    Comparison of Liquid Phase Ion Speciation in DEAB-CO2-H2O System with IPAB-CO2-H2O System Using 13C NMR Techniques

    AbstractThe 13C NMR techniques were selected to investigate the ion speciation in both DEAB and IPAB solvents. Calibration equations were established to quantify the exact ion concentration of free amines and protonated amines at different absorption times. The results show that the NMR method can be used to obtain both qualitative and quantitative information of the amine-CO2-H2O system. It is also the first time to monitor the components in the new solvent (IPAB) and establish its VLE model

    Non-wet kingfisher flying in the rain: The tumble of droplets on moving oriented anisotropic superhydrophobic substrates

    Extensive studies of antiwetting have been restricted to stationary substrates, while dewetting mechanisms on moving interfaces are still poorly understood. Due to the hydrophobic and anisotropic surface characteristics of kingfishers, they are able to easily change flight direction even under high-intensity precipitation. The present study aims to mechanistically analyze how the synergy of interfacial movement, anisotropy, and superhydrophobicity affects rapid dehydration. We have designed a droplet-conveyor system to simulate the bouncing of droplets on moving anisotropic superhydrophobic targets and performed simulations via the lattice Boltzmann algorithm. The moving interface can induce a directional tumbling behavior of the droplet and effectively avoid continuous wetting in the same region. We found that droplet tumbling is essentially caused by transformed depinning velocity vectors at the interface downstream. Also, the hang time of a tumbling droplet is positively related to the angle between the motion vector and the texture. The oriented anisotropic motion facilitates the tumbling of droplets and decreases their hang time by up to 23% as compared to that on a stationary inclined superhydrophobic surface. Similar interfacial process dehydration also occurs on a nonwet kingfisher flying in the rain, and we believe that these findings provide valuable new insights for high-efficiency water repellency of surface

    Community-based HIV-1 Early Diagnosis and Risk Behavior Analysis of Men Having Sex with Men in Hong Kong

    The increasing prevalence of HIV-1 among men having sex with men (MSM) calls for an investigation of HIV-1 prevalence and incidence in MSM by early diagnosis to assist with early preventive interventions in Hong Kong. The participants were recruited randomly from MSM communities within a one-year period. Rapid HIV Test (RHT) and real-time dried blood spot (DBS)-based quantitative polymerase chain reaction (DBS-qPCR) were used for the early diagnosis of 474 participants. Risk behavior analysis was performed by studying information obtained from the participants during the study period. The HIV-1 prevalence and incident rates in the studied MSM population were 4.01%(19/474) and 1.47% (7/474), respectively. Three infected participants were found at the acute phase of infection by DBS-qPCR. Only 46.4%(220/474) MSM were using condoms regularly for anal sex. HIV infection significantly correlated with unprotected receptive anal sex and syphilis infection. An increased number of infections was found among foreign MSM in Hong Kong. This study is the first to use DBS-qPCR to identify acutely infected individuals in a community setting and to provide both the prevalence and incident rates of HIV-1 infection among MSM in Hong Kong. The risk analysis provided evidence that behavior intervention strengthening is necessary to fight against the increasing HIV-1 epidemic among MSM in Hong Kong and surrounding regions in Asia.published_or_final_versio

    Genomic Analyses Reveal Mutational Signatures and Frequently Altered Genes in Esophageal Squamous Cell Carcinoma

    Esophageal squamous cell carcinoma (ESCC) is one of the most common cancers worldwide and the fourth most lethal cancer in China. However, although genomic studies have identified some mutations associated with ESCC, we know little of the mutational processes responsible. To identify genome-wide mutational signatures, we performed either whole-genome sequencing (WGS) or whole-exome sequencing (WES) on 104 ESCC individuals and combined our data with those of 88 previously reported samples. An APOBEC-mediated mutational signature in 47% of 192 tumors suggests that APOBEC-catalyzed deamination provides a source of DNA damage in ESCC. Moreover, PIK3CA hotspot mutations (c.1624G>A [p.Glu542Lys] and c.1633G>A [p.Glu545Lys]) were enriched in APOBEC-signature tumors, and no smoking-associated signature was observed in ESCC. In the samples analyzed by WGS, we identified focal (<100 kb) amplifications of CBX4 and CBX8. In our combined cohort, we identified frequent inactivating mutations in AJUBA, ZNF750, and PTCH1 and the chromatin-remodeling genes CREBBP and BAP1, in addition to known mutations. Functional analyses suggest roles for several genes (CBX4, CBX8, AJUBA, and ZNF750) in ESCC. Notably, high activity of hedgehog signaling and the PI3K pathway in approximately 60% of 104 ESCC tumors indicates that therapies targeting these pathways might be particularly promising strategies for ESCC. Collectively, our data provide comprehensive insights into the mutational signatures of ESCC and identify markers for early diagnosis and potential therapeutic targets

    Quantitative evaluation of the immunodeficiency of a mouse strain by tumor engraftments

Background: The mouse is an organism that is widely used as a mammalian model for studying human physiology or disease, and the development of immunodeficient mice has provided a valuable tool for basic and applied human disease research. Following the development of large-scale mouse knockout programs and genome-editing tools, it has become increasingly efficient to generate genetically modified mouse strains with immunodeficiency. However, due to the lack of a standardized system for evaluating the immuno-capacity that prevents tumor progression in mice, an objective choice of the appropriate immunodeficient mouse strains to be used for tumor engrafting experiments is difficult. Methods: In this study, we developed a tumor engraftment index (TEI) to quantify the immunodeficiency response to hematologic malignant cells and solid tumor cells of six immunodeficient mouse strains and C57BL/6 wild-type mouse (WT). Results: Mice with a more severely impaired immune system attained a higher TEI score. We then validated that the NOD-scid-IL2Rg-/- (NSI) mice, which had the highest TEI score, were more suitable for xenograft and allograft experiments using multiple functional assays.
