    Mixed mycobacterial infections in farmed sturgeons

    Based on microbiological and histopathological examinations and DNA sequencing, several outbreaks of mycobacteriosis in the reared sturgeons, including Chinese sturgeon (Acipenser sinensis Gray) and Amur sturgeon (Acipenser schrencki), were identified during 2009 to 2010. Forty-nine isolates of non-tuberculous mycobacteria(NTM)were isolated from 19 diseased sturgeons. In total, seven species of Mycobacterium were identified, namely, Mycobacterium chelonae, Mycobacterium marinum, Mycobacterium gordonae, Mycobacterium fortuitum, Mycobacterium szulgai, Mycobacterium arupense and Mycobacterium porcinum. Among them, M. marinum was found to be more prevalent (89.5%) compared with the other mycobacterial species. When two molecular biological methods, PCR-DGGE (denaturing gradient gel electrophoresis) analysis and rpoB gene library sequencing, were used to analyse the mycobacterial DNAs extracted from the diseased fish tissues, mixed infections of two or three mycobacterial species were found being the predominant infection form (94.7%) in sturgeon mycobacteriosis. M. marinum was the only one species that caused sturgeon mycobacteriosis alone. Virulence assay showed that M. marinum possessed stronger pathogenicity to zebrafish killing 100% of fish in 28days at 10(3) cfu/fish than the other species. These results suggested that M. marinum is the major pathogenic bacteria in sturgeon mycobacteriosis. To the best of our knowledge, this study is the first report on mycobacteriosis in farmed Chinese and Amur sturgeons as well as the first isolation of M. porcinum and M. arupense from fish

    Graphene-Based Anticancer Nanosystem and Its Biosafety Evaluation Using a Zebrafish Model

    In this paper, a facile strategy to develop graphene-based delivery nanosystems for effective drug loading and sustained drug release was proposed and validated. Specifically, biocompatible naphthalene-terminated PEG (NP) and anticancer drugs (curcumin or doxorubicin (DOX)) were simultaneously integrated onto oxidized graphene (GO), leading to self-assembled, nanosized complexes. It was found that the oxidation degree of GO had a significant impact on the drug-loading efficiency and the structural stability of nanosystems. Interestingly, the nanoassemblies resulted in more effective cellular entry of DOX in comparison with free DOX or DOX-loaded PEG-polyester micelles at equivalent DOX dose, as demonstrated by confocal microscopy studies. Moreover, the nanoassemblies not only exhibited a sustained drug release pattern without an initial burst release, but also significantly improved the stability of formulations which were resistant to drug leaking even in the presence of strong surfactants such as aromatic sodium benzenesulfonate (SBen) and aliphatic sodium dodecylsulfonate (SDS). In addition, the nanoassemblies without DOX loading showed negligible in vitro cytotoxicity, whereas DOX-loaded counterparts led to considerable toxicity against He La cells. The DOX-mediated cytotoxicity of the graphene-based formulation was around 20 folds lower than that of free DOX, most likely due to the slow DOX release from complexes. A zebrafish model was established to assess the in vivo safety profile of curcumin-loaded nanosystems. The results showed they were able to excrete from the zebrafish body rapidly and had nearly no influence on the zebrafish upgrowth.     Application of large-scale integrated vertical-flow constructed wetland in Beijing Olympic forest park: design, operation and performance

    This paper deals with the design, purification performance and public questionnaire of the integrated vertical-flow constructed wetland (IVCW) system built in Beijing Olympic Forest Park (BOFP). This IVCW system of 45 000 m2 has been operated to treat reclaimed wastewater and circulated lake water from Aohai lake. The first 6-month monitoring results in 2008 indicated that IVCW showed a statistically significant removal of TP and OP from reclaimed water and the removal of BOD5 from circulated lake water from Aohai lake. The questionnaire of Willingness To Pay (WTP) by visitors to the BOFP showed that 98.1% of the respondents were in favour of the construction of the wetland and approved the performance of IVCW. More importantly, during the periods of Beijing 2008 Olympic Games and Paralympic Games, the IVCW system helped to provide amenable surroundings for sight-seeing athletes and audiences, as well as tourists. The IVCW also helped to embody the concept of Harmony Between Human and Nature, a theme of the Olympics Games.     An unprecedented organocatalytic asymmetric Michael/cyclization cascade reaction of 3-isothiocyanato oxindoles and. 3-nitroindoles has-been disclosed. A wide range of enantioenriched polycyclic, spirooxindoles, containing three contiguous chiral centers With two of them having quaternary stereocenters, could be smoothly obtained with satisfactory results (up to 99%-yield, >99:1 dr, and 96% ee). This methods very promising because the reaction is scalable, and the versatile transformations of the products into other spirocyclic oxindoles are also feasible
