    Expression of a Shrimp Kunitz-type Protease Inhibitor in Pichia pastoris and Activity Analysis

    SKPI(shrimp Kunitz-type protease inhibitor)是日本囊对虾(Marsupenaeus japonicus)体内的一个小分子多肽,含有一个Kunitz型结构域,属于丝氨酸蛋白酶抑制剂。目前已知丝氨酸蛋白酶抑制剂在节肢动物免疫系统中起着非常重要的作用,为了了解SKPI在对虾天然免疫系统中的作用,首先对其进行了重组表达。从日本囊对虾肝胰腺中扩增skpi的cDNA片段,插入改造后的pPIC9K酵母表达载体,获得的重组质粒转化至毕赤酵母GS115进行表达。由于改造的pPIC9K载体加入了6-His标签,因此利用Ni Sepharose High Performance对SKPI进行了高效纯化。初步的活性研究表明,重组表达的SKPI能特异性地抑制胰蛋白酶的水解活性。SKPI(shrimp Kunitz-type protease inhibitor) from Marsupenaeus japonicus is a member of serine protease inhibitors which play an important role in the arthropod immunity.To fully understand its function in the innate immunity of shrimp,the skpi gene was cloned into a modified pPIC9K vector with a 6-His tag and expressed by Pichia pastoris GS115.The secretory SKPI was purified from the medium with high purity by using Ni Sepharose High Performance.This results also indicated that the purified SKPI could inhibit the activity of trypsin specifically.国家高技术研究与发展计划项目(No.2006AA100311);; 福建处科技厅资助重点项目(No.2006N0039)~

    Litter and Caloric Value of Casuarina equisetifolia Plantation in Coastal Sandy Area

    采用定位研究的方法,在福建惠安赤湖防护林场定期收集木麻黄林分的凋落物,开展凋落物的归还量和热值的研究.结果表明,木麻黄林凋落物年归还量为14.17thm-2a-1,小枝占总归还量的72.19%,其他各组分占27.81%.凋落物灰分含量小枝4.39%~5.65%,枝条3.52%~5.65%,球果2.63%~4.23%;凋落物干重热值为小枝20.51~21.68kJ/g,枝条19.63~20.21kJ/g,球果18.27~20.63kJ/g;凋落物去灰分热值为小枝21.48~22.69kJ/g,枝条21.48~22.69kJ/g,球果18.91~21.49kJ/g.图1表8参26The litter of Casuarina equisitiforia plantation in Hui′an County, Fujian was regularly collected, and its return amount and caloric value were studied. Result showed that the return amount of litter was 14.17 t hm-2 a-1, with leaf accounting for 72.19% and the others for 27.81%. The litter ash contents of leaf, branch and cone ranged between 4.39%~5.65%, 3.52%~5.65% and 2.63%~4.23%, respectively. The litter gross caloric values of leaf, branch and cone were 20.51~21.68 kJ/g, 19.63~20.21 kJ/g and 18.27~20.63 kJ/g, and the ash free caloric values of them were 21.48~22.69 kJ/g, 21.48~22.69 kJ/g and 18.91~21.49 kJ/g, respectively.     Anticorrosion Properties of Modified Nano-TiO_2 Films Prepared by Sol-Gel Method

