    研究如何快速、精确地匹配三基色rgb的比例得到任意颜色的光具有重要意义.提出一种结构简单易于实现的rgb-lEdS配色模型,通过振幅调制电流来改变三基色lEd辐射功率,从而调节各基色在混合色中的亮度比例,使得rgb-lEdS能够匹配到任意颜色.同时给出使用该模型进行配色的精确度评价指标.在此模型基础上提出一种基于二分法的rgb配色算法,使得混合颜色的色品坐标能够快速、精确地匹配到指定范围.仿真实验证明了算法的正确性和有效性.该算法收敛速度快,配色精确度高,具有很高的实用价值.The way to match the ratio of RGB trichromatic lights to get arbitrary color of light quickly and accurately is of great significance.An easily realizable RGB-LEDs color matching model with simple structure was proposed.An arbitrary color can be matched by changing trichromatic LED radiation power in the way of current amplitude modulation,which in turn adjusts the proportion of each color′s luminous intensity.At the same time,an accuracy evaluation indicator for color matching model was given.Based on the model,a dichotomy algorithm of RGB color matching is employed so that the chromaticity coordinates of mixed color can be quickly matched to a specified scope.The simulation results prove the correctness and validity of the algorithm.This algorithm has a high value in practical application for its rapid convergence and high precision color matching.863半导体照明工程重大专项(2006AA03A175);福建省重大专项(2006H0092);福建省自然科学基金(2008J0030

    目的克隆小鼠的SIrT5基因,及制备SIrT5多克隆抗体。方法提取小鼠肝细胞总rnA进行rT-PCr,PCr法扩增SIrT5基因,将此基因克隆至PET-28A载体,转化E.COlI bl21(dE3),经异丙-β-d-硫代吡喃半乳糖苷(IPTg)诱导表达后,利用nI 2+-nTA亲和层析柱纯化;纯化的目的蛋白免疫bAl/C小鼠后,收获血清并鉴定。结果成功构建了SIrT5原核表达载体,并能在宿主菌E.COlI bl21(dE3)中高效表达;利用纯化的蛋白制备了理想的多克隆抗体。结论利用分子克隆技术,获得了高纯度的SIrT5蛋白并制备了多克隆抗体,为进一步研究奠定了基础。Objective To construct prokaryotic expression vector of SIRT5and prepare its polyclonal antibody.Methods Total RNA isolated from mice liver cell was RT-PCR.Sirt5was amplified by RT-PCR and cloned into pET-28aplasmid,and the recombinant plasmid was transformed into E.coli BL21(DE3).After induction by IPTG,the fusion protein of SIRT5was produced,the protein was purified by Ni 2+-NTA afinity chromatography.Bal/c mice were immuned with the purified protein of SIRT5,and mice serum was collected for identification.Results SIRT5was successfully constructed and highly expressed in E.coli BL21(DE3).The purified protein was used to prepare polyclonal antibody.Conclusion Using moleccular cloning techonlogy,purified SIRT5protein can be obtained and prepared its polyclonal antibody,which may provide an foundation for further investigation

    目的构建耻垢分枝杆菌末端结合蛋白ku原核表达载体,并进行表达纯化及酶学测定。方法以耻垢分枝杆菌STr.MC2 155基因组dnA为模板,PCr法扩增ku基因,构建PQE-30-ku重组质粒,在表达宿主菌E.COlI M15中诱导表达蛋白,nI2+-nTA亲和层析柱纯化,最后通过凝胶迁移电泳实验测定ku蛋白与5′粘末端、3′粘末端和平末端3种不同末端dnA结合情况。结果成功构建了ku原核表达载体PQE-30-ku,并能在宿主菌E.COlI M15中表达,纯化后具有结合不同末端dnA的酶活性。结论 ku基因克隆到原核宿主菌中能进行表达、纯化,纯化后的蛋白具有结合不同末端dnA的生物学活性,为分枝杆菌非同源末端连接的研究奠定了基础。Objective To construct the prokaryotic expression vector of Ku binding protein of mycobacterium smegmatis terminal,and expressed determination of purification and characterization.Methods Ku gene was amplified from Mycobacterium smegmatis str.MC2 155 genomic DNA by PCR and then cloned into expression vector pQE-30.Finally the recombinant plasmid was expressed in E.coli M15.After induction by IPTG,the fusion protein of Ku was produced,the protein was purified by Ni2+-NTA afinity chromatography,detected it by EMSA.Results Ku was successfully constructed and expressed in E.coli M15.It could be purified and bind different terminal DNA(5'overhangs,3'overhangs and blunts).Conclusion Ku gene could be cloned into the host bacterium and successfully expressed and purified,the purified protein was biologically active,it could bind different terminal DNA,which might be provide an foundation for further research

    采用电阻应变测量技术研究了带有初始随圆度的弯管在纯内压作用下的应力分布 .实验结果表明 ,弯头横截面上的环向应力和轴向应力分布均呈相似的“W”型 ,但各点环向应力的绝对值均大于轴向应力 .最大应力在外弧线的环向 ,比直管高出 6 2 .7% ,最小应力在中线弧处 ,为负值 .这种应力分布情况也与弯管受平面弯矩时完全不同 .The stress distribution of a curved pipe with an initial elliptic cross section under internal pressure was studied by using electric resistance strain method. The experimental results showed that both the circumferential and axial stresses around the cross section varied in a similar "W" shape, but the former was larger than the latter in absolute valume. The circumferential stress of the extrados is about 62.7% higher than the circumferential stress of straight pipe with the same section size and pressure, while the stress of the middle curve is less then zero. It is also indicated that the stress curves under internal pressure are different from those under in plane bending moments both in shape and in the location of the maximal stress
