    The public face of event volunteering at the 2006 Commonwealth Games The media perspective

    Volunteers play a major role in mega and major events, both cultural and sporting, contributing invaluable human resources. Frequently described (and, maybe, over-emphasised) as the 'unsung heroes' of such events, relatively little is known about this group of workers when compared with volunteers who give regular and long-term commitment to their area of interest. This paper focuses on media representation and perceptions of volunteering at the 2006 Commonwealth Games in Melbourne. The exploratory study adopts an extended timeframe and reports on media representation of volunteers over the full event cycle from recruitment through training and the actual event, concluding with consideration of the Games' aftermath. The findings highlight various positive and negative aspects of the media representation of volunteering at this type of mega event and how this has the potential to influence its public face. Conclusions focus on the implications for the media management of volunteering at major events and suggestions for future research in this area

    If money talks, what does it say? Varieties of capitalism and business financing of parties

    Do business contributions to political parties convey different messages in different countries, and, if so, why? This is the first cross-national study of firm behaviour in political finance. It understands motivations for contributions to parties as either ideological or pragmatic. Motivation is inferred by quantitatively relating the payments of 960 firms to variations in political competition in three countries over periods of between seven and seventeen years. In co-ordinated Germany, a small number of firms make ideological payments. In liberal Canada and Australia, large numbers of firms made pragmatic payments. Australia’s left-right party system created an awareness of policy risk, which motivated ideological payments, but there was no ideological bias in business financing of politics in Canada’s unusually non-ideological party system. The statistical analysis is supplemented by a qualitative investigation of discrete and reciprocal exchanges between businesses and political parties

    Metformin Treatment Attenuates Brain Inflammation and Rescues PACAP/VIP Neuropeptide Alterations in Mice Fed a High-Fat Diet.

    High-fat diet (HFD)-induced comorbid cognitive and behavioural impairments are thought to be the result of persistent low-grade neuroinflammation. Metformin, a first-line medication for the treatment of type-2 diabetes, seems to ameliorate these comorbidities, but the underlying mechanism(s) are not clear. Pituitary adenylate cyclase-activating peptide (PACAP) and vasoactive intestinal peptide (VIP) are neuroprotective peptides endowed with anti-inflammatory properties. Alterations to the PACAP/VIP system could be pivotal during the development of HFD-induced neuroinflammation. To unveil the pathogenic mechanisms underlying HFD-induced neuroinflammation and assess metformin's therapeutic activities, (1) we determined if HFD-induced proinflammatory activity was present in vulnerable brain regions associated with the development of comorbid behaviors, (2) investigated if the PACAP/VIP system is altered by HFD, and (3) assessed if metformin rescues such diet-induced neurochemical alterations. C57BL/6J male mice were divided into two groups to receive either standard chow (SC) or HFD for 16 weeks. A further HFD group received metformin (HFD + M) (300 mg/kg BW daily for 5 weeks) via oral gavage. Body weight, fasting glucose, and insulin levels were measured. After 16 weeks, the proinflammatory profile, glial activation markers, and changes within the PI3K/AKT intracellular pathway and the PACAP/VIP system were evaluated by real-time qPCR and/or Western blot in the hypothalamus, hippocampus, prefrontal cortex, and amygdala. Our data showed that HFD causes widespread low-grade neuroinflammation and gliosis, with regional-specific differences across brain regions. HFD also diminished phospho-AKT(Ser473) expression and caused significant disruptions to the PACAP/VIP system. Treatment with metformin attenuated these neuroinflammatory signatures and reversed PI3K/AKT and PACAP/VIP alterations caused by HFD. Altogether, our findings demonstrate that metformin treatment rescues HFD-induced neuroinflammation in vulnerable brain regions, most likely by a mechanism involving the reinstatement of PACAP/VIP system homeostasis. Data also suggests that the PI3K/AKT pathway, at least in part, mediates some of metformin's beneficial effects

    ViP-CNN: Visual Phrase Guided Convolutional Neural Network

    As the intermediate level task connecting image captioning and object detection, visual relationship detection started to catch researchers' attention because of its descriptive power and clear structure. It detects the objects and captures their pair-wise interactions with a subject-predicate-object triplet, e.g. person-ride-horse. In this paper, each visual relationship is considered as a phrase with three components. We formulate the visual relationship detection as three inter-connected recognition problems and propose a Visual Phrase guided Convolutional Neural Network (ViP-CNN) to address them simultaneously. In ViP-CNN, we present a Phrase-guided Message Passing Structure (PMPS) to establish the connection among relationship components and help the model consider the three problems jointly. Corresponding non-maximum suppression method and model training strategy are also proposed. Experimental results show that our ViP-CNN outperforms the state-of-art method both in speed and accuracy. We further pretrain ViP-CNN on our cleansed Visual Genome Relationship dataset, which is found to perform better than the pretraining on the ImageNet for this task.Comment: 10 pages, 5 figures, accepted by CVPR 201

    Structural effects and potential changes in growth factor signalling in penis-projecting autonomic neurons after axotomy

