Canine semen (Canis familiaris) cryopreservation on french straw in Medellín, Antioquia
The objective of this work was to apply the canine semen cryopreservation methodology in French straw in Medellín-Antioquia. A seminal collection was carried out to eight adult canines, by gloved hand method, each ejaculate was assessed macroscopically and microscopically and subsequently diluted with Triladyl® with a final concentration of 80x106 sperm/ml. Semen samples were packed in 0.5 ml straws and a quick-freezing process was performed (nitrogen vapors for 25 minutes and then transferred directly to liquid nitrogen, french straw method). In fresh semen, a milky whitish appearance, a volume of 2.27 ml (± 1.40), a concentration of 388.5x106 sperm/mL (± 228.069), individual motility of 79% (± 4%) and vigor of 3.96 (± 0.327) were observed. 123 straws were obtained, of which 27 were taken randomly and individual motility (51 ± 19%) and vigor (2.89 ± 1.02) were evaluated. In conclusion, the application of the methodology of quick-freezing by french straw, using Triladyl® as a cryoprotectant, is suggested as an efficient protocol for cryopreservation of canine semen in Medellín-Antioquia.
El objetivo de este trabajo fue aplicar la metodología de criopreservación de semen canino en pajilla francesa en el municipio de Medellín- Antioquia. Se realizó una colecta seminal a ocho caninos adultos, por método de mano enguantada, cada eyaculado fue evaluado macroscópica y microscópicamente y posteriormente diluido con Triladyl® a una concentración final de 80 x 106 espermatozoides/ml. Se empacaron en pajillas de 0,5 ml y fueron sometidas a criopreservación con un descenso de temperatura rápido, sometiendo las pajillas a vapores de nitrógeno durante 25 minutos y luego transferidas directamente a nitrógeno líquido (método de pajilla francesa). En el semen fresco, se observó una apariencia blanquecina lechosa, un volumen de 2.27 ml (±1.40) una concentración de 388.5 x106 espermatozoides/ mL (±228.069), una motilidad individual de 79% (± 4%) y un vigor de 3.96 (±0.327). Se obtuvieron 123 pajillas, de las cuales se tomaron 27 aleatoriamente y se les evaluó la motilidad individual (51 ± 19%) y vigor (2.89 ± 1.02). En conclusión, la aplicación de la metodología de criopreservación rápida por pajilla francesa, usando Triladyl® como crioprotector, se sugiere como protocolo eficiente para la criopreservación de semen canino en el municipio de Medellín.

Efecto del Resveratrol sobre la congelación y vitrificación de espermatozoides epididimarios de perro
Esta investigación evaluó el efecto del Resveratrol (RES) en la congelación
convencional (C) y vitrificación (V) de espermatozoides epididimarios caninos sobre la
criosupervivencia espermática. Veinte agrupaciones (pools) de 60 muestras
espermáticas epididimarios de 30 perros orquiectomizados fueron usados en esta
investigación. Cada pool fue diluido con TCG-YH (6% yema de huevo) y entonces 4
tratamientos fueron conformados según el tipo de criopreservación y la adición de 100
µM de RES: 1) C-RES, 2) C-Co[control], 3) V-RES, y 4) V-Co[control]. La congelación
se realizó exponiendo pajuelas (0,25 ml) con las muestras espermáticas (5% glicerol) a
vapores de nitrógeno líquido (NL2) estático. La vitrificación se hizo dejando caer gotas
de 30 μL de muestras (con 250 mM sacarosa) directamente al NL2. El tratamiento CRES incrementó (P<0,05) la motilidad total, velocidad curvilínea (VCL) rápida, la
amplitud del desplazamiento lateral de la cabeza (ALH), la integridad de membranas
plasmática y acrosomal, y redujo el estrés oxidativo. No obstante, el tratamiento V-RES
no provocó ningún efecto benéfico (P>0,05) después del calentamiento. Los
tratamientos de vitrificación incrementaron (P<0,05) la fragmentación de ADN
comparados con los tratamientos de congelación. El tratamiento C-RES incrementó
(P<0,05) las dimensiones de la cabeza de los espermatozoides después de la
congelación. En conclusión, el RES provocó un efecto beneficioso y estimulador de la
locomoción, protector de las membranas y ejerció un efecto antioxidante, durante y
después del proceso de congelación de espermatozoides epididimarios caninos; sin
embargo, su efecto se vio limitado en la vitrificación.This research assessed the effect of Resveratrol (RES) on the conventional slow
freezing (CS) and vitrification (V) of canine epididymal sperm on cryosurvival. Twenty
sperm pooled samples from 60 epididymal from 30 orchiectomized dogs were used in
this study. Each pool was diluted with TCG-YH (6% egg yolk), and then 4 treatments
were conformed based on the type of cryopreservation and the addition of 100 µM RES:
1) CS-RES, 2) CS-Co[control], 3) V-RES, and 4) V-Co[control]. Freezing was performed
by exposing 0.25 ml straws with sperm samples (with 5% glycerol) to static liquid nitrogen
(LN2) vapors. Vitrification was done by directly dropping 30-μL drops of samples (with
250 mM sucrose) into LN2. The CS-RES treatment increased (P<0.05) post-thaw values
of total motility, rapid curvilinear velocity (VCL), and the amplitude of lateral head
displacement (ALH), and integrity of plasma and acrosome membranes, and reduced
oxidative stress. However, the V-RES treatment did not induce any beneficial effect
(P>0.05) after warming. Vitrification treatments increased (P<0.05) DNA fragmentation
compared to freezing treatments. The CS-RES treatment increased (P<0.05) the
dimensions of sperm head after freezing. In conclusion, RES had a beneficial and
enhanced effect on motility, membrane protection, and exerted an antioxidant effect
during and after the freezing of canine epididymal sperm; however, its effect was limited
