    Bidirectional coupling of splicing and ATM signaling in response to transcription-blocking DNA damage

    In response to DNA damage cells activate intricate protein networks to ensure genomic fidelity and tissue homeostasis. DNA damage response signaling pathways coordinate these networks and determine cellular fates, in part, by modulating RNA metabolism. Here we discuss a replication-independent pathway activated by transcription-blocking DNA lesions, which utilizes the ATM signaling kinase to regulate spliceosome function in a reciprocal manner. We present a model according to which, displacement of co-transcriptional spliceosomes from lesion-arrested RNA polymerases, culminates in R-loop formation and non-canonical ATM activation. ATM signals in a feed-forward fashion to further impede spliceosome organization and regulates UV-induced gene expression and alternative splicing genome-wide. This reciprocal coupling between ATM and the spliceosome highlights the importance of ATM signaling in the cellular response to transcription-blocking lesions and supports a key role of the splicing machinery in this process

    MPLS is an advanced forwarding scheme which allows the network to achieve the benefits provided by traffic engineering techniques. The establishment of an end to end LSP between two IP/MPLS networks inter connected through an ATM backbone is still an open issue. This focuses in an MPLS ATM environment and address problem of provisioning a fast LSP establishment with certain QOS between 2 MPLS sub networks inter connected through an ATM backbone. The PNNI is used in ATM backbone as a routing and signaling protocol. In order to achieve the paper objectives and new PNNI elements are defined and evaluated. Voice has been the primary application in wireless network to date. However packet based application and higher bandwidth requirements to sustain these applications are ever emerging. As in wire line packet based networks while IP routing is well understood IP network by itself is still not achieved and its depart on the backbone that can offer QOS guarantee. MPLS is a newer technology that offer service information layer 2 switching and connection oriented that allows traffic engineering control traffic flows in the network.     Quality of Service over Specific Link Layers: state of the art report

    The Integrated Services concept is proposed as an enhancement to the current Internet architecture, to provide a better Quality of Service (QoS) than that provided by the traditional Best-Effort service. The features of the Integrated Services are explained in this report. To support Integrated Services, certain requirements are posed on the underlying link layer. These requirements are studied by the Integrated Services over Specific Link Layers (ISSLL) IETF working group. The status of this ongoing research is reported in this document. To be more specific, the solutions to provide Integrated Services over ATM, IEEE 802 LAN technologies and low-bitrate links are evaluated in detail. The ISSLL working group has not yet studied the requirements, that are posed on the underlying link layer, when this link layer is wireless. Therefore, this state of the art report is extended with an identification of the requirements that are posed on the underlying wireless link, to provide differentiated Quality of Service

    Induction of Stress Granule Assembly is Essential for the Orchestration of DNA Damage Response

    DNA damage provokes several responses including DNA repair, cell cycle regulation and apoptosis that collectively represent the DNA damage response (DDR). Here, we demonstrate that the DDR incorporates the activation of stress granule (SG) formation pathway as a mechanism to process destabilized RNAs. UV irradiation induced the assembly of SGs during the G2 phase and newly formed SGs appeared exclusively in the early G1 phase. SG assembly pathway was activated within the first hours after DNA damage, suggesting that the processing of destabilized RNAs is activated at an early stage. The induction of SGs and RNAi effector protein Argonaute 2 recruitment after UV exposure was independent of ATM and ATR signaling cascades. Apoptosis occurred only in SG-negative cells indicating that SGs promote cell survival after genotoxic stress. Analysis of several DNA damage/repair deficient MEFs revealed that the SG accumulation remained unaltered after UV exposure. Our results show that SGs are an essential component of DDR that is activated in parallel to the DNA damage kinase response networks

    This thesis examines and simulates a statistically multiplexed multiple access technique known as Advanced Time Division Multiple Access (ATDMA). The simulations were carried out in a multimedia traffic environment. Parameters that could optimise the network performance in terms of quality, reliability and capacity have been examined using a simulation model. This thesis also examines network architecture and signalling related issues. The simulation results were analysed to propose a suitable ATDMA frame structure in terms of the frame length and the organisation of traffic and reservation slots. The simulation results indicated that the performance of the ATDMA based system can be enhanced when delay insensitive data is transmitted as blocks of packets of a specific size. The simulation results also indicated that the performance of the ATDMA based system can be further enhanced when a video terminal is allocated a single traffic slot as opposed to multiple traffic slots. Further simulations have been carried out to determine the up-link traffic channel capacities and control channel capacities. This thesis also examined aspects that could further enhance the performance of an ATDMA based system

    Applications of satellite technology to broadband ISDN networks

    Two satellite architectures for delivering broadband integrated services digital network (B-ISDN) service are evaluated. The first is assumed integral to an existing terrestrial network, and provides complementary services such as interconnects to remote nodes as well as high-rate multicast and broadcast service. The interconnects are at a 155 Mbs rate and are shown as being met with a nonregenerative multibeam satellite having 10-1.5 degree spots. The second satellite architecture focuses on providing private B-ISDN networks as well as acting as a gateway to the public network. This is conceived as being provided by a regenerative multibeam satellite with on-board ATM (asynchronous transfer mode) processing payload. With up to 800 Mbs offered, higher satellite EIRP is required. This is accomplished with 12-0.4 degree hopping beams, covering a total of 110 dwell positions. It is estimated the space segment capital cost for architecture one would be about 190Mwhereasthesecondarchitecturewouldbeabout190M whereas the second architecture would be about 250M. The net user cost is given for a variety of scenarios, but the cost for 155 Mbs services is shown to be about $15-22/minute for 25 percent system utilization