    应用溶胶-凝胶法和浸渍提拉技术在316l不锈钢表面分别制备TIO2纳米膜和 b-fE-CE改性的TIO2纳米膜. 采用场发射扫描电子显微镜(fE-SEM)、原子力显微镜(AfM)、拉曼光谱法和能量分散谱(EdS)对薄膜进行表征,通过电化学阻抗谱(EIS)和动电位阳极极化曲线的测试考察薄膜的耐蚀性及对不锈钢的保护性能. 结果表明:两种纳米薄膜均含锐钛矿型的TIO2纳米颗粒,纯TIO2纳米膜与改性后的纳米膜中颗粒直径分别约为15和10 nM. TIO2/316l不锈钢和 b-fE-CE-TIO2/316l不锈钢膜电极浸泡在0.5 MO.ll-1 nACl溶液后,后者的电化学反应电阻较大,动电位阳极极化曲线的稳定钝化区较宽,击穿电位更高,说明改性的纳米膜的耐蚀性及其保护性能更好.TiO2 and B-Fe-Ce-modified TiO2 films were synthesized on the surfaces of 316L stainless steel(316L SS) substrates using a sol-gel and dip-coating method.The properties of the films were characterized by field emission scanning electron microscopy,atomic force microscopy,Raman spectroscopy and energy dispersive spectrometry.The corrosion resistance of the films and their ability to protect stainless steel were investigated by electrochemical impedance spectroscopy and potentiodynamic polarization curves.Both TiO2 and B-Fe-Ce-modified TiO2 films were composed of anatase nanoparticles about 15 and 10 nm in diameter,respectively.Impedance spectra of the stainless steel substrates coated TiO2 films contained semicircles for capacitive reactance in 0.5 mol.L-1 NaCl solution,but the charge transfer resistance of the B-Fe-Ce-TiO2/316L SS electrode was higher than that of the TiO2/316L SS electrode.The potentiodynamic anodic polarization curve of the B-Fe-Ce-TiO2/316L SS electrode showed a larger stable passive region and a higher breakdown potential than the TiO2/316L SS electrode,indicating that the modified film had better corrosion resistance and protective properties for 316L SS.国家高技术研究发展计划项目(863)(2009AA03Z327);国家自然科学基金(21073151和21173177)资助---

    厦门地区无偿献血者HEV隐性感染情况及基因型分析

    目的了解厦门地区无偿献血者中戊型肝炎的感染情况。方法对2005年3月—2007年4月厦门地区20389人(次)无偿献血者做整群抽样,捕获法ElISA检测IgM抗-HEV、并对IgM抗-HEV阳性样本做HEVrT-PCr检测,经测序后分析其基因型及序列同源性。结果20389份无偿献血者血样本中,IgM抗-HEV阳性率为0.91%(186/20389);186例IgM抗-HEV阳性中有4例经rT-PCr检测为HEV阳性。献血者中病毒血症总阳性率为2/10000(4/20389),4例病毒血症中Ⅰ型与Ⅳ型感染各2例。结论厦门地区无偿献血者中,存在HEV隐性感染者。厦门市卫生局医学科研项目资助课题(编号:WSK0516

    Differential cognitive responses to guqin music and piano music in Chinese subjects: an event-related potential study

    目的研究古琴(一种古老的中国乐器)和钢琴音乐对认知的影响。方法记录和分析了中国被试在两种音乐背景(古琴音乐,钢琴音乐)下完成听觉oddball任务的行为和事件相关电位(event-related potential,ERP)数据。结果中国被试在本土文化的音乐环境(古琴音乐)下,前额区诱导出更大的P300,这一结果和已有的相关研究是相符的。同时,不同音乐背景对ERP产生的影响在N1和LPC(包括P300和P500)上也表现出差别:中国被试在古琴音乐背景下比钢琴音乐背景下表现出更多的右前侧颞叶的参与。结论因为古琴音乐的五声调式和汉语发音的音调具有对应关系,因此我们推断在古琴音乐下所表现出的这种特性与被试的汉语环境有关。Objective To compare the cognitive effects of guqin (the oldest Chinese instrument) music and piano music. Methods Behavioral and event-related potential (ERP) data in a standard two-stimulus auditory oddball task were recorded and analyzed. Results This study replicated the previous results of culture-familiar music effect on Chinese subjects: the greater P300 amplitude in frontal areas in a culture-familiar music environment. At the same time, the difference between guqin music and piano music was observed in N1 and later positive complex (LPC: including P300 and P500): a relatively higher participation of right anterior-temporal areas in Chinese subjects. Conclusion The results suggest that the special features of ERP responses to guqin music are the outcome of Chinese tonal language environments given the similarity between Guqin’s tones and Mandarin lexical tones

