Hematological and biochemical response in the blood of Alburnus tarichi (Actinopterygii: Cypriniformes: Cyprinidae) exposed to tebuconazole
Background. Lake Van, the second-largest soda lake in the world, has a pH 9.8 value. A fish, locally known as Van fish, Alburnus tarichi (Güldenstädt, 1814), is an endemic fish of Lake Van and known also as pearl mullet, tarek, or Van bleak. Tebuconazole is a widely used pesticide around Lake Van. In this study, we will focus on the effects of tebuconazole on Van fish blood to provide critical information on the environmental risk assessment of pesticides in various aquatic environments. Materials and methods. The Van fish were exposed to tebuconazole for 24, 48, 72, and 96 h at a concentration of 2.5 mg ·L–1. Subsequently, the resulting hematological and biochemical parameters were determined. Results. There was a statistically significant decrease in erythrocytes (RBC), hemoglobin (Hb), and hematocrit (Hct) in the blood parameters (P < 0.05). The levels of serum, aspartate aminotransferase (AST), alanine aminotransferase (ALT), lactate dehydrogenase (LDH), urea, and creatine increased significantly (P < 0.05). The serum cortisol level increased significantly at all hours after the administration of tebuconazole (P < 0.05). Conclusion. As a result, toxicity caused by pesticides caused negative changes in the biochemical and hematological values of Van fish. Changes in these parameters have shown that it can be used as a biomarker for toxicity
Factor IX gene mutations in turkish haemophiliacs
Hemofili B hastalığının nedeni, kanın temel fonksiyonları
arasında bulunan koagülasyon mekanizmasında
etkin olan faktör IX’un konjenital eksikliği veya
disfonksiyonel olmasıdır. Faktör IX’u kodlayan gen, X
kromozomunun q27.1 bandında lokalize olup, 34 kilobaz
(kb) uzunluğunda ve 8 ekzon ile 7 introndan oluşmaktadır.
Hem genotipik hem de fenotipik olarak oldukça heterojen
bir yapıda olan hemofili B hastalığının şiddeti, gendeki
mutasyonun lokalizasyonu ve yapısıyla yakından ilgilidir.
Bu çalışmadaki temel amaç, yüksek heterojenite gösteren
faktör IX geninde, hızlı, efektif ve kesin sonuç veren otomatik
kapiller jel elektroforezi ile DNA baz dizileme yöntemini
kullanarak, kesin moleküler tanıya gidilmesi, yöntemin
avantajlarının belirlenmesi ve Türk
popülasyonundaki mutasyon profilinin ortaya çıkartılmasına
yönelik çalışmalara ışık tutmasıdır.
Yapılan çalışmada, klinik olarak tanısı konmuş 13 hemofili
B hastasında, otomatik kapiller jel elektroforezi yöntemi
ile DNA dizi analizi yapılarak mutasyonlar taranmış ve
2’si yeni olmak üzere 11 farklı mutasyon tanımlanmıştır.
Tespit edilen mutasyonlar en çok ( %33 ) 8. ekzon bölgesinde
gözlenmiştir. Ayrıca, faktör IX geninin CpG’den
zengin bölgelerinde mutasyonların daha çok ( %36 ) oldu-
ğu görülmüş ve missens mutasyonların faktör IX geninde
hastalığa en fazla ( %46 ) neden olan mutasyon tipi oldu-
ğu, literatürle de uyum gösteren bir şekilde ortaya konmuştur.
Sonuç olarak, kapiller DNA dizi analizi yönteminin, faktör
IX geni mutasyonlarının tanımlanmasında hızlı ve etkili
bir yaklaşım olduğunu ve bu çalışmanın sonuçlarının,
Türk popülasyonunun faktör IX geni mutasyon profilinin
belirlenmesi ile ilgili başka çalışmalara da ışık tutacağını
düşünmekteyiz.Hemophilia B is an X linked coagulopathy
due to deficiency of clotting factor IX. The gene for factor
IX is situated on the long arm of the X chromosome at
band Xq27.1, and spans 34kb. It consists of eight exons
and seven introns. The clinical and molecular basis of
hemophilia B is heterogeneous and the clinical severity is
related to the location and type of genetic mutation.
The main purpose of this study was to perform molecular
diagnosis of hemophilia B disease by DNA sequence
analyses of the promoter, poly A and coding regions of the
factor IX gene. To evaluate the efficiency of this
technique in the diagnosis of factor IX gene mutations and
light up further studies that aim to obtain mutation profile
of the factor IX gene spesific to Turkish population.
