    Comparison of Two Year Classes of Hybrid Grass Carp and Grass Carp for Aquatic Plant Control

    Two year classes of grass carp and F, hybrids resulting from bighead carp male x grass carp female were compared at various stocking densities for aquatic plant control. One and two year old grass carp exhibited higher survival rates and better growth rates than the same age hybrid grass carp. The presence of grass carp or hybrid grass carp decreased both Secchi disc transparencies and dissolved oxygen values. Grass carp had a greater negative effect upon these measurements because they removed the vegetation quicker than the hybrid grass carp. These apparent detrimental effects on water quality are necessary trade-offs for vegetation removal by any method. Grass carp and hybrid grass carp utilized Chara sp., Potamogeton pectinatus, Hydrodicton sp., Rhizoclonia sp., and Pithophora sp. Two year old hybrid grass carp required approximately twice as much time as the same age grass carp to eliminate dense growths of the vegetation listed above. One year old hybrid grass carp were slightly less effective than one year old grass carp at controlling these same plant species. However, it was extremely difficult for one or two year old hybrid grass carp to totally eliminate dense growths of these plant species except at high stocking densities. The use of mean vegetation heights to indirectly measure total plant biomass was unacceptable whenever unpreferred floating plant species were present. The hybrid grass carp appeared to be a poor alternative biological control for nuisance aquatic vegetation when compared directly to the grass carp

    A novel lncRNA as a positive regulator of carotenoid biosynthesis in Fusarium

    The fungi Fusarium oxysporum and Fusarium fujikuroi produce carotenoids, lipophilic terpenoid pigments of biotechnological interest, with xanthophyll neurosporaxanthin as the main end product. Their carotenoid biosynthesis is activated by light and negatively regulated by the RING-finger protein CarS. Global transcriptomic analysis identified in both species a putative 1-kb lncRNA that we call carP, referred to as Fo-carP and Ff-carP in each species, upstream to the gene carS and transcribed from the same DNA strand. Fo-carP and Ff-carP are poorly transcribed, but their RNA levels increase in carS mutants. The deletion of Fo-carP or Ff-carP in the respective species results in albino phenotypes, with strong reductions in mRNA levels of structural genes for carotenoid biosynthesis and higher mRNA content of the carS gene, which could explain the low accumulation of carotenoids. Upon alignment, Fo-carP and Ff-carP show 75-80% identity, with short insertions or deletions resulting in a lack of coincident ORFs. Moreover, none of the ORFs found in their sequences have indications of possible coding functions.     Age composition, growth, and reproduction of koi carp (Cyprinus carpio L.) in the lower Waikato, New Zealand

    A total of 566 koi carp (Cyprinus carpio) from the lower Waikato region were aged from scales and opercular bones, and growth was modelled with the von Bertalanffy growth function. There was no difference in growth rate between male and female carp. Growth of koi carp between zero and 3 years of age was lower than that of common carp in Europe and Australia. However, after 5 years of age the growth of koi carp was higher than that of common carp in Europe, but still below that of carp in Australia. Males rarely lived in excess of 8 years, whereas females lived to 12 years. Mean total fecundity calculated from 44 running-ripe females was 299 000 oocytes (±195 600 SD) (range 29 800–771 000). Relative fecundity ranged from 19 300 to 216 000 oocytes kg–1 total body weight, with a mean of 97 200 (±35 000 SD) oocytes kg–1. Feral koi carp in the Waikato are capable of multiple spawnings within their lifetimes. Within a spawning season, Waikato populations of feral koi carp contained females that spawned once, and females that had the potential to have spawned repeatedly.     China has a very rich genetic diversity in common carp (Cyprinus carpio) and the red common carp plays an important role in Chinese aquaculture and genetic studies. Selective breeding, particularly crossbreeding has been applied successfully to red common carps in China, and the products of these efforts have been in commercial use since the 1970s. However, knowledge of the quantitative and molecular genetics of these carps is limited. Studies were therefore undertaken to: (1) understand the genetic diversity and genetic relationship of red common carps in China; (2) understand the inheritance of color phenotype of Oujiang color carp; (3) select stable Oujiang color carp with fast growth rate and ornamental Oujiang color carp comparable with the Koi common carp from Japan; (4) study the culture performance and culture systems suitable for the Oujiang color carp in cages and paddies; (5) extend better quality fish and appropriate culture systems for small scale fish farmers in poor areas

