Expression of Fusion Protein of Parathyroid Hormone and Transferrin N-terminal Half-molecule in Pichia pastoris
利用重叠PCR技术将PTH(parathyroidhormone,甲状旁腺激素)基因与TFN(transferrinN_terminalhalf_molecule,转铁蛋白N端半分子)基因在体外融合,融合基因克隆至真核表达载体pPIC9中,转化毕赤酵母GS115。转化子经甲醇诱导后,融合蛋白得到了表达并分泌到发酵上清液中。经SPSepharoseFF阳离子交换层析、PhenylSepharoseFastFlow疏水层析纯化获得了纯度大于95%的PTH_TFN样品。Westernblot分析及腺苷酸环化酶实验证明融合蛋白中的PTH具有与抗PTH抗体结合能力及刺激腺苷酸环化酶的活性,铁饱和实验证明融合蛋白中的TFN和单独的TFN具有相同铁结合能力。因而TFN可望作为PTH的天然运输载体。The fused gene (PTH_TFN) of parathyroid hormone (PTH) gene and transferring N_terminal half_molecule (TFN) gene was amplified by multiple PCR and inserted into pPIC9 vector. The recombinant plasmid pPIC9_PTH_TFN was transformed into Pichia pastoris GS115 by PEG. After methanol induction, the target protein was expressed in fermentation supernatant at high level.The fused protein PTH_TFN with purity being higher than 95% was finally obtained after purification through two_step chromatography : SP Sepharose Fast Flow and Phenyl Sepharose Fast Flow.Western blot analysis and adenylate cyclase assay proved that the fused protein exhibited the bioactivity to stimulate cAMP synthesis and the ability to bind Fe ~3+ in the Fe ~3+ saturation study as the recombinant TFN did indicating that TFN could be used as the transcellar carrier of PTH.国家高技术研究与发展项目基金资助(No.2004AA215172)。~
先驱体法制备连续SiC自由薄膜及其发光性能
以聚碳硅烷(polycarbosilane,PCS)为先驱体,熔融纺出连续PCS自由原膜,并在190℃下对其进行1,2,3h和6h氧化交联,在900℃预烧及最终分别在1200℃和1300℃烧成,制得系列SiC自由薄膜。采用红外光谱、Raman光谱、X射线衍射、透射电镜与扫描电镜对薄膜进行微观结构与形貌分析。测量了薄膜的室温光致发光特性。结果表明:连续SiC自由膜均匀致密,含有β-SiC微晶、无定形SiOxCy及C簇;薄膜在410~450nm范围内有较强的蓝光发射,1200℃烧结的薄膜随交联时间增加,发光强度增大;而1300℃烧结的薄膜的发光强度相对下降,且交联时间越长强度下降越明显。412nm发光峰可归结于C簇发光;而435nm附近的峰则是薄膜中富含的Si—O,Si—C等键中的缺陷态构成的发光中心,在β-SiC晶粒中电子受到激发与缺陷态产生辐射复合引起发光以及量子表面效应共同作用的结果
连续含铝SiC自由膜的制备与发光特性研究
通过自制喷膜装置对聚铝碳硅烷(PACS)进行脱泡处理、熔融纺膜,并对其进行氧化交联、高温预烧及高温裂解终烧可制得连续含铝SiC自由薄膜。用扫描电镜(SEM)分析薄膜的形貌,通过红外光谱(FT-IR)分析氧化交联后薄膜的结构变化,通过电子探针(EPMA)、拉曼光谱(Raman)、X射线衍射(XRD)与场发射高分辨透射电子显微镜(HRTEM)对薄膜进行成分及微观结构分析,采用光致发光谱(PL)对薄膜的光学带隙和发光特性进行了研究。结果表明,熔融纺膜法与PACS先驱体法相结合可制得均匀、致密的耐高温连续含铝SiC自由薄膜,室温下表现出了320~440nm宽谱带发光,其发光峰可分别归因于-αSiC和C簇,且随着烧结温度的提高,发光强度增大
纪念五四宪法颁布暨全国人民代表大会成立五十周年理论研讨会纪要
今年 9月是五四宪法颁布、全国人民代表大会成立五十周年。五四宪法是新中国的第一部宪法 ,为我国的社会主义民主法制奠定了基础 ,具有划时代的意义。坚持和完善人民代表大会制度 ,是新的历史条件下推进政治体制改革和社会主义政治文明建设的重要内容。基于“以学术表达纪念、以纪念促进学术”的目的 ,中国社会科学院法学研究所于 2 0 0 4年 9月 5日举办了“纪念五四宪法颁布暨全国人民代表大会成立五十周年理论研讨会”。来自全国各地的 70余位代表莅临会议 ,研讨了诸多热点和前沿问题
3D-printed integrative probeheads for magnetic resonance
射频探头前端作为核磁共振设备的核心部件之一,极大程度的决定着系统实验性能的优劣。探头前端通常由射频线圈、射频电路及样品检测管道等部分组成。现有的射频线圈制作技术主要是通过手工或机械手段按照所需的线圈形状进行绕制。但是,当线圈结构较为复杂、不规则,或体积尺寸较小时,常规绕制方法便难以满足结构设计和制造的精度需求,因此造成线圈性能的劣化,增大检测区域的射频场不均匀性,对核磁共振检测产生负面影响。本研究中,利用3D打印熔融沉积制造或光敏树脂选择性固化技术精确加工出一体化磁共振探头前端,使用常温液态金属填充线圈模型管路形成射频线圈,搭建出稳定的一体化磁共振射频探头。利用高精度3D打印和液态金属灌注技术制备出包含有射频线圈和定制化样品管道结构在内的一体化磁共振射频探头前端,克服了传统磁共振三维微型线圈成型困难、与样品腔匹配程度差等问题,提高了探头的信噪比,为定制化的磁共振检测提供了新思路。

