    Experimental study on effect of Changrun Formula in regulating expression of AQP3 and AQP9 in colon mucosa of functional constipation rats

    目的:观察肠润方对功能性便秘模型大鼠结肠黏膜AQP3、AQP9表达的影响,探究其调控AQP3、AQP9表达的分子机制。方法:60只SD大鼠随机分为空白组、模型组、阳性对照组(麻仁丸组)、肠润方组、肠润方联合P38MAPK抑制剂SB203580组。用复方地芬诺酯制造大鼠便秘模型,采用免疫组织化学及实时荧光定量PCR(q RT-PCR)检测大鼠便秘模型近端及远端结肠黏膜AQP3、AQP9的表达;Western Blot检测信号传导通路相关分子P38MAPK及P-P38MAPK的表达。结果:造模成功后,模型组近端结肠黏膜AQP3表达较空白组明显上升,而远端结肠黏膜AQP9表达较空白组明显下降(t值分别为3.148和7.069,P值均0.05)。肠润方联合P38MAPK抑制剂SB203580治疗后,近端结肠黏膜AQP3 m RNA水平显著上升(t=5.922,P0.05). The level of AQP3 m RNA in proximal colonic mucosa was significantly increased(t=5.922, P<0.01), and AQP9 m RNA was significantly decreased in distal colonic mucosa(t=4.038, P<0.01); The relative expression of P-P38/P38 protein was significantly decreased(t=19.419, P<0.01). Conclusion: The therapeutic action ofChangrun Formula on functional constipation rat models might through inhibiting the expression of AQP3 in proximal colon, and accelerating on expression of AQP9 in distal colon, and the regulating mechanism might relate with the inhibition on P38 MAPK and AKT phosphorylation.福建省自然科学基金项目(No.2012D043)~

    [中文文摘]考察了PVA Zr (Ⅳ )膜催化乙酸和丁醇酯化反应的宏观反应动力学 ,认为膜催化酯化反应为扩散 反应联合控制 ,膜溶胀实验表明反应液各组分浓度不同于膜中各组分平均浓度 .假设膜催化反应分为 :膜溶胀前及溶胀后两个阶段 ,分别求出了此两个阶段反应动力学常数 .比较了PVA Zr (Ⅳ )催化膜和无催化活性层的PVA膜渗透性能.[英文文摘]The solubility of catalytic membrane of PVA-Zr(Ⅳ) was investigated and that the catalytic layer was found to swell in nearly two hours. The composition of the bulk liquid mixture was varied during the swelling and was different from that of the catalytic layer when the swelling equilibrium state was reached.Furthermore, the reaction rate constants of reaction kinetics equations were obtained for the esterification of acetic acid with n-butanol catalyzed by PVA-Zr(Ⅳ)catalytic membrane. The esterification was taken as a model reaction to study the permeation flux for the catalytic membrane was less than that for the non-catalytic layer membrane in the first reaction stage during swelling, and such comparision was reversed around four hours later when swelling equlibrium was rdached.国家自然基金资助项目 (No 50243014

    Well Width Dependence of Band Gap Renormalization of Single ZnO/MgZnO Quantum Well

    通过室温下的时间积分光致发光(Pl)谱,研究了阱宽lW渐变的znO/Mg0.1zn0.9O单量子阱在高激发强度下的能带重正化与阱宽的关系。实验中光生载流子浓度为n=1.6x1014CM-2,在lW从2.3 nM渐变到4.3 nM,Pl谱峰位的红移量从5.9 MEV变化到97.1 MEV。红移量随阱宽增大而增加,但增加率却逐渐减少。当lW>2αb(αb,znO体材料激子玻尔半径,约为2 nM)时,红移量逐渐呈现出饱和的趋势(100 MEV)。研究发现峰位的红移是由于多体效应所导致的能隙收缩以及在高的激发强度下带内填充效应的这两种机理相互竞争的结果。Band gap renormalization of ZnO/Mg0.1Zn0.9O single quantum well(QW) with gradual well width(Lw) is studied by room-temperature time integrated photoluminescence(PL) spectra at high excitation power density.The photo-generated carrier density is n=1.6×1014 cm-2 and the magnitude of red shift of PL spectrum peak increases from 5.9 meV to 97.1 meV with Lw changing from 2.3 nm to 4.3 nm.With Lw increaseing,the red shift increases but the increase rate gradually decreases.When Lw>2αB(αB,the exciton Bohr radius of ZnO bulk,is about 2 nm),the red shift starts to be gradually saturated.It is found that the red shift is the competition result of energy gap contraction due to many body effect and intraband filling effect at high excitation power density.The result is useful for designing and application of ZnO QW-based optoelectronic devices.国家973计划(2004CB719804);国家自然科学基金(60876007);广州市科技局推荐项目(2005Z3-C7451)资助课

