Aims: To investigate the effect of YKL-40 RNA interference on VEGF gene polymorphism expression in atherosclerotic mice. Methods: After the atherosclerosis models in mice were built, the mice were divided into three groups including control group, negative control group and observation group, which were separately given to normal saline, negative virus (5 × 107 TU) and YKL-40 RNA interference lentivirus. Then the whole blood DNA was extracted and genotyped in each group of mice and the expression of VEGF in each group of mice was detected by PCR, while the expression level of inflammatory factors in each group of mice was detected by ELISA. Meanwhile, the aortas of mice in each group were pathologically analyzed and the atherosclerosis of mice was detected. Results: Compared with the control group, the VEGF content in both the virus negative control group and the observation group was significantly increased(P<0.05). The detection rates of CC genotype and C allele at rs699947 of VEGF gene in the observation group were significantly higher than those in the control group and the virus negative control group, and the difference was statistically significant. There were no significant changes for the expression of HDL-C, LDL-C, TC and TG in mice of each group(P>0.05). Moreover, the levels of Lp-PLA2 and MCP-1 in the negative control group were significantly increased (P < 0.05), while those in the observation group were significantly decreased (P < 0.05) compared to that in control group. What’s more, the histomorphology of the observation group was significantly different from that of the control group and the virus negative control group. The thickness of the fibrous cap of the as plaque was significantly higher than that of the control group and the virus negative control group, but the plaque area and fat content were significantly lower than that of the control group and the virus negative control group and the NC group. Besides, there was no significant difference in lipid content, fiber cap thickness and plaque area between the control group and the virus negative control group. Conclusion: YKL-40 RNAi could improve the VEGF polymorphism, reduce the expression of LP⁃PLA2 and MCP⁃1, and significantly inhibit the occurrence and development of atherosclerosis, which was expected to provide a new target for the prevention and treatment of atherosclerosis

Advances in the ADAMTS Family in Cardiovascular Disease

    Get PDF
Cardiovascular disease is a serious threat to human life and health. The number of people who die from cardiovascular disease is up to 15 million every year, ranking the first cause of all causes of death. ADAMTS family (A Disintegrin and Metalloproteinase With Thrombospondin Motifs, ADAMTSs) are matrix-associated zinc metallopeptidases with secretory function. It has diverse roles in tissue morphogenesis, pathophysiological remodeling, inflammation, and vascular biology. Controlling the structure and function of the Extracellular Matrix (ECM) is the central theme of the biology of ADAMTSs. ADAMTSs mainly play a biological role by regulating the structure and function of extracellular mechanisms, and the abnormal expression or dysfunction of some family members is associated with cardiovascular diseases. ADAMTS family plays an important role in the occurrence and development of various cardiovascular diseases. This paper aims to study the role of ADAMTS family in cardiovascular diseases

Objective: Informing patient consultations and healthcare choices, clinical predictive models can offer patients tailored projections of the outcome. The most frequent elbow fractures in children are supracondylar humerus fractures, and clinical prediction models were still largely underutilized in these cases. By developing and verifying a prediction model to lower the risk of postoperative problems in children with supracondylar humerus fractures, this research sought to evaluate independent risk variables connected with the incidence of complications of supracondylar humerus fractures in children. Methods: We retrospectively studied 411 children with supracondylar humerus fractures treated surgically at our hospital from 2015 to 2019, and explored the independent risk factors affecting the prognosis of supracondylar humerus fractures in children in the study group using univariate and multifactorial Cox regression analysis, respectively. In addition, a prediction model based on the independent factors was constructed, a nomogram was made and data from the two cohorts were used to verify the feasibility and reliability of the model and visualize the data. Results: Height, older than eight years, weight, nerve damage, fracture type and with joystick technology of the child as independent risk factors influenced the prognosis of pediatric supracondylar humerus fractures in the modeling constructed by the training cohort, respectively. The results of the validation cohort were further screened for older than eight years, nerve injury and fracture type as independent prognostic factors. Conclusions: We were able to construct a predictive model based on a large genuine data sample, and clinical characteristics in this model could be used as independent predictors for reducing the occurrence of postoperative complications in supracondylar fractures. Combining basic vital signs and clinical risk factors into a simple and clear nomogram was more likely to result in the best treatment plan
