    Carotenoids - Effective Radical Scavengers for Healthy and Beautiful Skin

    Free radicals are involved in various diseases and skin aging. To reduce and prevent this risk, our body produces antioxidants that can neutralize free radicals. However, some antioxidants need to be taken up with food, so a balanced and varied diet is essential for human health and beauty, along with sufficient exercise. Vegetables, especially curly kale, show very good antioxidative capacity due to the presence of carotenoids. As the recommended daily intake of vegetables is usually not consumed, dietary supplements are a good possibility to ingest carotenoids in a controlled and natural way. The positive effect of carotenoid-based dietary supplements on the skin has already been shown in several studies on healthy volunteers. Innovative non-invasive measuring methods have shown that oil extracts from vegetables significantly reduce not only free radicals in the skin but also the age-related breakdown of collagen and have a positive effect on skin parameters such as wrinkle volume. Thus, a balanced mixture of different natural carotenoids contributes to maintaining health and beauty

    A Bright, Slow Cryogenic Molecular Beam Source for Free Radicals

    We demonstrate and characterize a cryogenic buffer gas-cooled molecular beam source capable of producing bright beams of free radicals and refractory species. Details of the beam properties (brightness, forward velocity distribution, transverse velocity spread, rotational and vibrational temperatures) are measured under varying conditions for the molecular species SrF. Under typical conditions we produce a beam of brightness 1.2 x 10^11 molecules/sr/pulse in the rovibrational ground state, with 140 m/s forward velocity and a rotational temperature of approximately 1 K. This source compares favorably to other methods for producing beams of free radicals and refractory species for many types of experiments. We provide details of construction that may be helpful for others attempting to use this method.Comment: 15 pages, 14 figure

    Role of free radicals has been proposed in the pathogenesis of many diseases. Gastric cancer is a common disease worldwide, and leading cause of cancer death in India. Severe oxidative stress produces reactive oxygen species (ROS) and induces uncontrolled lipid peroxidation. Albumin, uric acid (UA) and Bilirubin are important physiological antioxidants. We aimed to evaluate and assess the role of oxidative stress (OS) and physiological antioxidant system in stomach cancer patients. Lipid peroxidation measured as plasma Thio Barbituric Acid Reactive substances (TBARS), was found to be elevated significantly (p=0.001) in stomach cancer compared to controls along with a decrease in plasma physiological antioxidant system. The documented results were due to increased lipid peroxidation and involvement of physiological antioxidants in scavenging free radicals but not because of impaired hepatic and renal functions

    Assay of the antioxidant capacity of foods using an iron(II)-catalysed lipid peroxidation model for greater nutritional relevance

    The formation of free radicals by the iron-catalysed Fenton reaction is a major cause of oxidative damage in the body. Here a common assay of antioxidant capacity, inhibition of the β-carotene-linoleic acid model of lipid peroxidation, has been modified by the addition of ferrous iron (final concentration 36 μmol/l), which makes the rate of oxidation of the lipids occur twenty-five times faster. Such an assay can simulate the oxidative damage to membrane lipids and low density lipoproteins occurring in the body in the presence of free iron. It thus may be nutritionally more relevant than traditional chemical assays of antioxidant capacity, as it measures pre-emptive antioxidant activity, i.e. activity which prevents free radicals being formed in the first place. Pre-empting their formation is likely to be more protective than scavenging of free radicals. The relative antioxidant activity of some food products found using this new assay was very different from that found using a radical-scavenging assay. Vitamin C, at 280 mg/l, was found to be sixty times better than blackcurrant puree in scavenging free radicals, but only one eighth as good as the blackcurrant puree in preventing iron-catalysed lipid peroxidation

    While free radicals and inflammation constitute major routes of neuronal injury occurring in neurodegenerative diseases, neither antioxidants nor nonsteroidal anti-inflammatory drugs (NSAIDs) have shown significant efficacy in human clinical trials. To explore the possibility that concurrent blockade of free radicals and PGE2-mediated inflammation might constitute a safe and effective therapeutic approach to certain neurodegenerative diseases, we have developed 2-hydroxy-5-[2-(4-trifluoromethylphenyl)-ethylaminobezoic acid (AAD-2004) as a derivative of aspirin. AAD-2004 completely removed free radicals at 50 nM as a potent spin trapping molecule and inhibited microsomal prostaglandin E synthase-1 (mPGES-1) with an IC50 of 230 nM. Oral administration of AAD-2004 blocked free radical formation, PGE2 formation, and microglial activation in the spinal motor neurons of SOD1G93A mice. As a consequence, AAD-2004 reduced autophagosome formation, axonopathy, and motor neuron degeneration, improving motor function and increasing life span. In these assays, AAD-2004 was superior to ibuprofen or riluzole. Gastric bleeding was not induced by AAD-2004 even at a dose 400-fold higher than that required to obtain maximal therapeutic efficacy in SOD1G93A mice. Targeting both mPGES-1 and free radicals may be a promising approach to reduce neurodegeneration in ALS and possibly other neurodegenerative diseases

    Abstract Background: Oxidative stress is responsible for some alterations in the chemical structure and, consequently, in the function of proteins, lipids, and DNA. Recent studies have linked oxidative stress to cancers, particularly thyroid cancer, but the mechanisms remain unclear. Here, we further characterize the role of oxidative stress in thyroid cancer by analyzing the expression of two selenium antioxidant molecules, glutathione peroxidase (GPx1) and thioredoxin reductase (TrxR1) in thyroid cancer cells. Methods: Samples of both healthy thyroid tissue and thyroid tumor were taken for analysis after total thyroidectomy. The expression of GPx1 and TrxR1 was revealed by Western blot analysis and quantified by densitometric analyses, while the evaluation of free radicals was performed by Electron Paramagnetic Resonance (EPR)-spin trapping technique. Results: Our results show a decrease in the expression of GPx1 and TrxR1 (− 45.7 and − 43.2% respectively, p < 0.01) in the thyroid cancer cells compared to the healthy cells. In addition, the EPR technique shows an increase of free radicals in tumor tissue, significantly higher than that found in healthy thyroid tissue (+ 116.3%, p < 0.01). Conclusions: Our findings underscore the relationship between thyroid cancer and oxidative stress, showing the imbalance of the oxidant/antioxidant system in thyroid cancer tissue. These results suggest that either the inability to produce adequate antioxidant defense or an increased consumption of antioxidants, due to the hyper-production of free radicals, may play a crucial role in thyroid cancer.
