    RESUMEN: Durante la expansi贸n y regeneraci贸n de los tejidos en desarrollo se produce una fase de r谩pida proliferaci贸n celular, que debe parar en un momento dado para asegurar la homeostasis del tejido. La epidermis es un epitelio escamoso estratificado, compuesto por c茅lulas en diferenciaci贸n terminal en su capa m谩s externa que deben ser constantemente sustituidas por c茅lulas que emergen de la capa mas interna. Por esta raz贸n, la epidermis se considera como un tejido adulto en desarrollo y sirve de modelo para el estudio de los mecanismos moleculares que regulan el balance entre la proliferaci贸n y la diferenciaci贸n celular. En esta tesis se han estudiado dichos mecanismo, prestando especial atenci贸n al papel desempe帽ado por la regulaci贸n del ciclo celular y la respuesta al da帽o en el ADN. Adem谩s, se han realizado b煤squedas g茅nicas con el objetivo de encontrar nuevas mol茅culas involucradas en la activaci贸n de la diferenciaci贸n escamosa.ABSTRACT: During expansion and regeneration developing tissues undergo a rapid phase of cell proliferation that needs to stop at some to control tissue homeostasis. The epidermis is an squamous stratified epithelia with a high self-renewal capacity. The epidermis is considered as an adult developing tissue and can be use as a model for the study of the molecular mechanism maintaining the balance between proliferation and differentiation. In this Thesis we have studied the role of the cell cycle checkpoints and the DNA damage response in the regulation of these mechanisms.     Ambiguous Medical Image Segmentation using Diffusion Models

    Collective insights from a group of experts have always proven to outperform an individual's best diagnostic for clinical tasks. For the task of medical image segmentation, existing research on AI-based alternatives focuses more on developing models that can imitate the best individual rather than harnessing the power of expert groups. In this paper, we introduce a single diffusion model-based approach that produces multiple plausible outputs by learning a distribution over group insights. Our proposed model generates a distribution of segmentation masks by leveraging the inherent stochastic sampling process of diffusion using only minimal additional learning. We demonstrate on three different medical image modalities- CT, ultrasound, and MRI that our model is capable of producing several possible variants while capturing the frequencies of their occurrences. Comprehensive results show that our proposed approach outperforms existing state-of-the-art ambiguous segmentation networks in terms of accuracy while preserving naturally occurring variation. We also propose a new metric to evaluate the diversity as well as the accuracy of segmentation predictions that aligns with the interest of clinical practice of collective insights

    1. Latar Belakang/Konteks: Sebagai generasi yang dekat dengan sosial media, generasi Z sangat mudah terpengaruh perubahan tren gaya hidup. Sosial media memberikan dampak signifikan pada pengambilan keputusan konsumen sebelum melakukan pembelian skincare, diketahui 81-90 persen manusia menelusuri ulasan online sebelum membeli suatu produk (Erskine, 2017) dan 67,7 persen dari keputusan pembelian dipengaruhi oleh ulasan online (Unni M. , 2020). 2. Permasalahan: Kuatnya pengaruh ulasan online berpotensi menyebabkan ketidaktepatan pemilihan produk yang sesuai dengan kebutuhan masing-masing konsumen karena mayoritas didasarkan hanya pada ulasan influencer atau opini konsumen lain. Secara umum pengguna skincare awam tidak mempertimbangkan keamanan produk dan kredibilitas sumber yang terpercaya. 3. Tujuan: Membangun basis pengetahuan untuk meningkatkan efisiensi bagi pengembang aplikasi pasar dan komunitas pengguna skincare. Fitur pembelajaran pada aplikasi yang nantinya dikembangkan berguna bagi pengguna skincare awam yang membutuhkan pengetahuan terstruktur guna mempercepat proses pembangunan rutin skincare. 4. Data dan Metode: Data yang dikumpulkan didapat melalui video dari dermatologis. Terdapat tujuh channel youtube dermatologis yang digunakan sebagai sumber data. Dermatologis terpilih adalah yang mengaku telah tersertifikasi. Data tersebut diolah menggunakan metode Zettlekasten, yaitu metode pencatatan dan pengembangan pengetahuan secara buttom up menjadi basis pengetahuan. 5. Hasil: Menyediakan basis pengetahuan yang akurat dan terpercaya tentang perawatan kulit, yang dapat membantu dalam membangun rutin. Perbaikan yang perlu dilakukan adalah beberapa sumber data harus terus relevan sehingga basis pengetahuan harus terus diperbarui dan dikelola secara berkala. Basis pengetahuan harus diuji untuk mengukur efektivitas dan ketepatan informasi. ================================================================================================ 1. Problems: Background phenomena have the potential to cause inaccuracies in selecting products that suit the needs of each consumer because the majority are based only on the reviews of influencers or other consumers. 3. Data and Methods: The data collected was obtained via video from a dermatologist. There are seven dermatology youtube channels used as data sources. Selected dermatologists are those who claim to be certified. The data is processed using the Zettlekasten method, which is a bottom-up method of recording and developing knowledge into a knowledge base. 5. Results: Provides accurate and reliable information about skin care, which can help solve skin problems. While the improvements that need to be made are that some data sources must continue to be relevant to the times so that the knowledge base must be updated and managed regularly, the knowledge base must be tested to determine the effectiveness and accuracy of the information

