Improved in vitro fertilization via zona thinning.
Mouse in vitro fertilization (IVF) can be used to rapidly expand mouse lines from a few males that carry the desired genotype or to maintain strains with poor breeding efficiency. This guide describes a detailed method using acidified Tyrode solution to open the zona pellucida, and increase the fertilization capacity of frozen mouse sperm. The embryos generated are suitable for transfer into pseudopregnant recipent females, culture or cryopreseration
Onco-Huā¢ Models: Humanized NSGā¢ and NSGā¢-SGM3 Mice for Immuno-Oncology
Understanding the interactions between human immune cells and tumors is paramount when devising treatment strategies that prevent tumor evasion of immune cells and improve cytotoxic responses. NSGā¢* and NSGā¢-SGM3* mice are a proven host for engraftment of human tumors or establishment of human immunity following hematopoietic stem cell transplantation. Here, we provide evidence that NSG and NSG-SGM3 mice engrafted with hematopoietic stem cells (collectively referred to here as āhumanized NSG miceā) can support the growth of allogeneic human tumors. The human tumors respond to standard-of-care chemotherapeutics, targeted therapies, and to immune check-point inhibitors clinically proven to initiate cytotoxic activity towards the tumor. Tumor-bearing, humanized NSG mice (the Onco- Huā¢* mouse offered exclusively from The Jackson Laboratory) are a new and valuable preclinical testing platform for immuno-oncology