    无偿献血者中隐匿性乙型肝炎病毒感染及表面抗原突变分析

    采用多种免疫学检测和核酸检测相结合的方法调查了我国南方某城市无偿献血者中隐匿性乙型肝炎病毒(HBV)感染的存在情况。结果在9023例乙肝表面抗原(HBsAg)阴性的无偿献血者中,共发现17例HBV DNA阳性,隐匿性HBV感染者的发生率为0.19%(95%CI:0.11~0.30%)。序列分析显示其中6例在HBsAg"a"表位(aa124~aa147)存在不同程度氨基酸突变,突变发生率为42.9%(6/14,有3例未扩增出"a"表位片段序列),G145R突变是该地区隐匿性HBV携带者中发生频率最高的突变(4/6,66.7%)。隐匿性HBV感染者中基因型C的比例(10/17)显著高于HBsAg阳性的HBV感染者(0/15,P<0.01)

    Experimental Investigation on Thermocapillary Drop Migration at Large Marangoni Number in Reduced Gravity

    Results from a space experiment on thermocapillary drop migration conducted on board the Chinese spacecraft ShenZhou-4 are presented in this paper. In the experiment, isolated drops of Fluorinert liquid moved in a matrix liquid of 5cst silicone oil at values of the Marangoni numbers (Ma) ranging up to 5500 and the interferometry images showed the temperature distribution inside the test cell. The drop migration velocity was measured. The experimental results show that the scaled drop migration velocity V/VYGB obviously decreases with Ma increasing the values up to 5500. The space experimental results are also compared with those from our early experiments, other space experiments, and some theoretical predictions

    通信作者:史建陆Tel:(0576)85199572 E-mail: zmfjmu@ gmail.com[中文文摘]目的研究纳米银羟基磷灰石的细胞毒性。方法采用能快速、准确、敏感地评价细胞增殖率和细胞毒性的MTT比色法,以纳米羟基磷灰石为对照组,纳米银羟基磷灰石为实验组,在材料浸提液中,分别检测人牙周膜成纤维样细胞的相对增殖率。另观察细胞在扫描电镜中的形态,从而分析比较涂层表面对细胞的毒性。结果与纳米羟基磷灰石相比,纳米银羟基磷灰石涂层中的纳米银粒子直径在200~400nm时体外细胞培养显示表现出较高的细胞增殖率,细胞毒性趋于0级,显示其具有良好的生物活性。结论纳米银羟基磷灰石涂层无细胞毒性。[英文文摘]Objective To study the cytotoxicity of a Nano-Ag/HAp composite coating on titanium surface.Methods A Nano-Ag/HAp composite coating was deposited on the surface of pure titanium by electrochemical deposition.In vitro celluar responses of the PDLF to the coating were also evaluated by MTT assay.Conventional HAp coatings and commercially pure titanium were taken as control.Results The Nano-Ag/Hap bioceramic coating has good crystallization and homogeneity on nano-scale surface morphology.The PDLF culture exhibits satisfactory bioactivity.Conclusions The Nano-Ag /HAp bioceramic coating on titanium surface has promising prospects in the future clinical application

