    Allelopathic effects of exotic mangrove species Laguncularia racemosa on leaf ultrastructure of Bruguiera gymnorhiza seedlings

    探讨外来速生红树植物拉关木(Laguncularia; racemosa)扩张过程中潜在的化感作用,为其入侵风险评估提供参考。通过室内栽培试验测定拉关木根、枝、叶、果的水浸液不同质量浓度(0.1; g·mL~(-1)、0.5 g·mL~(-1))对我国乡土红树植物木榄(Bruguiera; gymnorhiza)幼苗叶片超微结构的影响。结果表明,在拉关木根、叶、果的水浸液低浓度(0.1; g·mL~(-1))处理下,木榄幼苗的叶肉细胞内的叶绿体紧密排列在细胞壁周围,线粒体结构保持完好;而在拉关木枝的水浸液低浓度(0.1; g·mL~(-1))处理下,木榄幼苗叶肉细胞内的液泡轻微皱缩,出现轻微的质壁分离现象。拉关木枝、果水浸液高浓度(0.5; g·mL~(-1))处理下,木榄幼苗叶片叶肉细胞的叶绿体内的淀粉粒变多,变大,线粒体内部结构出现降解;拉关木根的水浸液高浓度(0.5; g·mL~(-1))处理下,部分叶绿体结构受到破坏,但线粒体结构基本保持较好;而在拉关木叶的水浸液高浓度(0.5; g·mL~(-1))处理下,木榄叶片叶肉细胞的超微结构基本保持完好,表明拉关木叶水浸液对木榄幼苗的化感抑制作用弱。本研究中拉关木各器官高浓度(0; .5 g·mL~(-1))水浸液对木榄幼苗叶片超微结构的化感作用强弱顺序为:枝>果>根>叶。In order to provide a reference for the invasion risk assessment of; fast-growing alien mangrove species of Laguncularia racemosa,the aqueous; extracts of different concentrations(0.1 mg·L~(-1),0.5 mg·L~(-1)) of; roots,stems,leaves and fruits of L. racemosa were used to determine the; allelopathic effects on the leaf ultrastructure of seedlings of; Bruguiera gymnorhiza native species in China by indoor cultivation; experiment. The results of this study show that when subjected to the; concentration of 0.1 mg·L~(-1) aqueous extracts from L. racemosa roots; and fruits,the number of chloroplasts in the mesophyll cells of B.; gymnorhiza seedlings increased and mitochondria were delimited by a; double mitochondria membrane,as well as showed clear cristae inside an; electrondense stroma;when subjected to the concentration of 0.1; mg·L~(-1) aqueous extracts of L. racemosa stems,the vacuoles of the; mesophyll cells in the mesophyll cells of B. gymnorhiza were slightly; shrunk and a slight wall separation was observed. Whereas subjected to; the concentration of 0.5 mg·L~(-1) aqueous extracts from L. racemosa; stems and fruits,the size and number of starch grains of chloroplast; were increased,and the mitochondria were deformed and seriously; degraded. When subjected to the concentration of 0.5 mg·L~(-1) aqueous; extracts from L. racemosa roots,some parts of chloroplast were; damaged,but the mitochondria structure basically remained intact;whereas; subjected to the concentration of 0.5 mg·L~(-1) aqueous extracts from L.; racemosa leaves,both the chloroplast and mitochondria in mesophyll cells; of B. gymnorhiza seedlings basically remained intact,which showed that; the inhibitory effect of the aqueous extract of L. racemosa leaves was; weak. When subjected to the concentration of 0.5 mg·L~(-1) aqueous; extracts from different organs of L. racemosa,the allelopathy intensity; order on the ultrastructure of leaves of B. gymnorrhiza seedlings was:; stem>fruit>root>leaves.国家自然科学基

    Tis11是哺乳动物基因TTP在果蝇中的同源物,其基因位于果蝇X染色体11B9区.它含有两个保守的顺式Cys-X8-Cys-X5-Cys-X3-His(CCCH)锌指结构.它与mRNA的稳定性有关.为了进一步从动物水平研究Tis11的生物学功能,作者利用P因子不精确剪切的特性来获得Tis11基因敲除果蝇.以P因子插入果蝇19949为起始材料,经过一系列杂交后,挑取了700株P因子被切除的果蝇.通过PCR筛选和鉴定,从这700株果蝇中获得一株果蝇279,它的Tis11基因的第2个外显子中包括翻译起始位点的区域被删除

