    Cardiovascular instrumentation for spaceflight

    The observation mechanisms dealing with pressure, flow, morphology, temperature, etc. are discussed. The approach taken in the performance of this study was to (1) review ground and space-flight data on cardiovascular function, including earlier related ground-based and space-flight animal studies, Mercury, Gemini, Apollo, Skylab, and recent bed-rest studies, (2) review cardiovascular measurement parameters required to assess individual performance and physiological alternations during space flight, (3) perform an instrumentation survey including a literature search as well as personal contact with the applicable investigators, (4) assess instrumentation applicability with respect to the established criteria, and (5) recommend future research and development activity. It is concluded that, for the most part, the required instrumentation technology is available but that mission-peculiar criteria will require modifications to adapt the applicable instrumentation to a space-flight configuration

    Development and Applications of Advanced Ultrasound Techniques for Characterization and Stimulation of Engineered Tissues

    Mechanobiology is central in the development, pathology, and regeneration of musculoskeletal tissues, in which mechanical factors play important roles. Therefore, there is a need for methods to characterize the composition and mechanical properties of developing musculoskeletal tissues over time. Ultrasound elastographic techniques have been developed for noninvasive imaging of spatial heterogeneity in tissue stiffness. However, their application for quantitative assessment of tissue mechanical properties, especially viscoelastic properties, has not been exploited. Additionally, ultrasound energy may be used to apply mechanical stimulation to engineered constructs at the microscale, and thereby to enhance tissue regeneration. We have developed a multimode ultrasound viscoelastography (MUVE) system for assessing microscale mechanical properties of engineered hydrogels. MUVE uses focused ultrasound pulses to apply acoustic radiation force (ARF) to deform samples, while concurrently measuring sample dimensions using coaxial high frequency ultrasound imaging. We used MUVE to perform creep tests on agarose, collagen, and fibrin hydrogels of defined concentrations, as well as to monitor the mechanical properties of cell-seeded constructs over time. Local and bulk viscoelastic properties were extracted from strain-time curves through fitting of relevant constitutive models, showing clear differences between concentrations and materials. In particular, we showed that MUVE is capable of mapping heterogeneity of samples in 3D. Using inclusion of dense agarose microbeads within agarose, collagen and fibrin hydrogels, we determined the spatial resolution of MUVE to be approximately 200 μm in both the lateral and axial directions. Comparison of MUVE to nanoindentation and shear rheometry showed that our ultrasound-based technique was superior in generating consistent, microscale data, particularly for very soft materials. We have also adapted MUVE to generate localized cyclic compression, as a means to mechanically stimulate engineered tissue constructs at the microscale. Selected treatment protocols were shown to enhance the osteogenic differentiation of human mesenchymal stem cells in collagen-fibrin hydrogels. Constructs treated at 1 Hz at an acoustic pressure of 0.7 MPa for 30 minutes per day showed accelerated osteogenesis and increased mineralization by 10 to 30 percent, relative to unstimulated controls. In separate experiments, the ultrasound pulse intensity was increased over time to compensate for changes in matrix properties over time, and a 35 percent increase in mineralization was achieved. We also extended the application of a previously-developed spectral ultrasound imaging (SUSI) technique to an animal model for early detection of heterotopic ossification (HO). The quantitative information on acoustic scatterer size and concentration derived from SUSI was used to differentiate tissue composition in a burn/tenotomy mice model from the control model. Importantly, HO foci were detected as early as one week after injury using SUSI, which is 3-5 weeks earlier than when using conventional micro-computed tomography. Taken together, these results demonstrate that ultrasound-based techniques can non-invasively and quantitatively characterize viscoelastic properties of soft materials in 3D, as well as their composition over time. Ultrasound pulses can also be used to stimulate engineered constructs to promote musculoskeletal tissue formation. MUVE, SUSI, and ultrasound stimulation can be combined into an integrated system to investigate the roles of matrix composition, static mechanical environment, and dynamic mechanical stimuli in tissue regeneration, as well as the interactions of these factors and their evolution over time. Ultrasound-based techniques therefore have promising potential in noninvasively characterizing the composition and biomechanics, as well as providing mechanical intervention in native and engineered tissues as they develop over time.PHDBiomedical EngineeringUniversity of Michigan, Horace H.     Quantitative elasticity measurement of urinary bladder wall using laser-induced surface acoustic waves

    The maintenance of urinary bladder elasticity is essential to its functions, including the storage and voiding phases of the micturition cycle. The bladder stiffness can be changed by various pathophysiological conditions. Quantitative measurement of bladder elasticity is an essential step toward understanding various urinary bladder disease processes and improving patient care. As a nondestructive, and noncontact method, laser-induced surface acoustic waves (SAWs) can accurately characterize the elastic properties of different layers of organs such as the urinary bladder. This initial investigation evaluates the feasibility of a noncontact, all-optical method of generating and measuring the elasticity of the urinary bladder. Quantitative elasticity measurements of ex vivo porcine urinary bladder were made using the laser-induced SAW technique. A pulsed laser was used to excite SAWs that propagated on the bladder wall surface. A dedicated phase-sensitive optical coherence tomography (PhS-OCT) system remotely recorded the SAWs, from which the elasticity properties of different layers of the bladder were estimated. During the experiments, series of measurements were performed under five precisely controlled bladder volumes using water to estimate changes in the elasticity in relation to various urinary bladder contents. The results, validated by optical coherence elastography, show that the laser-induced SAW technique combined with PhS-OCT can be a feasible method of quantitative estimation of biomechanical properties