    Common carp (Cyprinus carpio L.) alters its feeding niche in response to changing food resources: direct observations in simulated ponds

    We used customized fish tanks as model fish ponds to observe grazing, swimming, and conspecific social behavior of common carp (Cyprinus carpio) under variable food-resource conditions to assess alterations in feeding niche. Different food and feeding situations were created by using only pond water or pond water plus pond bottom sediment or pond water plus pond bottom sediment and artificial feeding. All tanks were fertilized twice, prior to stocking and 2 weeks later after starting the experiment to stimulate natural food production. Common carp preferred artificial feed over benthic macroinvertebrates, followed by zooplankton. Common carp did not prefer any group of phytoplankton in any treatment. Common carp was mainly benthic in habitat choice, feeding on benthic macroinvertebrates when only plankton and benthic macroinvertebrates were available in the system. In the absence of benthic macroinvertebrates, their feeding niche shifted from near the bottom of the tanks to the water column where they spent 85% of the total time and fed principally on zooplankton. Common carp readily switched to artificial feed when available, which led to better growth. Common carp preferred to graze individually. Behavioral observations of common carp in tanks yielded new information that assists our understanding of their ecological niche. This knowledge could be potentially used to further the development of common carp aquaculture

    Genomic organization of nucleolin gene in carp fish: Evidence for several genes

Here we report that at least three different genes codify for this protein in carp fish (Cyprinus carpio). This is the first description of the genomic organization of nucleolin in a teleost. The carp nucleolin gene includes 8.8 kb and contains 16 exons. Promoter cis regulatory elements are similar to constitutive genes, i.e., a putative TATA box, three G/C boxes, and three pyrimidine-rich boxes. As in other species, carp nucleolin gene introns host three snoRNA codifying sequences: U23 from the H/ACA family and two C/D box snoRNAs, U20 and U82. Both U20 and U82 span a complementary sequence with carp 18S rRNA. Additionally, we identified two cDNAs coding for nucleolin, confirming the existence of several nucleolin genes in carp. Amino acidderived sequence from carp cDNAs differ from mammal protein because they span additional acidic domains at the amino end, whose functional significance remains unclear. We performed amino acid sequence comparison and phylogenetic analyses showing that the three isoforms of carp nucleolin, which we describe herein, cluster in two groups. cNUC1 probably diverges from cNUC2 and cNUC3 as result of ancestral fish-specific genome duplication, indeed C. carpio is a tetraploid fish

    Management of broodstock and quality control of fish seed in Hungary

    Common carp (Cyprinus carpio) breeding has a long tradition in Hungary. However, recent economic changes in Eastern Europe and new developments in aquaculture necessitated the need for ensuring quality of the brood stock used in hatcheries and the legal and institutional frameworks needed to implement the program. In addition to good research and development programs and gene banking, it became essential to establish an appropriate legal framework, organize, coordinate and control breeding activities, and provide financial support. It was a major breakthrough for carp breeding when C.carpio was recognized as one of the cultivated animals in the Animal Breeding Act in 1993. The Carp Breeding Section of the Hungarian Fish Producers Association plays an important role in carp breeding programs. Thirteen breeding farms of the Carp Breeding Section have 24 certified C.carpio varieties. In Hungary, about 80 % of the seed used as stocking for commercial production are from high quality certified breeders