    Treatment of Hepatocellular Carcinoma in Mice with Locally Administered Epirubicin-loaded Poly(D,L)-lactic Acid Microspheres

    [中文文摘]目的研究表柔比星-聚乳酸缓释微球(EPI-PLA-MS)局部给药治疗肝癌的效果。方法复乳-溶剂挥发法制备EPI-PLA-MS。将40只昆明小鼠随机分为5组,每组8只,分别腹腔注射不同剂量的游离表柔比星(FEPI),计算最大耐受量(MTD)。H22皮下实体瘤肝癌荷瘤小鼠3组,每组5只,分别用生理盐水(normalsaline,NS)、空白微球和含药微球(含EPI9mg/kg)瘤内注射给药,2周后取瘤称重。H22腹水型肝癌荷瘤小鼠3组,每组5只,分别用NS、空白微球和含药微球(含EPI9mg/kg)腹腔注射给药,计算动物生命延长率。结果FEPI腹腔注射的MTD为9mg/kg。EPI-PLA-MS瘤内给药后含药微球组和空白微球组的抑瘤率分别为40.35%和36.09%。腹腔给药后能显著延长荷瘤鼠的生存时间,含药微球组和空白微球组生命延长率分别为153.49%和142.22%。结论EPI-PLA-MS是一种有效低毒的药物新剂型,在局部治疗肝癌方面具有良好的临床应用前景。[英文文摘]Objective To study the effectiveness of treating hepatocellular carcinoma(HCC)in mice with locally administered epirubicin-loaded poly(D,L)-lactic acid microspheres(EPI-PLA-MS).Methods EPI-PLA-MS was prepared with double emulsion solvent evaporation technique.Five groups of mice(n=8 in each group)were intraperitoneally injected with five different doses of free epirubicin(FEPI),and the maximum tolerated dose(MTD)was calculated.Then 15 mice with transplanted subcutaneous H 22 HCC were divided into three group s ( n = 5) , which were respectively intratumorally injected with normal saline (NS) , blank micro-spheres, and EPI-PLA-MS (with 9mg/kg of EPI) 1 After two weeks the tumorswere excised and weighed1 Another 15 mice with transp lanted H22 ascites HCC were divided into three groups (n=5) , which were intraperitonealy injected with the same drugs, and the increased life span were registered exactly1 Resutls TheMTD of intraperito neally injected FEPIwas 9mg/kg1 The tumour inhibiting rateswas 40135% and 36109% when EPI-PLA-MS were administered by intratumoral injection to the mice with subcutaneous H22 HCC1 It significantly p rolonged the survival time of mice with H22 ascites HCC and the increased life span by 153149% and 142122% when EPI-PLA-MS were intraperitoneally administered1 Conclusion EPI-PLA-MS is a new sustained release preparation with high-ef-ficacy and low-toxicity in treating HCC and has shown promising prospects when administerd locally

The crosslinked polymer matrix acted as structure-directing agent in a pyrolyzing process, whereas the functional precursor was converted into the skeleton of the micropatterns. The inorganic micropatterns could be further catalytically functionalized to grow CNT and ZnO nanorod arrays by simply changing different functional precursors.     Identification and preliminary analysis of a novel full-length cDNA encoding retinoid X receptor 2 from Schistosoma japonicum

    通信作者 E-mail: [email protected][中文文摘]目的克隆编码日本血吸虫视黄酸X受体2(SjRXR2)蛋白的全长cDNA,并对其进行初步研究。方法利用cDNA末端快速扩增技术(RACE)获得SjRXR2蛋白全长编码cDNA。利用生物信息学技术,对基因结构进行初步分析。利用实时荧光定量(Real time)PCR技术对该基因在日本血吸虫不同时期虫体中的转录情况进行分析。应用在线抗体表位预测软件获得SjRXR2配体结合区抗原性较强的一个多肽序列,合成该多肽片段,并免疫小鼠制备抗血清。利用Western blot技术分析该蛋白在日本血吸虫中的表达。结果采用RACE技术成功获得了SjRXR2蛋白全长编码cDNA,总长度为5 960bp,其完整开放阅读框为4 308 bp,编码1 435个氨基酸,预测分子量为159 kDa。生物信息学分析表明该基因编码的蛋白质序列具有核受体家族2的典型结构域特征,且与曼氏血吸虫RXR2有较高的相似性。Real time PCR分析表明,该基因在21、42 d龄日本血吸虫虫体内有较高的转录水平。Western blot分析表明,小鼠SjRXR2多肽免疫血清可特异性识别日本血吸虫虫体150 kDa蛋白。结论成功获得了编码SjRXR2蛋白的全长 cDNA,并制备了针对该蛋白的特异性多克隆抗体,为进一步研究该蛋白的功能奠定了基础。 .国家自然科学基金(31172315);公益性行业(农业)科研专项(20090303036
