水平均明显下调,内质网应激因子GRP78表达明显上调。而在耐药株r BT474中给药后这12种代谢相关因子的mRNA表达水平均明显上调。结论耐药株r BT474可能通过调控糖脂代谢以及细胞应激等过程使细胞耐受性增强。福建省自然科学基金面上项目(2012J01417

    对比增强3D STIR SPACE序列对子宫内膜异位症患者骶丛神经异常的显示

    目的探讨增强 3D STIR SPACE序列对盆腔子宫内膜异位症患者骶丛神经异常的显示价值,并与增强3D VIBE序列进行对比。方法病例组包括30名子宫内膜异位症患者,对照组包括20名健康女性,均接受盆腔MRI检查,包括对比增强3D STIR SPACE序列和增强3D VIBE序列。评估骶丛神经显示的图像质量以及子宫内膜异位症患者中的骶丛神经侵犯。结果对照组和病例组中,3D STIR SPACE序列和3D VIBE序列均能很好地显示骶丛神经纤维(P<0.001)。阅片者应用3D-STIR-SPACE序列对病变的诊断信心分数一致性较强(Kappa=0.684,P<0.001),应用3D-VIBE序列的诊断信心分数一致性一般(Kappa=0.130,P=0.386)。结论对比增强T2-SPACE序列和3D VIBE序列能够清晰显示骶丛神经,前者对盆腔子宫内膜异位症患者骶丛神经异常的显示更有优势

    Study on the Relationship between Structure of Supramolecular Ion Material and Performance of Humidity Sensing

    相对湿度是许多领域的关键参数,环境湿度与人们的生活密切相关,因此对湿度进行测量和控制是各个领域中值得关注的问题之一. 在前期的研究中,作者制备了一种新型的超分子离子材料(SIM),它是由基于咪唑的双阳离子(如1,10-双(3-甲基咪唑-1-基)癸烷,C10(mim)2)和电活性二阴离子(如2,2\'-连氮基-双(3-乙基苯并噻唑啉-6-磺酸),ABTS)组成的,发现其对湿度具有敏感且快速的响应. 在此基础上,本文制备了6种不同碳链(C4,C6,C8,C10,C12,C14)的咪唑基化合物,发现其中3种(C10,C12,C14)可与ABTS形成水稳定的SIM. 循环伏安法、计时电流法以及石英晶体微量天平表征了这些超分子离子材料的湿度传感性能,发现基于C12的SIM具有最佳的湿度传感性能. 同时,SEM结果显示随着碳链的增加,离子材料的厚度变薄并且形态变得不规则. 因此,作者认为疏水作用和材料比表面积均会影响湿度传感的灵敏度. 本研究为发展新的湿度响应的离子传感材料奠定了基础.Humidity measurement and control is one of the most notable issues in various areas, such as climate, industry, agriculture, electronics, especially human comfort and health. In our previous study, we have found that a new kind of supramolecular ionic material (SIM), consisting of an imidazolium-based dication (e.g., 1,10-bis(3-methylimidazolium-1-yl) decane, C10(mim)2) and electroactive dianionic (e.g., 2,2\'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid), ABTS), shows ultrasensitive and ultrafast response towards humidity sensing. Herein we prepared six kinds of imidazolium-based dications with different carbon chain lengths (i.e., C4, C6, C8, C10, C12, C14), and found that three of them (i.e., C10, C12, C14) could form water-stable SIM with ABTS. Cyclic voltammetry, chronoamperometry and quartz crystal microbalance were used to characterize the humidity sensing performance of these supramolecular ionic materials. The results show that the C12-based SIM has the best humidity sensing performance compared with the other two kinds of SIM. Moreover, SEM images show that the thickness of the ionic material became thinner with the increase of the carbon chain length. We, thus, conclude that both the hydrophobic interaction and the specific surface area of SIMs would contribute to the performance of humidity response.
