    Post-translational modifications (PTMs) of proteins are crucial to guarantee the proper biological functions in immune responses. Although protein phosphorylation has been extensively studied, our current knowledge of protein pyrophosphorylation, which occurs based on phosphorylation, is very limited. Protein pyrophosphorylation is originally considered to be a non-enzymatic process, and its function in immune signaling is unknown. Here we identify a metabolic enzyme, UDP-N-acetylglucosamine pyrophosphorylase 1 (UAP1) as a pyrophosphorylase for protein serine pyrophosphorylation, by catalyzing the pyrophosphorylation of interferon regulatory factor 3 (IRF3) at serine (Ser) 386 to promote robust type I interferon (IFN) responses. Uap1 deficiency significantly impairs the activation of both DNA and RNA viruses-induced type I IFN pathways, and the Uap1 deficient mice are highly susceptible to lethal viral infection. Our findings demonstrate protein pyrophosphorylation as a novel type of PTM in the regulation of antiviral responses, and provide insights into the crosstalk between metabolism and innate immunity.THIS DATASET IS ARCHIVED AT DANS/EASY, BUT NOT ACCESSIBLE HERE.     This is raw data of appropriate depth for harvesting lincluding yield, shoot and root biomass, root system traits in different diameter. The field experiments were conducted in Kaiyuan City (103o15’E, 23o42’ N, 1117m A) of Yunnan Province, in the south-west of China. The field soil soil pH was 7.5 (tested by potentiometry); The organic matter was tested by the potassium dichromate method; Soil nutrients were tested by flme atomic absorption spectrophotometry; and soil mechanical composition was tested by sedimentation method. The experiment was rainfed, and the groundwater level is approximately 2 m below the ground surface. The climate in this region is high temperature during summer, with long winter and early spring severe drought periods. The highest values of precipitation and temperature occur in June and July. The experiment was conducted from 2016 to 2018 on sugarcane cultivar YZ081609 (Zhao et al., 2019). The plant crop was planted on 22 April 2016 with a conventional planting density of 120,000 buds ha-1 (double buds cane setts). The planting depth (cane setts bed to soil surface) was approximately 10 cm, with about 10 ~ 15 cm of earth up before grand growth. So, after plant crop was harvested, there was a soil layer of 20 ~ 25 cm upper the original cane setts. Fertilizer N P K (20-12-18) was applied during the early elongation period of sugarcane at the rate of 1200 kg ha -1 on March 10, 2017 and 2018. Cultivation and crop management procedures were otherwise consistent with conventional cropping practices. Based on the density of underground buds on ratoon stool and their germination potential, we divided the underground buds into 3 types.Type 1 is the terminal bud in top soil, they were lowest in amount and germinate the fastest, the roots distributed mainly in upper soil; type 2 distributed at the depth of 5 ~ 10 cm, most of them are active bud that germinate fast, and their roots distributed deeper; type 3 are mostly dormant bud distributed bellow 10 cm with the poorest germination. Accordingly, the harvesting depths (treatment) were -0 cm, -5 cm and -10 cm for T1, T2 and T3. For each harvesting, sugarcane plants were cut down by a sharp hoe manually. In order to maintain the accuracy of harvesting depth, a half side of the soil surrounding the stool were removed by the relative depth of each treatment before conducting the harvest. Plant crop was harvested on 9 March 2017, first ratoon crop was harvested on 4 March 2018 when the sucrose content reached to top (matured). The plot size was 30 m2 (5 rows wide Γ— 6 m long). Three treatments were randomized in each of the 3 replications. The data of each plant came from the data of the cluster dividing the millable number of the cluster

    Low temperatures affect plant growth, development, productivity, and ecological distribution. Expression of the C-repeat-binding factor (CBF) transcription factors is induced by cold stress, which in turn activates downstream cold-responsive (COR) genes that are required for the acquisition of freezing tolerance. Inducer of CBF expression 1 (ICE1) is a master regulator of CBFs, and ICE1 stability is crucial for its function. However, the regulation of ICE1 is not well understood. Here, we report that mitogen-activated protein kinase 3 (MPK3) and MPK6 interact with and phosphorylate ICE1, which reduces its stability and transcriptional activity. Consistently, the mpk3 and mpk6 single mutants and the mpk3 mpk6 double mutant show enhanced freezing tolerance, whereas MPK3/MPK6 activation attenuates freezing tolerance. Phosphor-inactive mutations of ICE1 complement freezing sensitivity in the ice1-2 mutant. These combined results indicate that MPK3/MPK6 phosphorylate and destabilize ICE1, which negatively regulates CBF expression and freezing tolerance in plants

    A hybrid MPI-CUDA approach for nonequispaced discrete Fourier transformation

    Nonequispaced discrete Fourier transformation (NDFT) is widely applied in all aspects of computational science and engineering. The computational efficiency and accuracy of NDFT has always been a critical issue in hindering its comprehensive applications both in intensive and in extensive aspects of scientific computing. In our previous work Yang et al. (2018), a CUNFFT method was proposed and it shown outstanding performance in handling NDFT at intermediate scale based on CUDA (Compute Unified Device Architecture) technology. In the current work, we further improved the computational efficiency of the CUNTTF method using an efficient MPI-CUDA hybrid parallelization (HP) scheme of NFFT to achieve a cutting-edge treatment of NDFT at super extended scale. Within this HP-NFFT method, the spatial domain of NDFT is decomposed into several parts according to the accumulative feature of NDFT and the detailed number of CPU and GPU nodes. These decomposed NDFT subcells are independently calculated on different CPU nodes using a MPI process-level parallelization mode, and on different GPU nodes using a CUDA thread-level parallelization mode and CUNFFT algorithm. A massive benchmarking of the HP-NFFT method indicates that this method exhibit a dramatic improvement in computational efficiency for handling NDFT at super extended scale without loss of computational precision. Furthermore, the HP-NFFT method is validated via the calculation of Madelung constant of fluorite crystal structure, and thereafter verified that this method is robust for the calculation of electrostatic interactions between charged ions in molecular dynamics simulation systems
