Kurzzusammenfassung Ján Cvengroš Metall-vermittelte Strategien zur Totalsynthese der Antibiotika Pestalon und Patulin und struktureller Analoga.   Die vorliegende Arbeit behandelt synthetische Untersuchungen zur Totalsynthese der Naturstoffe Pestalon und Patulin und deren struktureller Analoga mit potentiell interessanten biologischen Aktivitäten. Beide Zielmoleküle verfügen über bemerkenswerte Strukturmotive, die die Entwicklung neuartiger effizienter Synthesemethoden erfordern. Pestalon ist ein natürliches marines Polyketid mit interessanter Struktur und starker antibakterieller Aktivität. Das zentrale Strukturmotiv, ein hochsubstituiertes Benzophenon, das auch in verwandten biologisch aktiven Substanzen vertreten ist, erregte unser Interesse an der Entwicklung einer einfachen und variablen Synthese. Erste Untersuchungen beschäftigten sich mit der Synthese zweier aromatischer Synthesebausteine. Die Darstellung eines „unsymmetrischen“ Orcinol-Bausteins konnte durch Optimierung der Desymmetrisierung von 1,3-Dimethoxyaromaten mittels selektiver Demethylierung unter Verwendung von Lithiumthioethanolat signifikant verbessert werden. Bei der Synthese des zweiten Bausteins wurden verschiedene Strategien zur Einführung von Kohlenstoff-Substituenten (Prenyl-, Formyl-) unter Verwendung Metall-vermittelter Reaktionen untersucht. Dabei wurde eine neuartige Tandem-Reaktion (Allylstannylierung und Heck-Olefinierung) entdeckt, die einen einfachen Zugang zu Indanol-Derivaten eröffnet, ausgehend von ortho-Halobenzaldehyden.  Die Kupplung der beiden Bausteine zum Aufbau des Pestalonsgerüstes und verschiedene Folgetransformationen erwiesen sich als unerwartet schwierig. Deshalb wurde noch eine alternative Strategie zur Synthese von hoch substituierten Benzophenonen durch 4+2-Cycloaddition untersucht. In diesem Zusammenhang wurden neue Zugänge zu den erforderlichen Dien- und Dienophil-Reaktionspartnern realisiert.  Auch wenn die Totalsynthese des Pestalons nicht mehr abgeschlossen werden konnte, so konnten doch effiziente neue Wege zu hoch substituierten Arenen erschlossen und verschiedene Pestalonanaloga für biologische Untersuchungen bereitgestellt werden. In einem zweiten Teil der Dissertation wurde eine neue Strategie  zur Synthese des antibakteriellen Mykotoxins Patulin ausgelotet. Die avisierte Synthese basiert auf einer Pd-katalysierten Kaskadenreaktion zum Aufbau des hoch funktionalisierten Bicyclus und einer anschliessenden photochemischen Gerüstumlagerung. Diese Untersuchungen führten zur Entwicklung einer neuartigen Synthese von γ-Ylidenbutenoliden. Abstract Ján Cvengroš Metal-based strategies for the total synthesis of the antibiotics pestalone and patulin and structural analogues  The presented work deals with synthetic studies towards the total synthesis of the natural products pestalone and patulin. Both molecules exhibit attractive biological activities and also possess striking structural features, which call for novel and efficient synthetic methodologies. Pestalone represents a natural marine polyketide having a potent antibacterial activity and an interesting architecture. The main structural motif, a highly substituted benzophenone skeleton also found in other biologically active compounds, challenges the development of efficient and flexible synthetic strategies for its assembly. Our first generation approach dismantled the pestalone molecule into two suitable aromatic building blocks, the preparation of which concerned the majority of this work. The synthesis of an “unsymmetrical” orcinol derivative was significantly improved through desymmetrization of a 1,3-dimethoxyarene in a highly efficient manner using lithium thioethanolate as an O-demethylating agent. In the course of the synthesis of the second building block several methods were evaluated for the introduction of carbon-substituents (namely prenyl and formyl groups) using metal-mediated reactions. This lead to the discovery of a novel and unique tandem process (allylstanylation/Heck reaction) which opens access to indanol derivatives in a very efficient manner starting from ortho-halo-benzaldehydes. The key coupling of the two arene building blocks and subsequent transformations were extensively studied but turned out to be unexpectedly difficult. Therefore, an alternative strategy for the construction of the highly substituted benzophenone moiety through [4+2]-cycloaddition was investigated. In this context new synthetic routes to the required Diels-Alder reaction partners (dienes and dienophiles) were delineated.  Despite the fact that the total synthesis of pestalone itself could not be completed, efficient new synthetic entries to highly substituted arenes were developed and interesting structural analogs were made available for the biological investigations. In a second part of the thesis, a new strategy for the synthesis of the antibacterial mycotoxin patulin was examined. The envisioned synthesis is based on a Pd-catalyzed cascade reaction for the construction of the densely functionalized bicyclic system followed by a photochemical rearrangement. These investigations led to the discovery and development of a novel and facile route towards γ-ylidenebutenolides.