    BACKGROUND: The responses of adult parasympathetic ganglion neurons to injury and the neurotrophic mechanisms underlying their axonal regeneration are poorly understood. This is especially relevant to penis-projecting parasympathetic neurons, which are vulnerable to injury during pelvic surgery such as prostatectomy. We investigated the changes in pelvic ganglia of adult male rats in the first week after unilateral cavernous (penile) nerve axotomy (cut or crush lesions). In some experiments FluoroGold was injected into the penis seven days prior to injury to allow later identification of penis-projecting neurons. Neurturin and glial cell line-derived neurotrophic factor (GDNF) are neurotrophic factors for penile parasympathetic neurons, so we also examined expression of relevant receptors, GFRα1 and GFRα2, in injured pelvic ganglion neurons. RESULTS: Axotomy caused prolific growth of axon collaterals (sprouting) in pelvic ganglia ipsilateral to the injury. These collaterals were most prevalent in the region near the exit of the penile nerve. This region contained the majority of FluoroGold-labelled neurons. Many sprouting fibres formed close associations with sympathetic and parasympathetic pelvic neurons, including many FluoroGold neurons. However immunoreactivity for synaptic proteins could not be demonstrated in these collaterals. Preganglionic terminals showed a marked loss of synaptic proteins, suggesting a retrograde effect of the injury beyond the injured neurons. GFRα2 immunofluorescence intensity was decreased in the cytoplasm of parasympathetic neurons, but GFRα1 immunofluorescence was unaffected in these neurons. CONCLUSION: These studies show that there are profound changes within the pelvic ganglion after penile nerve injury. Sprouting of injured postganglionic axons occurs concurrently with structural or chemical changes in preganglionic terminals. New growth of postganglionic axon collaterals within the ganglion raises the possibility of the formation of aberrant synaptic connections between injured and un-injured ganglion neurons. Together these changes demonstrate a broader effect on the pelvic autonomic circuitry than simply loss of neuroeffector connections. These structural changes are accompanied by potential changes in neurotrophic factor signalling due to altered expression of receptors for members of the GDNF family. Together our results advance understanding of the responses of pelvic autonomic nerve circuits to injury and may assist in designing strategies for promoting regeneration

    PACAP and VIP Mitigate Rotenone-Induced Inflammation in BV-2 Microglial Cells.

    Rotenone is a commercial pesticide commonly used to model Parkinson's disease (PD) due to its ability to induce dopaminergic degeneration. Studies have confirmed that rotenone causes microglial activation, which seems to contribute to the toxic effects seen in rodent models. Pituitary adenylate cyclase-activating polypeptide (PACAP) and vasoactive intestinal peptide (VIP) are two structurally related neuropeptides that have robust neuroprotective and anti-inflammatory properties. However, their ability to regulate microglial activity in response to rotenone is not fully understood. Using rotenone as an inflammatory stimulus, we tested whether PACAP or VIP could mitigate microglial activation in BV2 microglial cells. Rotenone dose-dependently reduced cell viability and the percentage of apoptotic cells. It also increased the release of nitric oxide (NO) in culture media and the expression of microglial activation markers and pro-inflammatory markers, including CD11b, MMP-9 and IL-6, and heightened the endogenous levels of PACAP and its preferring receptor PAC1. Co-treatment with PACAP or VIP prevented rotenone-induced increase of NO, CD11b, MMP-9 and IL-6. These results indicate that both PACAP and VIP are able to prevent the pro-inflammatory effects of rotenone in BV2 cells, supporting the idea that these molecules can have therapeutic value in slowing down PD progression

    Pancreatic Polypeptide Controls Energy Homeostasis via Npy6r Signaling in the Suprachiasmatic Nucleus in Mice

    SummaryY-receptors control energy homeostasis, but the role of Npy6 receptors (Npy6r) is largely unknown. Young Npy6r-deficient (Npy6r−/−) mice have reduced body weight, lean mass, and adiposity, while older and high-fat-fed Npy6r−/− mice have low lean mass with increased adiposity. Npy6r−/− mice showed reduced hypothalamic growth hormone releasing hormone (Ghrh) expression and serum insulin-like growth factor-1 (IGF-1) levels relative to WT. This is likely due to impaired vasoactive intestinal peptide (VIP) signaling in the suprachiasmatic nucleus (SCN), where we found Npy6r coexpressed in VIP neurons. Peripheral administration of pancreatic polypeptide (PP) increased Fos expression in the SCN, increased energy expenditure, and reduced food intake in WT, but not Npy6r−/−, mice. Moreover, intraperitoneal (i.p.) PP injection increased hypothalamic Ghrh mRNA expression and serum IGF-1 levels in WT, but not Npy6r−/−, mice, an effect blocked by intracerebroventricular (i.c.v.) Vasoactive Intestinal Peptide (VPAC) receptors antagonism. Thus, PP-initiated signaling through Npy6r in VIP neurons regulates the growth hormone axis and body composition

I am interested in where these borders wear thin and threaten to break and disrupt social order. I explore the bodies of gay pride parades because it is bodies such as these that threaten the borders of corporeal acceptability