By this aim, blood samples from thirteen clinically
diagnosed hemophilia B patients were analysed by the
automatic capiller gel electrophoresis technique. At least
one mutation was identified in the patients, except one
case with mild hemophilia B. Of 11 identified molecular
abnormalities, two were new mutations according to
“Hemophilia B mutation bank” data of the recognised
mutations. The most frequently seen mutations (33%)
were localised at the exon 8 of the factor IX gene. The
frequency of mutations was higher in the CpG rich regions
of the gene and missense mutations was regarded as the
most common type of mutations causing severe
hemophilia B and this conclusion was in accordance with
the literature.
In conclusion, we suggested that capiller DNA sequencing
is a fast and efficient approach to identify mutations in the
factor IX gene and the results of this study will light up
further studies in obtaining the factor IX gene mutation
profile of the Turkish population
Dvantages of the combination of VNTR linkage and direct mutation analysis in prenatal and postnatal diagnosis of phenylketonuria
Bu çalışmada, Fenilketonüri tanısı konulmuş hasta bir çocuğa sahip 20 ailede, hasta çocuğun kardeşlerinin taşıyıcı olup olmadıklarım saptamak için PAH geni dışındaki VNTR polimorfizimlerinden faydalanarak alel segregasyonu yapılmıştır. Hastalarda IVS10nt546 mutasyonun varlığını araştırmak için Ddel restriksiyon enzimi direkt mutasyon analizi yöntemi uygulanmıştır. DNA venöz kandan ekstrakte edilmiştir. Ekstragenik VNTR dizileri PCR kullanılarak amplifiye edilmiş ve alel segregasyonu yapmak için de PCR ürünü jelde yürütülmüştür. İnformatif ailelerde hasta çocuğun kardeşlerinin taşıyıcılığı tesbit edilmiştir. Yirmi aileden 18 tanesi informatif, 9 çocuk taşıyıcı ve 6 çocuk sağlam bulunmuştur. İnformatif ailelere prenatal tanı yapılabileceği bildirilmiştir.
Aynı hasta çocuklarda IVS10nt546 mutasyonuna yönelik olarak amplifiye edilen PAH geni PCR ürünü DdeI restrik- siyon enzimi ile direkt mutasyon analizine tabi tutulmuştur. Toplam 40 alelden 12 sinde (%30) IVS10nt546 mutasyonu bulunmuştur. PAH-VNTR polimorfizimleri ile alel segregasyonu yapılamayan bir ailede hasta çocuğun IVS10nt546 mutasyonunu homozigot olarak taşıması, kardeşinin PKU taşıyıcısı olduğunu ortaya koymamızı sağlamıştır. Yalnız başına PAH-VNTR polimorfizimleri ile taşıyıcı tespiti başarısı %90 iken, DdeI restriksiyon enzimi direkt mutasyon analizi yöntemiyle birlikte kullanıldığında başarı oranı %95 e çıkmıştır.In this dissertation, allele segregation was performed utilizing VNTR polymorphisms outside the PAH gene in order to determine whether the siblings of a child who has been diagnosed with PKU are carriers. Blood samples from 20 children who had PKU and their parents as well as their siblings were studied. Ddel restriction enzyme direct mutation analysis technique was applied to determine whether the IVS10nt546 mutation existed in patients. DNA was extracted from the venus blood sample. The extragenic VNTR sequences were amplified using PCR, and the PCR product was run on the gel for ellele segragation. With this process, it was determined whether a sibling of a sick child is a carrier in “informative” families. With this method, 18 families out of 20 were found to be “informative”; 9 children, carriers; 6 children, normal. It was concluded that the prenatal diagnosis was applicable to those informative families.
In the samples from the PKU patients, the PAH gene which was amplified by PCR regionally including the IVS10nt546 mutation. IVS10nt546 mutation was found in 12 alleles out of 40 (30%) by digesting the PCR products with DdeI restriction enzyme. In a noninformative family, where the allele segregation with PAH-VNTR polymorphism failed, a sick child was found to carry IVS10nt546 mutation homozgote. This led to the conclusion that the sibling of the child was a PKU carrier. While the success rate in carrier determination was 90% with PAH-VNTR polymorphism alone, the rate increased to 95% when DdeI restriction enzyme direct mutation analysis was used additionally