However, the fabrication of MR radio frequency probeheads still face difficulties in integration, customization and miniaturization. Here, we utilized 3D printing and liquid metal filling techniques to fabricate integrative radio frequency probeheads for MR experiments. The 3D-printed probehead with micrometer precision generally consists of liquid metal coils, customized sample chambers and radio frequency circuit interfaces. We screened different 3D printing materials and optimized the liquid metals by incorporating metal microparticles. The 3D-printed probeheads are capable of performing both routine and nonconventional MR experiments, including in situ electrochemical analysis, in situ reaction monitoring with continues-flow paramagnetic particles and ions separation, and small-sample MR imaging. A Study on the Competitive Strategies in the e-reader Industry:An Application of Scenario Analysis
電子書閱讀器在iPad及平板電腦未出現前,以電子紙為主的閱讀器是電子書閱讀的主流產品,但在iPad及平板電腦出現後,電子書閱讀器面臨強大的競爭威脅。面對未來競爭的不確定性,電子書閱讀器是否能主宰閱讀器市場,抑或證明只是過渡期的產品,是個值得深入探討的問題。
本研究選擇電子書閱讀器、平板電腦裝置兩大陣營作為分析對象,先以產業分析探討電子書閱讀器業者所面對的技術發展與產業環境,同時探討數位內容業者所扮演的角色,對電子書閱讀器業者未來發展所造成的影響。除了對產業現狀進行了解外,本研究利用產業情境模擬分析法,建構未來產業可能的產業情境,並加以分析,對電子書閱讀器產業內的主要廠商,提出以下的策略建議:
一、對於電子書閱讀器的品牌業者的策略建議: 兩面下注
對於Amazon和B&N兩品牌業者而言,他們對於電子書閱讀器相關的電子紙技術及硬體的設計開發,並不需投入資源與設備投資。在面對平板電腦的興起與彩色化的需求,品牌業者很明確地可以兩面下注,吸收各種不同需求的客戶。對於需要優越閱讀感受的客戶,業者可提供電子紙式的閱讀器。而對於需要多媒體功能的消費者,業者可提供以LCD顯示的閱讀器或平板電腦。
二、對於電子紙業者的策略建議: 保留彈性
iPad及平板電腦的興起,電子紙業者受到極大的衝擊與不確定性,而其投入龐大的資金與技術開發,使得此類業者有很高的退出成本。因此建議此業者對於新技術開發與投資應保留彈性,以目前擁有的技術和資源,降低成本,提升競爭力。待情勢明朗後,再作出對自己有利的決策,以降低不確定性的風險。
此外,由於我國擁有豐富的電子紙專利,並擁有完整的產業供應鏈,若有優異的產品策略及商業模式,勢必能在此產業競爭中脫穎而出。Before the launching of iPad and Tablet PC, e-ink based e-reader is the dominant reading device for e-books. However, e-reader has faced strong competitive threat since iPad entered into the market. Facing future uncertainty, whether e-reader could maintain dominance in the e-reader market or just a transitional product is a critical issue that is worth for further research.
The present study selects e-reader and tablet PC as targets for systematic analyses. In the first part, we analyze the industry structure and the technology development of the e-reader industry. In the meantime, the study also reviews the role that the digital content providers play in the e-reader industry. In addition to studying the e-reader industry, we apply the scenario analysis method to analyze the future development and structure of the e-reader industry under which potential contenders can formulate their strategies. Based on scenario analysis, we suggest the following strategic alternatives for e-reader companies.
For the e-reader brand companies, the study suggested these companies can invest and develop both e-paper based and LCD based e-reader or tablet pc to attract different targets of customers. However, the e-paper manufacturers are under severe impact and uncertainty due to the debut of iPad and tablet PC. The huge capital investment and technology development make these businesses have high exit costs. The study suggested that this industry for new technology development and investment should remain flexible and conservative until the situation is clearer.
As Taiwanese companies preempt substantial number of patents of e-reader technologies and have competitive industry supply chain, these companies shall maintain competitive edge in the e-reader industry if their strategies are right