    Evaluation of macrophyte control in 38 Florida lakes using triploid grass carp

    Florida’s large number of shallow lakes, warm climate and long growing season have contributed to the development of excessive growths of aquatic macrophytes that have seriously interfered with many water use activities. The introduction of exotic aquatic macrophyte species such as hydrilla ( Hydrilla verticillata ) have added significantly to aquatic plant problems in Florida lakes. The use of grass carp ( Ctenopharyngodon idella ) can be an effective and economical control for aquatic vegetation such as hydrilla. Early stocking rates (24 to 74 grass carp per hectare of lake area) resulted in grass carp consumption rates that vastly exceeded the growth rates of the aquatic plants and often resulted in the total loss of all submersed vegetation. This study looked at 38 Florida lakes that had been stocked with grass carp for 3 to 10 years with stocking rates ranging from < 1 to 59 grass carp per hectare of lake and 1 to 207 grass carp per hectare of vegetation to determine the long term effects of grass carp on aquatic macrophyte communities. The median PAC (percent area coverage) value of aquatic macrophytes for the study lakes after they were stocked with grass carp was 14% and the median PVI (percent volume infested) value of aquatic macrophytes was 2%. Only lakes stocked with less than 25 to 30 fish per hectare of vegetation tended to have higher than median PAC and PVI values. When grass carp are stocked at levels of > 25 to 30 fish per hectare of vegetation the complete control of aquatic vegetation can be achieved, with the exception of a few species of plants that grass carp have extreme difficulty consuming. If the management goal for a lake is to control some of the problem aquatic plants while maintaining a small population of predominately unpalatable aquatic plants, grass carp can be stocked at approximately 25 to 30 fish per hectare of vegetation

    Anticarbamylated protein antibodies are associated with long-term disability and increased disease activity in patients with early inflammatory arthritis:Results from the Norfolk Arthritis Register

    Objectives: Anticarbamylated protein (anti-CarP) antibodies are a novel family of autoantibodies recently identified in patients with inflammatory arthritis. The aim of this study was to investigate their association with long-term outcomes of disability and disease activity over 20 years’ follow-up in a cohort of patients with inflammatory polyarthritis (IP).  Methods: Norfolk Arthritis Register recruited adults with recent-onset swelling of ≥2 joints for ≥4 weeks from 1990 to 2009. At baseline, Health Assessment Questionnaire (HAQ) and 28 joint disease activity scores (DAS28) were obtained, and C reactive protein, rheumatoid factor (RF), anticitrullinated protein antibodies (ACPA) and anti-CarP antibodies were measured. Further HAQ scores and DAS28 were obtained at regular intervals over 20 years. Generalised estimating equations were used to test the association between anti-CarP antibody status and longitudinal HAQ and DAS28 scores; adjusting for age, gender, smoking status, year of inclusion and ACPA status. Analyses were repeated in subgroups stratified by ACPA status. The relative association of RF, ACPA and anti-CarP antibodies with HAQ and DAS28 scores was investigated using a random effects model.  Results: 1995 patients were included; 1310 (66%) were female. Anti-CarP antibodies were significantly associated with more disability and higher disease activity, HAQ multivariate β-coefficient (95% CI) 0.12 (0.02 to 0.21), and these associations remained significant in the ACPA-negative subgroups. The associations of RF, ACPA and anti-CarP antibodies were found to be additive in the random effects model.  Conclusions: Anti-CarP antibodies are associated with increased disability and higher disease activity in patients with IP. Our results suggest that measurement of anti-CarP antibodies may be useful in identifying ACPA-negative patients with worse long-term outcomes. Further, anti-CarP antibody status provided additional information about RF and ACPA

    Control of macrophytes by grass carp (ctenopharyngodon idella) in a Waikato drain, New Zealand

    Hornwort (Ceratophyllum demersum L.) and other aquatic macrophytes have historically been mechanically removed from the Rangiriri drain and Churchill East drain to maintain drain efficiency. As an alternative control method for the high plant biomass that accumulates at the end of summer, the effect of stocking diploid grass carp (Ctenopharyngodon idella L.) on the aquatic vegetation was evaluated in these Waikato drainage systems. At the start of the trial, both drains had a low diversity of aquatic macrophytes, and of the nine species (including the emergents), seven were exotic. Two months after grass carp were released to Churchill East drain (the treated drain) the four submerged and floating macrophyte species became scarce in the main drain. Over the same period, these species increased in biomass in Rangiriri drain (the untreated drain), where hornwort became dense and surface-reaching and remained so for the duration of the trial. However, grass carp did not control submerged vegetation in smaller side drains or the shallow, upper parts of the main drain, or the marginal sprawling species and emergent species. The cost of leasing the grass carp was similar to the cost of clearing the drains mechanically, but grass carp provided continuous weed control. However, subsequent to this trial, 62 dead grass carp were found in Churchill East drain in February 2001, and weed cover subsequently increased. This illustrates that grass carp management in New Zealand agricultural drains can be problematic due to periodic fish kills