    Cutaneous anaplastic large T-cell lymphoma in a cat - case report

    ABSTRACT Lymphoma is the most common lymphoproliferative disorder in cats. However, cutaneous lymphomas are uncommon in this species and can be classified as epitheliotropic or non-epitheliotropic. Epitheliotropic lymphomas are typically of T-cells and have tropism for epidermis and/or adnexal epithelium. Non-epitheliotropic lymphomas can be of either T-cell or B-cell and involve predominantly the dermis. The present study describes a case of multifocal cutaneous anaplastic large T-cell lymphoma. The skin nodules were multiple and variable in size, alopecic and erythematous in appearance and randomly distributed. Immunohistochemistry revealed positivity for the CD3 antigen, consistent with T-lymphocytes. This neoplasm should be remembered whenever multiple cutaneous nodules are observed in cats

    Gesti贸n ambiental y manejo de residuos s贸lidos en un distrito de la provincia de Luya, Amazonas

    La investigaci贸n tuvo el objetivo de determinar la relaci贸n entre la gesti贸n ambiental y el manejo de residuos de un distrito de la Provincia de Luya, Amazonas. Se realiz贸 una investigaci贸n de tipo b谩sica, de enfoque cuantitativo, con dise帽o no experimental, transversal, correlacional. La poblaci贸n estuvo conformada por 117 familias de la entidad estudiada, de la cual a trav茅s de un muestreo probabil铆stico aleatorio simple se tom贸 una muestra de 90 familias. La t茅cnica de recolecci贸n de datos fue la encuesta y se emple贸 como instrumento dos cuestionarios, los cuales fueron validados por juicio de tres expertos; as铆 mismo, la confiabilidad se obtuvo mediante el coeficiente de Alfa de Cronbach, del cual se obtuvo un valor mayor a 0.9, presentando una confiabilidad alta para los cuestionarios de ambas variables. Se obtuvo un coeficiente de correlaci贸n Rho de Spearman de 0.808** y un valor Sig. de 0.000<0.05, valores que demuestran la relaci贸n existente entre las variables, de manera que se acepta la hip贸tesis alternativa y se rechaza la hip贸tesis nula. Concluyendo que existe una correlaci贸n alta entre la gesti贸n ambiental y el manejo de residuos s贸lidos de un distrito de la Provincia de Luya, Amazonas

    Identifizierung pr盲diktiver und prognostischer Biomarker in unterschiedlichen Tumorkompartimenten des 枚sophagealen Adenokarzinoms

    Das 枚sophageale Adenokarzinom zeigt eine global steigende Inzidenz und hat mit einer 5-Jahres-脺berlebensrate von weniger als 25% eine schlechte Prognose. Personalisierte Therapieans盲tze sind selten und prognostische/pr盲diktive Biomarker des Tumormikromilieus sind unzureichend charakterisiert. Die kumulative Promotion n盲hert sich dieser Problematik in drei unterschiedlichen Schwerpunkten. 1. Zur Identifizierung Kompartiment-spezifischer Biomarker wurde eine Methode entwickelt, welche als kosteng眉nstige Alternative zum sc-Seq Expressionsprofile individueller Zelltypen generiert. Dabei erfolgt die Extraktion der RNA nicht aus Einzelzellen, sondern aus flowzytometrisch-getrennten Zellkompartimenten. Die Separation der Proben in Epithelzellen, Immunzellen und Fibroblasten wurde durch verschiedene Verfahren validiert und eine suffiziente Ausbeute an RNA auch f眉r kleine Gewebemengen gezeigt. 2. Biomarker des Immunzellkompartiments als therapeutische Angriffspunkte wurden in einem Patientenkollektiv von bis zu 551 Patienten auf ihre Bedeutung beim EAC 眉berpr眉ft. Es zeigte sich eine Expression der Immuncheckpoints LAG3, VISTA und IDO auf TILs durch IHC und RNA-Sonden basierte Verfahren in einem relevanten Anteil (LAG3: 11,4%, VISTA: 29%, IDO: 52,6%). Es konnte eine prognostisch g眉nstige Bedeutung der VISTA, LAG3 und IDO Expression gezeigt werden. Durch den Vergleich von Genexpressionsprofilen aus therapienaiven und vorbehandelten Tumoren konnte zudem ein immunsuppressiver Effekt von neoadjuvanten Therapiekonzepten auf das Tumormikromilieu des EACs gezeigt werden. Dabei kam es zur verminderten Expression von Checkpoints und Anzahl TILs nach (Radio-) Chemotherapie. 3. Im Tumorzellkompartiment wurde die Rolle von Amplifikationen in ErbB-Rezeptor abh盲ngigen Signalwegen durch FISH-Technik und Immunhistochemie evaluiert. Es fanden sich KRAS Amplifikationen in 17,1%, PIK3CA Amplifikationen in 5% sowie eine HER2/neu-脺berexpression in 14,9% der untersuchten Tumore