    中国物理海洋学研究70年:发展历程、学术成就概览

    本文概略评述新中国成立70年来物理海洋学各分支研究领域的发展历程和若干学术成就。中国物理海洋学研究起步于海浪、潮汐、近海环流与水团,以及以风暴潮为主的海洋气象灾害的研究。随着国力的增强,研究领域不断拓展,涌现了大量具有广泛影响力的研究成果,其中包括:提出了被国际广泛采用的"普遍风浪谱"和"涌浪谱",发展了第三代海浪数值模式;提出了"准调和分析方法"和"潮汐潮流永久预报"等潮汐潮流的分析和预报方法;发现并命名了"棉兰老潜流",揭示了东海黑潮的多核结构及其多尺度变异机理等,系统描述了太平洋西边界流系;提出了印度尼西亚贯穿流的南海分支(或称南海贯穿流);不断完善了中国近海陆架环流系统,在南海环流、黑潮及其分支、台湾暖流、闽浙沿岸流、黄海冷水团环流、黄海暖流、渤海环流,以及陆架波方面均取得了深刻的认识;从大气桥和海洋桥两个方面对太平洋–印度洋–大西洋洋际相互作用进行了系统的总结;发展了浅海水团的研究方法,基本摸清了中国近海水团的分布和消长特征与机制,在大洋和极地水团分布及运动研究方面也做出了重要贡献;阐明了南海中尺度涡的宏观特征和生成机制,揭示了中尺度涡的三维结构,定量评估了其全球物质与能量输运能力;基本摸清了中国近海海洋锋的空间分布和季节变化特征,提出了地形、正压不稳定和斜压不稳定等锋面动力学机制;构建了"南海内波潜标观测网",实现了对内波生成–演变–消亡全过程机理的系统认识;发展了湍流的剪切不稳定理论,提出了海流"边缘不稳定"的概念,开发了海洋湍流模式,提出了湍流混合参数化的新方法等;在海洋内部混合机制和能量来源方面取得了新的认识,并阐述了混合对海洋深层环流、营养物质输运等过程的影响;研发了全球浪–潮–流耦合模式,推出一系列海洋与气候模式;发展了可同化主要海洋观测数据的海洋数据同化系统和用于ENSO预报的耦合同化系统;建立了达到国际水准的非地转(水槽/水池)和地转(旋转平台)物理模型实验平台;发展了ENSO预报的误差分析方法,建立了海洋和气候系统年代际变化的理论体系,揭示了中深层海洋对全球气候变化的响应;初步建成了中国近海海洋观测网;持续开展南北极调查研究;建立了台风、风暴潮、巨浪和海啸的业务化预报系统,为中国气象减灾提供保障;突破了国外的海洋技术封锁,研发了万米水深的深水水听器和海洋光学特性系列测量仪器;建立了溢油、危险化学品漂移扩散等预测模型,为伴随海洋资源开发所带来的风险事故的应急处理和预警预报提供科学支撑。文中引用的大量学术成果文献(每位第一作者优选不超过3篇)显示,经过70年的发展,中国物理海洋学研究培养了一支实力雄厚的科研队伍,这是最宝贵的成果。这支队伍必将成为中国物理海洋学研究攀登新高峰的主力军

    Damage to mangroves from extreme cold in early 2008 in southern China

    通讯作者Author for correspondence (E-mail: [email protected])2008年初,我国南方19个省经历了50年一遇的持续低温雨雪冰冻天气。极端气候对华南沿海各省的红树林区造成不同程度的危害。2008年3月,在我国南方各省红树林区的10个代表性地点,对这次寒害造成的红树植物伤害程度进行了系统的调查。结果表明:冬季低温对红树林的影响极为显著,特别是在低纬度的海南、广西和广东湛江,由于极端低温正值夜间退潮,对红树林的影响更为显著;在纬度较高的福建,本地红树种类秋茄(Kandelia obovata)、桐花树(Aegiceras corniculatum)和白骨壤(Avicennia marina)及引种的木榄(Bruguiera gymnorrhiza)等,由于长期适应于冬季较低的气温或在种植前经过抗寒锻炼,具有较强的抗寒能力。各地主要红树植物中,广布种秋茄、桐花树和白骨壤最为耐寒,其耐寒性均大于红树科的木榄、海莲(Bruguiera sexangula)和红海榄(Rhizophora stylosa)。海桑(Sonneratia caseolaris)对温度的敏感性最强,抗寒能力最低,因此,即使在其原产地海南也受到较为严重的寒害,在纬度更高的引种地出现大面积受害甚至全部死亡,而从孟加拉国引种的无瓣海桑(Sonneratia apetala)却显示出一定的抗寒能力。同一地点的红树植物幼苗的抗寒能力低于大树。此次寒害也造成了苗圃场的种苗大量死亡,成熟的植株提前落花落果,这势必会影响后继一两年内红树林的自然更新和人工造林。因此,在未来红树林造林或人工引种中,一定要考虑到红树植物的抗寒能力。国家自然科学基金(30671646和30700092);厦门大学“闽江学者”启动基
