    A unified approach to linking experimental, statistical and computational analysis of spike train data

    A fundamental issue in neuroscience is how to identify the multiple biophysical mechanisms through which neurons generate observed patterns of spiking activity. In previous work, we proposed a method for linking observed patterns of spiking activity to specific biophysical mechanisms based on a state space modeling framework and a sequential Monte Carlo, or particle filter, estimation algorithm. We have shown, in simulation, that this approach is able to identify a space of simple biophysical models that were consistent with observed spiking data (and included the model that generated the data), but have yet to demonstrate the application of the method to identify realistic currents from real spike train data. Here, we apply the particle filter to spiking data recorded from rat layer V cortical neurons, and correctly identify the dynamics of an slow, intrinsic current. The underlying intrinsic current is successfully identified in four distinct neurons, even though the cells exhibit two distinct classes of spiking activity: regular spiking and bursting. This approach – linking statistical, computational, and experimental neuroscience – provides an effective technique to constrain detailed biophysical models to specific mechanisms consistent with observed spike train data.Published versio

    Carbon Isotope Constraints on the Deglacial CO2 Rise from Ice Cores

    The stable carbon isotope ratio of atmospheric CO2 (d13Catm) is a key parameter in deciphering past carbon cycle changes. Here we present d13Catm data for the past 24,000 years derived from three independent records from two Antarctic ice cores. We conclude that a pronounced 0.3 per mil decrease in d13Catm during the early deglaciation can be best explained by upwelling of old, carbon-enriched waters in the Southern Ocean. Later in the deglaciation, regrowth of the terrestrial biosphere, changes in sea surface temperature, and ocean circulation governed the d13Catm evolution. During the Last Glacial Maximum, d13Catm and atmospheric CO2 concentration were essentially constant, which suggests that the carbon cycle was in dynamic equilibrium and that the net transfer of carbon to the deep ocean had occurred before then

    Observations of solar wind ion charge exchange in the comet Halley coma

    Giotto Ion Mass Spectrometer/High Energy Range Spectrometer (IMS/HERS) observations of solar wind ions show charge exchange effects and solar wind compositional changes in the coma of comet Halley. As the comet was approached, the He(++) to proton density ratio increased until about 1 hour before closest approach after which time it decreased. Abrupt increases in this ratio were also observed in the beginning and near the end of the so-called Mystery Region (8.6 - 5.5(10)(exp 5) km from the comet along the spacecraft trajectory). These abrupt increases in the density ratio were well correlated with enhanced fluxes of keV electrons as measured by the Giotto plasma electron spectrometer. The general increase and then decrease of the He(++) to proton density ratio is quantitatively consistent with a combination of the addition of protons of cometary origin to the plasma and loss of plasma through charge exchange of protons and He(++). In general agreement with the solar wind proton and He(++) observations, solar wind oxygen and carbon ions were observed to charge exchange from higher to lower charge states with decreasing distance to the comet. The more abrupt increases in the He(++) to proton and the He(++) to O(6+) density ratios in the mystery region require a change in the solar wind ion composition in this region while the correlation with energetic electrons indicates processes associated with the comet

    The mystery of South Africa's ghost workers in 1996: measurement and mismeasurement in the manufacturing census, population census and October Household Surveys

    This paper compares estimates of total manufacturing employment from South Africa's 1996 manufacturing census, the 1996 population census and the October Household Surveys from 1995, 1996 and 1997. Findings show that there are 300 000 too few manufacturing workers recorded in the 1996 population census. Furthermore there are other inconsistencies between these data sources. Several possible explanations for the deficit in manufacturing workers are put forward, but none are compelling. It is concluded that aggregate employment series constructed from household surveys should be treated with caution.

    Reply to comment by B. Andreotti et al. on "Solving the mystery of booming sand dunes"

    This reply addresses three main issues raised in the comment of Andreotti et al. [2008]. First, the turning of ray paths in a granular material does not preclude the propagation of body waves and the resonance condition described by Vriend et al. [2007]. The waveguide model still holds in the dune for the observed velocities, even with a velocity increase with depth as implied by Andreotti et al. [2008]. Secondly, the method of initiation of spontaneous avalanching does not influence the booming frequency. The frequency is independent of the source once sustained booming starts; it depends on the subsurface structure of the dune. Thirdly, if all data points from Vriend et al. [2007] are included in the analysis (and not an average or selection), no correlation is observed between the sustained booming frequency and average particle diameter
