    Dependability where the mobile world meets the enterprise world

    As we move toward increasingly larger scales of computing, complexity of systems and networks has increased manifold leading to massive failures of cloud providers (Amazon Cloudfront, November 2014) and geographically localized outages of cellular services (T-Mobile, June 2014). In this dissertation, we investigate the dependability aspects of two of the most prevalent computing platforms today, namely, smartphones and cloud computing. These two seemingly disparate platforms are part of a cohesive story—they interact to provide end-to-end services which are increasingly being delivered over mobile platforms, examples being iCloud, Google Drive and their smartphone counterparts iPhone and Android. ^ In one of the early work on characterizing failures in dominant mobile OSes, we analyzed bug repositories of Android and Symbian and found similarities in their failure modes [ISSRE2010]. We also presented a classification of root causes and quantified the impact of ease of customizing the smartphones on system reliability. Our evaluation of Inter-Component Communication in Android [DSN2012] show an alarming number of exception handling errors where a phone may be crashed by passing it malformed component invocation messages, even from unprivileged applications. In this work, we also suggest language extensions that can mitigate these problems. ^ Mobile applications today are increasingly being used to interact with enterprise-class web services commonly hosted in virtualized environments. Virutalization suffers from the problem of imperfect performance isolation where contention for low-level hardware resources can impact application performance. Through a set of rigorous experiments in a private cloud testbed and in EC2, we show that interference induced performance degradation is a reality. Our experiments have also shown that optimal configuration settings for web servers change during such phases of interference. Based on this observation, we design and implement the IC 2engine which can mitigate effects of interference by reconfiguring web server parameters [MW2014]. We further improve IC 2 by incorporating it into a two-level configuration engine, named ICE, for managing web server clusters [ICAC2015]. Our evaluations show that, compared to an interference agnostic configuration, IC 2 can improve response time of web servers by upto 40%, while ICE can improve response time by up to 94% during phases of interference

    Biosensors

    A biosensor is defined as a detecting device that combines a transducer with a biologically sensitive and selective component. When a specific target molecule interacts with the biological component, a signal is produced, at transducer level, proportional to the concentration of the substance. Therefore biosensors can measure compounds present in the environment, chemical processes, food and human body at low cost if compared with traditional analytical techniques. This book covers a wide range of aspects and issues related to biosensor technology, bringing together researchers from 11 different countries. The book consists of 16 chapters written by 53 authors. The first four chapters describe several aspects of nanotechnology applied to biosensors. The subsequent section, including three chapters, is devoted to biosensor applications in the fields of drug discovery, diagnostics and bacteria detection. The principles behind optical biosensors and some of their application are discussed in chapters from 8 to 11. The last five chapters treat of microelectronics, interfacing circuits, signal transmission, biotelemetry and algorithms applied to biosensing

    Transformational Leadership and the Adoption of Sustainability and Asset Management Practices in Ground Transportation organizations

