The Molecular Mechanism of the Regulation of AMPK Phosphorylation and Activity by Hypo2

Abstract

腺苷酸活化蛋白激酶(AMP-activatedproteinkinase,AMPK)是细胞重要的“能量感受器”,在调节代谢和能量平衡等方面具有重要的作用。近年来研究显示,AMPK不仅在调节糖脂代谢、血压及免疫功能等方面扮演重要角色,同时还参与了多种代谢性疾病的发生和发展,因此AMPK相关作用机制的研究可能为肥胖、糖尿病(DM)、代谢综合征(MS)和肿瘤的治疗提供新的途径。我们实验室检测到了一个崭新的与AMPK相互作用的蛋白命名为Hypo2,它们的结合随着葡萄糖饥饿时间的延长而增强。我们发现随着Hypo2的表达量上升AMPKα1的第172位苏氨酸的磷酸化水平也随之上升,从而上调了AMPK的活性。...As a metabolic sensor, the serine/threonine protein kinase AMP -activated protein kinase(AMPK) promotes the adaptation of cells to signals arising from nutrients, hormones, and growth factors.