    Design and integration of a parallel, soft robotic end-effector for extracorporeal ultrasound

    Objective: In this work we address limitations in state-of-the-art ultrasound robots by designing and integrating a novel soft robotic system for ultrasound imaging. It employs the inherent qualities of soft fluidic actuators to establish safe, adaptable interaction between ultrasound probe and patient. Methods: We acquire clinical data to determine the movement ranges and force levels required in prenatal foetal ultrasound imaging and design the soft robotic end-effector accordingly. We verify its mechanical characteristics, derive and validate a kinetostatic model and demonstrate controllability and imaging capabilities on an ultrasound phantom. Results: The soft robot exhibits the desired stiffness characteristics and is able to reach 100% of the required workspace when no external force is present, and 95% of the workspace when considering its compliance. The model can accurately predict the end-effector pose with a mean error of 1.18+/-0.29mm in position and 0.92+/-0.47deg in orientation. The derived controller is, with an average position error of 0.39mm, able to track a target pose efficiently without and with externally applied loads. Ultrasound images acquired with the system are of equally good quality compared to a manual sonographer scan. Conclusion: The system is able to withstand loads commonly applied during foetal ultrasound scans and remains controllable with a motion range similar to manual scanning. Significance: The proposed soft robot presents a safe, cost-effective solution to offloading sonographers in day-to-day scanning routines. The design and modelling paradigms are greatly generalizable and particularly suitable for designing soft robots for physical interaction tasks

    MODELLING AND IN VIVO MONITORING OF THE TIME DEPENDENT MECHANICAL PROPERTIES OF TISSUE ENGINEERING SCAFFOLDS

    When organs and tissue fail either due to pre-existing disease progression or by accidental damage, current state of the art treatment involves the replacement of the damaged or diseased tissue with new donor derived organs/tissue. The limitations of these current approaches include a limited supply of tissue for treatments and the immune response of the patient’s own body against the new implanted tissue/organs. To solve these issues, tissue engineering aims to develop artificial analogs derived from a patient’s own cells instead of donor tissue/organs for treatment. To this end, a promising approach, known as scaffold-based tissue engineering, is to seed engineered constructs or scaffolds with cells to form artificial analogs, which then develop with time into new tissue/organs for implantation. The mechanical properties of the scaffold play a critical role in the success of scaffold-based treatments, as the scaffold is expected to provide a temporary support for the generation of new tissue/organs without causing failure at any time during the treatment process. It is noted that due to the degradation of scaffold in the treatment process, the mechanical properties of the scaffold are not constant but change with time dynamically. This raises two scientific issues; one is the representation of the time-dependent mechanical properties and the other one is the monitoring of these properties, especially in the in vivo environments (i.e., upon the implantation of scaffolds into animal/patient bodies). To address these issues, this research is aimed at performing a novel study on the modelling and in vivo monitoring of the time dependent mechanical properties of tissue engineering scaffolds. To represent the time-dependent mechanical properties of a scaffold, a novel model based on the concept of finite element model updating is developed. The model development involves three steps: (1) development of a finite element model for the effective mechanical properties of the scaffold, (2) parametrizing the finite element model by selecting parameters associated with the scaffold microstructure and/or material properties, which vary with scaffold degradation, and (3) identifying selected parameters as functions of time based on measurements from the tests on the scaffold mechanical properties as they degrade. To validate the developed model, scaffolds were made from the biocompatible polymer polycaprolactone (PCL) mixed with hydroxyapatite (HA) nanoparticles and their mechanical properties were examined in terms of the Young modulus. Based on the bulk degradation exhibited by the PCL/HA scaffold, the molecular weight was selected for model updating. With the identified molecular weight, the finite element model v developed was effective for predicting the time-dependent mechanical properties of PCL/HA scaffolds during degradation . To monitor and characterize scaffold mechanical properties in vivo, novel methods based on synchrotron-based phase contrast imaging and finite element modeling were developed. The first method is to represent the scaffold mechanical properties from the measured deflection. In this method, the phase contrast imaging is used to characterize the scaffold deflection caused by ultrasound radiation forces; and the finite element modelling is used to represent the ultrasonic loading on the scaffold, thus predicting the mechanical properties from the measured deflection. The second method is to characterize the scaffold degradation due to surface erosion, which involves the remote sensing of the time dependent morphology of tissue scaffolds by phase contrast imaging and the estimation of time dependent mass loss of the scaffolds from the sensed morphology. The last method is to relate the elastic mechanical property and nonlinear stress-strain behavior to the scaffold geometry, both changing with time during surface erosion. To validate the above methods, scaffolds was made from varying biomaterials (PLGA and PCL) and their mechanical properties (degradation, mass loss, and elastic modulus) were examined experimentally. The results obtained illustrate the methods developed in this research are effective to monitor and characterize scaffold mechanical properties. The significance of this research is that the model developed for the scaffold mechanical properties can be used in the design of scaffolds with the desired mechanical properties, instead of the trial and error methods typical in current scaffold design; and that these novel monitoring methods based on synchrotron imaging can be used to characterize the scaffold time-dependent mechanical properties in the in vivo environments, representing an important advance in tissue engineering
