Rapid identification of pathogens using UPPCR with SSCP and RFLP analysis

目的　建立一种快速检测病原菌的方法以利于临床诊断。方法　选取 10种具有代表性的病原菌 ,采用通用引物PCR(UPPCR)对其 16SrRNA基因进行扩增 ,并对PCR产物进行限制性酶切片段长度多态性分析 (RFLP)和单链构象多态性分析 (SSCP)。结果　RFLP电泳图谱呈现多态性 ,但半数菌的图谱两两相同或相似 ;SSCP电泳图谱各异 ,可以相互区分。结论　UPPCR SSCP技术能快速简便地检测病原菌Objective To establish a method for the identification of pathogens for fast diagnosis. Methods By universal primer polymerase chain reaction (UPPCR), the 16S ribosomal RNA genes of ten representative pathogens were amplified. Then the PCR products were analyzed by single strand configuration polymorphism (SSCP) and restrict fragment length polymorphism (RFLP). Results RFLP map expressed polymorphism, but there were either the same or similar figures between two or among three pathogens in half of test strains. However, there were significantly different bands in all of the pathogens by SSCP. Conclusion UPPCR SSCP analysis can rapidly and conveniently identify bacterial pathogens. Correlation of C3/Ig Two component determined Circulating Immune Complexes with Humoral Immunityin Healthy Individuals

采用直线相关分析法，较系统地研究了健康人 C3/ Ig 双特异性循环免疫复合物（ C3/ Ig M T C I C、 C3/ Ig g T C I C 和 C3/ Ig A T C I C）与 Ig M 、 Ig g、 Ig A 和 C3 的相关性.结果发现， C3/ Ig M T C I C 和 C3/ Ig g T C I C 含量主要取决于血清总 Ig M 和 Ig g 量； C3/ Ig g T C I C 与复合的 Ig M 、复合的 Ig g、总的 C3 呈正相关，而与复合的 C3 呈负相关.这些结果提示， Ig g 在健康人体液免疫中的重要作用.同时说明，健康人机体免疫反应是复杂多样的，深入研究 C3/ Ig T C I C 与体液免疫的相关性及其形成机理，将为揭示健康人的体液免疫机制提供重要参考.Correlation of C3/Ig two component determined circulating immune complexes(C3/IgM TCIC、C3/IgG TCIC和C3/IgA TCIC)with IgM,IgG,IgA and C3 has been characterized in healthy individuals.The results showed that the levels of C3/IgM TCIC and C3/IgG TCIC depended mainly on those of total serum IgM and IgG.And C3/IgG TCIC were positively correlated to complexed IgM,complexed IgG and total serum C3,whereas negatiely to complexed C3.These findings suggest that IgG plays an important role in humoral immunity in healthy individuals.Moreover,further study on correlation mechanism of Ig/C3 TCIC with humoral immunity will provide useful data for understanding of immunity in healthy individuals.福建省自然科学基