    The unprecedented challenges the infrastructure domain has been facing for the past few decades are forcing the authorities and owners of relevant organizations, particularly within the ground transportation sector, to introduce more innovative measures and to mitigate risks by embracing and adopting sustainability and asset management practices. This study examined the relationship between transformational leadership and the embracing and adopting of sustainability and asset management practices in the U.S. ground transportation sector. Further, this study assessed the mediation effect of organizational innovation on the relationship between transformational leadership style and the leaders\u27 perception in terms of adaptation of sustainability and asset management practices. Moreover, the study investigated the moderation effect of organization type (public and private) on the relationship between transformational leadership and the perception of adopting sustainability and asset management practices. Structural Equation Modeling – Partial Least Square (SEM-PLS) technique was used to analyze the quantitative data collected using a questionnaire survey distributed to a sample of leaders working in the ground transportation sector in the U.S. The results revealed a positive and significant relationship between transformational leadership and the leaders\u27 perception of adopting asset management and sustainability practices. Moreover, the results supported the mediation influence (partial mediation) of organizational innovation on the relationship between transformational leadership and the leaders\u27 perception of adopting sustainability and asset management. The results also revealed the prominent leadership style (transformational) that embraces sustainability and asset management practices. The findings claim that organization type does not have a significant influence on the relationship between transformational leadership and the research\u27s outcomes (adoption of sustainability and asset management). Moreover, according to the results, no significant moderation effect was detected of organization type (public vs. private) on the relationship between transformational leadership and the leaders\u27 perception of embracing and adopting sustainability and asset management practices. The study attempts to fill an important gap in the body of knowledge by investigating the influencing role of leadership style on the leaders\u27 willingness to adopt and embrace sustainability and asset management practices and applications in the ground transportation sector. In addition, the study investigated the mediation effect of leadership styles on the organization\u27s climate for innovation that, in turn, influences the adoption of sustainability and asset management practices. As the results revealed the prominent leadership style that is more likely to embrace sustainability and asset management practices, ground transportation authorities and owners are expected to effectively and seriously consider hiring, recognizing, promoting and endorsing leaders with transformational skills and traits. Further, as the results revealed a positively mediating influence of organizational innovation on the relationship between transformational leadership style and the adoption of sustainability and asset management. ground transportation organizations are urged to identify the factors that both facilitate and inhibit the development of non-technological innovation in their work environment

    The use of computer-aided drug design in small molecule drug discovery

    Drug discovery is one of the most challenging research fields that contributes to the birth of novel drugs for therapeutic use. Due to the complexity and intricate nature of the research, lengthy processes are involved in identifying potential hit molecules for a therapeutic target. To shorten the time required to reach the hit-to-lead stage, computer-aided drug design (CADD) has been used to expedite the process and reduce laboratory expenses. Common strategies used within CADD involve structure-based drug design (SBDD) and ligand-based drug design (LBDD). Both strategies were used extensively in two projects showing the complementarity of each strategy throughout the process. In this work, two separate drug discovery projects are detailed: Design, synthesis and molecular docking study of novel tetrahydrocurcumin analogues as potential sarcoplasmic-endoplasmic reticulum calcium ATPases (SERCA) inhibitors – details the identification, synthesis and testing of potential hit candidate(s) targeting SERCA by using SBDD Filamenting temperature-sensitive mutant Z (FtsZ) as therapeutic target in ligand-based drug design – details the identification, synthesis and testing of potential hit molecule(s) targeting FtsZ In the first project, homology modelling and virtual compound library screening were utilised as the SBDD methods to identify potential hit molecules for testing in P-type calcium ATPases such as SERCA. Preliminary results have found compound 20, an analogue of tetrahydrocurcumin, to show some SERCA inhibitory effect at 300µM based on a SERCA-specific calcium signalling assay performed via fluorometric imaging plate reader. Molecular docking study has also reflected this outcome with desirable ligand-protein binding energies found for 20 when compared with other tested ligands. Pharmacophore screening was used as the main LBDD method in the second project to identify probable hit candidates targeting FtsZ. Potential ligands were synthesised, and tested for antibacterial effect in Bacillus Subtilis strain 168 (Bs168) and Streptococcus pneumoniae strain R6 (SpnR6) cells. One of the tetrahydrocurcumin analogues, compound 4, was found to have minimum inhibitory concentration (MIC) ≤ 10 µM in Bs168 cells and ≤ 2 µM in spnR6 cells. The IC50 values for 4 were 9.1 ± 0.01 µM and 1 ± 0.01 µM in Bs168 and SpnR6 cells respectively. The MIC of 4 was found to be very similar to the MIC of compound 1, a known hit compound targeting against Bs168 cells. On the other hand, the MIC of 4 was lower than the MIC (> 64 µg/mL) of a well-known FtsZ inhibitor, PC190723, against S. pneumoniae. Subsequent molecular docking analyses were completed to evaluate the ligand-protein binding energies to correlate against the testing results. Both compounds 20 and 4 possess some structural similarities and differences that may confer their different effects in these protein targets, which render both with potentials to become the next lead molecules for future development
