    The role of Mycoepoxydiene in inhibiting the progression of human cholangiocarcinoma (CCA)

    环氧二烯(MED)是海洋木栖真菌菜豆间坐壳菌(Diaporthephaseolorum)的次级代谢产物,是一个以含有氧桥的八元环二烯为骨架的化合物,已有研究发现其具有很好的抗肿瘤活性,但是该化合物在胆管癌发生和发展过程中的作用并不清楚,我们用胆管癌细胞株SK-ChA-1和Mz-CHA-1通过细胞水平首次揭示了MED在胆管癌中的抗肿瘤活性。 首先我们通过MTT试验发现MED显著抑制胆管癌细胞的生长,这提示我们MED在胆管癌中可能具有良好的抗肿瘤活性。接着我们进一步研究了MED抑制胆管癌细胞生长的机理。通过Westernblotting实验我们发现MED显著诱导胆管癌细胞的凋亡,并且上调抗凋亡蛋...Mycoepoxydiene(MED)is a secondary metabolic product of Diaporthe phaseolorum which contains an oxygen-bridged cyclooctadiene core and an α,β-unsaturated δ-lactone moiety.     Down-regulation of TSPO expression doesn't affect the productions of TNF-α,IL-1β and IL-6 in LPS-stimulated BV-2 microglia

    目的研究相对分子质量(Mr)18 000转运蛋白(TSPO)基因表达下调对脂多糖(lPS)诱导bV-2小胶质细胞分泌Tnf-α,Il-1β和Il-6的影响。方法以rnA干扰技术建立TSPO基因表达下调的细胞模型,实时荧光定量PCr(QrT-PCr)和WESTErn blOT法检测转染TSPO SIrnA bV-2细胞TSPO MrnA及蛋白水平表达的效果;用QrT-PCr法检测TSPO基因下调后小胶质细胞bV-2在lPS作用下分泌Tnf-α、Il-1β和Il-6的MrnA水平表达的情况;ElISA检测TSPO基因下调小胶质细胞bV-2在lPS作用下分泌Tnf-α、Il-1β和Il-6的蛋白水平表达的变化。结果成功建立了TSPO基因下调的细胞模型,稳定表达TSPO SIrnA细胞的TSPO MrnA和蛋白水平表达均明显下降,TSPO基因下调后bV-2细胞在lPS作用下分泌Tnf-α、Il-1β和Il-6的量无变化。结论下调TSPO的表达对lPS刺激引起的小胶质细胞Tnf-α、Il-1β和Il-6的分泌无明显影响。Objective To evaluate the effect of translocator protein( TSPO,Mr18 000) on the productions of TNF-α,IL-1βand IL-6 in lipopolysaccharide( LPS)-stimulated BV-2 cells.Methods RNA interference technique was used to decrease TSPO expression in BV-2 cells.Western blotting was performed to assess the level of TSPO protein in BV-2 cells transfected with TSPO siRNA.Then the mRNA and protein levels of TNF-α,IL-1β,and IL-6 were measured in LPS-stimulated BV-2 cells by real-time quantitative PCR( qRT-PCR) and ELISA,respectively.Results The level of TSPO protein obviously decreased in BV-2 cells transfected with TSPO siRNA.However,knockdown of TSPO had no effect on the productions of TNF-α,IL-1βand IL-6 in LPS-stimulated BV-2 cells.Conclusion Down-regulated TSPO is not directly involved in regulating TNF-α,IL-1βand IL-6 productions induced by LPS in microglia.国家自然科学基金(81071182); 福建省医学创新项目(2009-CXB-46); 厦门大学生命科学学院细胞生物和肿瘤工程教育部重点实验室开发基金(2009101); 福建医科大学非直属附属医院科研发展专项基金(2008031

