    Modeling Influence of Temperature and Salinity on Growth Rate of Larva in Noble Scallop,Chlamys nobilis(Reeve) by Response Surface Methodology(RSM)

    采用中心复合设计(CEnTrAl COMPOSITE dESIgn,CCd)的试验设计法研究了温度和盐度2个环境因子对华贵栉孔扇贝幼虫生长的影响。同时采用响应曲面分析法及基于响应曲面基础之上的满意度函数法确立了生长的二次回归方程,同时进行优化处理得到温度与盐度的最佳组合。结果表明,在华贵栉孔扇贝幼虫生长过程中温度与盐度在一定的范围内的互作效应不显著,其中温度效应较为明显。温度对壳长瞬时生长影响的一次项效应和二次项效应均达到显著水平(P<0.000 1)。盐度对壳长瞬时生长影响一次项效应显著(P<0.000 1),然而,盐度对壳长瞬时生长影响的二次项效应达到显著水平(P<0.000 1)。分析响应曲面法优化温度和盐度对华贵栉孔扇贝幼虫壳长瞬时生长率的影响,其结果显示最佳温度与盐度组合为25.33℃与29.42‰,此条件下瞬时生长率为7.91%,其满意度函数值达到85.57%。在生产实践中,严格控制温度和盐度范围,使其在最优值范围内,将会显著提高苗种生产效率。In this study,the combined effects of temperature and salinity on the instantaneous growth rate(IGR) of larva in noble scallop,Chlamys nobilis,was evaluated using the central composite orthogonal quadratic design and the response surface methodology.The results showed that the linear effects of temperature on IGR was statistically highly significant(P<0.000 1);the linear effects of salinity on IGR was significant(P<0.000 1).The interactions between temperature and salinity on IGR was not significant;the quadratic effects of temperature and salinity on the IGR was highly significant(P<0.000 1);temperature was more important in influencing growth of larvaof C.nobilis.By applying the simultaneous optimization technique,the optimum factor combination,i.e.,25.33 ℃ and 29.42‰ was found out,at which the optimal IGR(7.91%)arrived simultaneously,with the desirability value as high as 85.57 %.It is anticipated that application of the optimal temperature-salinity combination to practice would improve the productive efficiency of C.nobilis.国家自然科学基金(31160528); 国家海洋公益性行业科研专项(201405020-5); 海洋公益专项科研基金(201205021); 南海生物资源开发与利用协同创新中

    前梯度蛋白2(AnTErIOr grAdIEnT 2,Agr2)是一种分泌蛋白,在多种腺癌中过表达,是一个潜在的肿瘤诊断和预后判断的标志物。用质粒PgEX4T-1构建Agr2蛋白原核表达载体(PgEX4T-1-△SPAgr2),IPTg诱导表达,考马斯亮蓝染色和WESTErn blOT验证纯化的Agr2蛋白。结果表明,利用大肠埃希菌培养,原核表达载体(PgEX4T-1-△SP-Agr2)在25℃、0.1MMOl/l IPTg诱导16H后Agr2蛋白大量表达,分离纯化得到具有生物活性的成熟Agr2蛋白,这对于开展Agr2生物机制的研究,以及开发Agr2的诊断和治疗试剂具有重要的意义。Anterior gradient-2(AGR2) is a secreted protein and overexpressed in many tumor tissues of glands.Thus,AGR2 is considered to be a promising biomarker for early diagnosis and prognosis on cancer.It was used to express AGR2 by prokaryotic expresion vector pGEX4T-l-ASP-AGR2 in E.coli.After induction with IPIG,the protein was assessed with Coomassie bright blue gel and Western blot.The results showed that the AGR2 vector was induced with IPTG at 25℃ for 16 hours to get high expression of pure and bioactive AGR2 protein.Our results suggest potential uses in the study the mechanism of AGR2 as a secreted prote of the tumor.In a broader sense,they also has important theoretical and practical significance for early diagnosis and prognosis of cancer.福建省自然科学基金项目(2012J01155); 中央高校基本科研业务费项目; 厦门大学药学院院长基金项

    HCV 广东株5’NCRc DNA的克隆及序列测定

    摘 要 从广东省1例慢性丙型肝炎病人血清中提取HCV RN A,随机引物逆转录为cDN A后用HCV 5’ 端非编码区( 5’ NCR)特异引物进行聚合酶链反应( PC R)。扩增产物302 bp,经补齐和提纯后插入pUC 19质 粒,获得的重组质粒pUN采用双脱氧链终止法测定核苷酸序列,与国内外多个株比较,核苷酸同源性介乎 92. 69%~ 100% , 其中与HCV Ⅱ ( 1b)型的同源性最大。本文的测序结果可为引物设计提供依据。获得的HCV cDNA在常规PCR步骤中用于设立有效的模板对照, 对消除假阴性及评估试剂有重要意义

    本研究采用荧光显微技术对皱纹盘鲍(Halilotis discus hannai)♀×西氏鲍(H.gigantea)♂的受精过程及受精卵的早期发育进行了观察,并采用FITC-anti-α-tubulin对精子入卵位置进行标记.结果显示西氏鲍的精子可正常激活皱纹盘鲍的卵子,精子进入卵子2-5 min后受精卵开始第一次卵裂,释放出第一极体.随着受精卵的发育卵子完成第二次减数分裂,排出第二极体,同时形成早期精卵原核.精卵原核膨胀并彼此融合,受精卵开始进行有丝分裂完成第一次卵裂.通过FITC标记的anti-α-tubulin蛋白免疫荧光观察,发现精子在入卵时会在卵膜表面,形成一个类似于受精孔的结构.The current study was conducted on the fertilization process and the early development of fertilized eggs by the fluorescence microscopy technique. The position of sperm into the eggs was labeled by FITC - anti - α - tubulin. The results showed that sperm of H. gigantea penetrated H. discus hannai eggs. The first cleavage and the first polar body were released in after 2 -5min of sperm - egg fusion. With the completion of the second meiotic division of the fertilized egg, the second polar body was released, forming the early male and female pronucleus. Then two pronucleus began to swell and started mutual fusion, and the fertilized eggs began to undergo mitosis to complete the first cleavage. The immunofluorescent observation with the FITC labeled anti - α - tubulin probe detected a pore structure similar to micropylar on the surface of egg membrane.海南省高等学校科学研究项目(HNKY2016-43

    探讨了一种基于SystemC的RISCCPU建模方法,建立了一个SPARC精简指令集CPU的整数单元(IU)行为级模型.该模型作为指令集仿真器(ISS),基本达到了RISCCPU的功能要求,为程序提供了一个CPU模拟平台,减少了软硬件协同设计的周期,这对片上系统SOC的实现具有重要的意义.In this paper, a behavior model of Interger Unit(IU) which is a part of SPARC RISC is designed with SystemC.
