    Classification Model of HCC Patients and Healthy People Established from Mass Spectrometry Data

    通过对质谱数据处理和分析,建立肝细胞癌(HCC)病人与健康人的分类模型,可以用来判别样本是否来源于肝癌病人。运用表面加强激光解析电离-飞行时间质谱(SELDI-TOF-MS)获取肝癌病人和健康人血清蛋白指纹图谱数据,并运用偏最小二乘(PLS)变量筛选法建立分类模型,最终得到分类模型的交叉检验相关系数达0.96以上,判别准确率大大提高。同时对模型进行分析,找出对肝癌病人和健康人的差异有重要影响的因素或变量。这些变量为30个质荷比区间内特定蛋白的峰强度值,反映这些质荷比区间内蛋白量的增加或减少与肝癌的形成有密切关系,可作为重要的生物标志物进一步加以研究。并且采用所得模型的拟合值等一些信息做分类图,能较好地表达回归模型的分类效果。Classification models of hepatocellular carcinoma(HCC) patients and healthy people were built from mass spectrometry data,which could be used for the detection of HCC.Surface-enhanced laser desorption and ionization time-of-flight mass spectrometry(SELDI-TOF-MS) technique was applied to get the data of serum protein from HCC patients and healthy people.Then PLS variable selection method was used to deal with the data and to establish the classification model.The cross validation relativity coefficients of the model cames to over 0.96.Furthermore the important factors or variables that discriminated HCC patients and healthy people were found by analyzing the model.The 30 variables were several peak intensities of protein from some m/z sections,which could express the up-regulation or down-regulation of protein in the sections.As potential biomarkers,the proteins may be closely related to the formation of HCC,which can be deeply studied.The classification figures constructed by the fitting value of the model in the article are clear and intuitive,and can express the discrimination effect of the model well.福建省自然科学基金(批准号:2007J0290)资

    Construction of SSH library from haemocyte of variously colored abalone challenged with bacteria and differential expression analysis of macrophage expressed protein

    以雌性杂色鲍为对象,以大肠杆菌、副溶血弧菌、溶壁微球菌、表皮葡萄球菌、金黄色葡萄球菌的混悬液做为攻毒菌,利用抑制性差减杂交(SSH)技术构建细菌攻毒的杂色鲍血淋巴细胞SSHcDNA文库。随机挑取生长菌落110个克隆子,进行菌液PCR鉴定,计算文库重组率为98.18%,文库容量为1.37×106pfu。将重组子测序,经BLAST一致性搜索比对分析,有一重组片段含有穿孔素(Perforin)保守结构域,为巨噬细胞表达蛋白(MEP)类穿孔素部分cDNA序列,片段大小为1551bp,连续编码517个氨基酸残基,申请GenBank登录号为EU272049。经半定量PCR和荧光定量PCR差异显示分析,发现在细菌感染状态下MEP基因在血淋巴细胞中存在明显的上调表达现象。Abalones are considered to be the most precious delicacy from the sea, and become very important commercial seafood in aquaculture worldwide. Variously colored abalone (Haliotis diversicolor Reeve, 1846) has been widely cultured on the southeast coast for more than twenty years. However, abalone culture frequently suffers from bacterial infection and mass mortality of reared abalones causes serious economic losses. Unfortunately, knowledge of the defense mechanism in this animal is still lacking. In this study, using suppression subtractive hybridization (SSH) technology, a forward SSH li-brary was constructed from haemocytes of H. diversicolor, with the content of 1.37×106 pfu and the recombinant rate of98.18%. After the recombinant plasmids were sequenced, partial cDNA of macrophage expressed protein (MEP) was recognized based on BLAST searches in NCBI, with the size of 1 551 bp, and continuously encoding 517 amino acids. Semi-quantitative PCR and quantitative real-time PCR results showed that MEP cDNA was distinctly up-regulated in haemocytes of the bacterial-challenged group compared to the unchallenged group. The gene information obtained from this library will provide new insights into the immune mechanism of H. diversicolor and facilitate future study of target genes involved in the response to invading microorganisms.国家高技术研究发展计划项目(863计划)(编号:2007AA091406)资助~

    GaNAs/GaAs量子阱的激子局域化以及退局域化

    我們利用光熒光 (PL)以及時間分辨光譜 (TRPL)研究了用MBE生長在GaAs襯底上的GaNAs/GaAs量子阱的激子局域化以及退局域化。研究發現 ,在低溫下用連續光 (CW)激發 ,由于GaNAs中勢振蕩所產生的局域激子發光是所測量到光譜的主要發光來源。然而在脈沖激發下 ,情況完全不同。在高載流子密度激發或者高溫下GaNAs/GaAs量子阱中例外 ,一個高能端的PL峰成為了主要的發光來源。通過研究 ,我們將這個新的發光峰指認為量子阱中非局域激子復合的PL峰。這個發光峰在溫度和激發強度的變化過程中與局域激子相互競爭。我們相信這一過程也是許多文獻所報道的在InGaN和AlGaN等氮化物中經常觀測到的發光峰位隨溫度“S”形變化的主要根

    物理保定是野生动物饲养管理的重要工作内容之一,在疾病诊治过程中起了重要作用。为了解决中型非人灵长类动物在临床检查和治疗等操作时的物理保定问题,本研究根据中型非人灵长类动物的生物学特点和兽医临床需要,研制了一种压缩保定治疗笼。该笼具为长方体结构,通过滑轮、推拉杆等装置控制后侧面移动,以改变笼体空间大小,实现压缩功能;并配有操作窗口、可拆卸喂食(水)盘、折叠式输液架等结构。临床应用结果表明,该笼具对长臂猿、金丝猴、猕猴等多种中型非人灵长类动物都能实现压缩保定,对提高临床检查效率和治疗效果起到了积极作用

    应用响应面分析方法(Response Surface Method,RSM),对转人源胸腺肽α1基因聚球藻7942的培养基及培养条件进行优化,优化后摇瓶培养转化藻的比增殖速率(μ值)可达0.500d-1,10 L全自动光生物反应器中培养转化藻μ值可达0.979 d-1,分别比正交实验提高了33%和161.1%,建立高密度培养的数字模型预测实验结果,预测值与实验值之间的相对误差在±5.0%以内,为响应面方法在微藻细胞培养中的广泛应用奠定了基础。The response surface method(RSM)was used to optimize the medium and culture conditions for transgenic Synechococcus sp.PCC7942 with human thymosin α1 gene.The results were as follows: NaHCO3 36 mg/L,KNO3 1.3 g/L,initial pH 10.0,light intensity 1800 lx.Under these conditions,the specific growth rate(μ) of the transgenic strain could reach 0.500 d-1 through shake-flask culture,when cultivated in 10L autophotobioreactor,the μcould increase to 0.979 d-1,with the maximal yield of 0.299 g/ L·d.There was no reduction of the bioactivity of UB-Tα1 in these culture processes