Methods: Totally 349 middle school students aged 12 - 19 years were selected, including junior high school students and senior high school students. They were assessed with the Adolescent Online Aggressive Behav- ior Scale (AOABS), Adolescent Aggressive Behavior Questionnaire, and State-Trait Anger Expression Inventory-2 (STAXI-2). Results: Girls got significantly higher scores than boys on anger temper factor [ (2. 0±0. 7) vs. ( 1.9±0. 7), P 〈0. 05]. Senior high school students got significantly higher scores than junior high school students on trait anger subscale [ (2. 1±0. 6) vs. ( 1.9±0. 6), P 〈 0. 01] and anger expression-out factor [ (2. 3±0. 5) vs. (2. 2±0. 6), P 〈 0.01 ]. Aggressive behavior, no matter online or offline, were positively correlated with trait anger and anger expression (r = 0. 14±0. 57, Ps 〈 0.05). Meanwhile, significant negative correlations were noted between the factors of anger control and offline aggressive behavior as well as online instrumental aggressive behavior ( r = - 0. 19 - -0. 11, Ps 〈0.05). The results of stepwise regression showed that the scores of anger reaction and anger expression were positively correlated with scores of aggressive behavior(β = 0. 13 -0. 37, P 〈 0. 05). The score of anger control-out was negatively correlated with the score of reactive aggression(β= -0. 01, P 〈 0. 05). Explained variation rate of aggressive behavior ranged from 19. 1% to 35.7%. Conclusion: The online aggression amongmiddle school students are probably related with trait anger and anger expression

    Impact of sleep duration on cognitive functions among preschoolers

    目的:探讨学龄前儿童白天睡眠以及晚上睡眠对其注意力、记忆力以及语言能力等认知能力的影响.方法:采用皮博迪图片词汇测验、图片划销测试以及空间工作记忆任务对杭州和北京的94名学龄前儿童的认知能力进行测试,并采用自编问卷调查儿童的人口学相关情况以及白天和晚上的睡眠情况.结果:学龄前儿童白天睡眠时长以及总睡眠时长的年级差异有统计学意义(F0.05(3,90)=6.346,P=0.001;F0.05(3,90)=2.925,P=0.038),晚上睡眠时长差异无统计学意义.晚上睡眠时长以及总睡眠时长与儿童的注意力和工作记忆广度呈显著正相关(r=0.202~ 0.282),与语言能力的相关差异无统计学意义.白天睡眠时长与各认知测试的得分相关均无统计学意义.回归分析结果显示,总睡眠时长对儿童的注意力以及工作记忆广度有较好的预测作用,进一步分析显示,晚上睡眠时长对儿童的注意力以及工作记忆广度有较好的预测作用.结论:总睡眠时长尤其是晚上睡眠时长可能影响学龄前儿童的注意力以及工作记忆等认知能力.中国科学院心理健康重点实验室经费、“十二五”国家科技支撑计划重点项目; 北京市教育科学十二五规划项目; Fund of Key Laboratory of Mental Health,Institute of Psychology,Chinese Academy of Sciences; the Mega-projects of Science Research for the 12th Five-Year Plan; the Project of 12th Five-Year Plan on Beijing&amp;#39;s Education中文核心期刊要目总览(PKU)中国科技核心期刊(ISTIC)中国科学引文数据库(CSCD)06933-9374

    目的建立注意缺陷多动障碍患者(AdHd)哌甲酯缓释片清除率的群体药动学模型。方法前瞻性收集闽南地区服用哌甲酯缓释片治疗的78例AdHd患者的消除相血药浓度(n=165),采用非线性混合效应模型(nOnMEM)法分析数据。结果最终模型为Cl(l·H-1)=254·(WT/34.7)0.663·0.884(如果合用丙戌酸钠,否则为1)·EXP(ETA(Cl)),WT为体质量(kg)。结论成功建立AdHd患者哌甲酯缓释片治疗的群体药动学模型,根据患者的生理用药资料,可估算其清除率,为制定给药方案提供依据。AIM To establish a clearance model for methylphenidate prolonged-release tablets(MPH)in patients with attention deficit/hyperactivity disorder(ADHD)by population pharmacokinetic method.METHODS Serum concentration data(n=165)in elimination phase were collected prospectively from 78 ADHD patients in south Fujian during their routine clinical treatment.Nonlinear mixed effects modeling(NONMEM)was employed to establish the population pharmacokinetic model of the MPH clearance(CL).RESULTS The final model was:CL(L·h-1)=254·(WT/34.7)0.663·0.884(if taking sodium valproate,otherwise it equaled to 1)·EXP(ETA(CL))in which WT was weight(kg).CONCLUSION A population pharmacokinetic model is established successfully and proposed to estimate the individual CL for patients taking MPH in terms of patients' characteristics and dosing history,and to establish a prior dosage regimen.福建省卫生厅青年科研课题(编号2013-2-02); 厦门市卫生局科研立项(编号3502Z20077079