    Experimental Study of the Abalone Shell on Cataract induced by Oxidative Stress

    目的研究石决明提取物对体外培养的晶状体氧化应激性白内障形成的作用及机制。方法离体培养小鼠晶状体,应用不同浓度的石决明提取物预孵育晶状体24h后,加入1mm过氧化氢,继续培养3小时后恢复正常培养,72小时后观察小鼠晶状体混浊程度,测定晶状体组织培养液中的乳酸脱氢酶(LDH)含量,晶状体组织中还原型谷胱甘肽(GSH)含量及超氧化物歧化酶(SOD)活力。结果石决明提取物在1~2 mg/ml浓度范围内减轻氧化应激造成的晶状体混浊,减少晶状体LDH的释放,提高组织内GSH含量和SOD活力。结论石决明提取物可减轻氧化应激白内障的形成,其作用主要与石决明提取物提高内源性抗氧化系统有关。Objective To study the effect of the abalone shell extract on oxidative stress induced cataract formation and its mechanism in cultured mouse lens in vitro. Methods The cultured mouse lens were pretreated with the abalone shell extract in different concentrations for24 hours,and then 1mm hydrogen peroxide was added and continued incubating for 3 hours,and they were changed to normal culture media.After 72 hours,the opacity of each lens was observed under an anatomical microscope,the content of lactate dehydrogenase( LDH)leakages,the content of the reduced glutathione( GSH) and activity of superoxide dismutase( SOD) in lens tissue were assayed. Results Abalone shell extract in the concentration range of 1 ~ 2 mg / ml reduced the lens opacity caused by oxidative stress,alleviated the release of LDH,and increased GSH content and SOD activity in cultured lens.     魁蚶(Scapharca broughtonii)热休克蛋白90(HSP90)基因的克隆及转录表达分析

    热休克蛋白90(HSP90)作为一种研究的常规免疫基因,在许多物种都有过报道。本研究从构建的魁蚶转录组文库中筛选得到的HSP90基因部分序列为基础,通过RACE技术获得其c DNA全长序列(命名为Sb HSP90),以期明确魁蚶HSP90基因的结构特征、组织分布及其对病原菌刺激的免疫变化规律。序列和结构分析表明,该c DNA全长2707bp,编码一个由728个氨基酸组成的多肽,该多肽含有HSP90家族共有的5个签名序列,C端高度保守的MEEVD短肽序列和ATPase结构域;预测蛋白的分子量(Mw)为83.72k Da,理论等电点(p I)为4.85;预测该蛋白无信号肽,具有4个糖基化位点。同源性及系统分析表明,Sb HSP90基因与软体动物的HSP90相似性达到83%以上,其中与长牡蛎(Crassostrea gigas)和海湾扇贝(Argopecten irradians)相似度最高达86%,与甲壳动物HSP90的相似度都在81%左右,与脊椎动物HSP90-α和HSP90-β的同源性都很接近。实时荧光定量PCR(q RT-PCR)结果表明:Sb HSP90 m RNA在魁蚶血细胞、斧足、鳃、外套膜、闭壳肌和肝胰腺和中均有表达,斧足中的表达量相对较高,而在肝胰腺中的表达量则相对较低;注射鳗弧菌后,相对于对照组,Sb HSP90基因在所检测的每个组织中m RNA水平上的表达量都显著上调(P<0.01),而且具有显著的时间依赖性和瞬时表达趋势。国家自然科学基金项目,31602142号;;中国水产科学研究院基本科研业务费项目,2017GH07号;;浙江省重中之重学科开放基金,KF2015007

    本文叙述了使用差热分析、X射线衍射、偏光显微镜和扫描电子显微镜等方法对钠化钒渣中几种钒酸钠物相分析研究的结果。水溶性钒酸钠有三种,偏钒酸钠—NaVO_3、焦钒酸钠—Na_4V_2O_7、正钒酸钠—Na_3VO_4。钠化钒渣中最主要的钒酸钠是正钒酸钠和少量的焦钒酸钠。焦正钒酸钠的水溶性较偏钒酸钠好。为了获得较纯的各种钒酸钠单矿物,用化学纯的V_2O_5、Na_2CO_3试剂,按照Na_2O/V_2O_5克分子比为1:1、2:1、8:1的配比,在高温差热分析上加热至900℃合成了偏、焦、正钒酸钠矿物,取得了三种矿物的热化学数据,对三种矿物的晶体结构和晶体光性进行分析测定。偏钒酸钠晶体呈柱状和板状,淡黄色,N_p1.74,N_m、N_p>1.74,二轴晶正光性。焦钒酸钠晶体呈羽扇状和束状,淡黄至白色,N_p=1.630±0.015,N_m=1.635,N_g=1.643±0.003二轴晶正光性。正钒酸钠晶体呈树枝状和粒状等,黄褐色,等轴晶系,光性异常,N=1.610±0.010。焦、正钒酸钠吸附水或溶水后重结晶可形成Na_4V_2O_7·12H_2O、Na_3VO_4·12H_2O、Na_3VO_4·16H_2O等多种含水相
