    Massive Spread of OXA-48 Carbapenemase-Producing Enterobacteriaceae in the Environment of a Swiss Companion Animal Clinic.

    BACKGROUND Companion animal clinics contribute to the spread of antimicrobial resistant microorganisms (ARM) and outbreaks with ARM of public health concern have been described. METHODS As part of a project to assess infection prevention and control (IPC) standards in companion animal clinics in Switzerland, a total of 200 swabs from surfaces and 20 hand swabs from employees were collected during four days in a medium-sized clinic and analyzed for extended spectrum beta-lactamase-producing Enterobacteriaceae (ESBL-E), carbapenemase-producing Enterobacteriaceae (CPE), vancomycin-resistant enterococci (VRE), and methicillin-resistant staphylococci (MRS). RESULTS A total of 22 (11.0%) environmental specimen yielded CPE, 14 (7.0%) ESBL-E, and 7 (3.5%) MRS; MR Staphylococcus aureus were isolated from two (10.0%) hand swabs. The CPE isolates comprised Escherichia coli, Klebsiella pneumoniae, Enterobacter hormaechei, Citrobacter braakii, and Serratia marcescens. Whole genome sequencing revealed that all CPE carried closely related blaOXA-48 plasmids, suggesting a plasmidic spread within the clinic. The clinic exhibited major deficits in surface disinfection, hand hygiene infrastructure, and hand hygiene compliance. CPE were present in various areas, including those without patient contact. The study documented plasmidic dissemination of blaOXA-48 in a companion animal clinic with low IPC standards. This poses a worrisome threat to public health and highlights the need to foster IPC standards in veterinary clinics to prevent the spread of ARM into the community

    Educational intervention to improve infection prevention and control practices in four companion animal clinics in Switzerland

    Background: Infection prevention and control (IPC) practices vary among companion animal clinics, and outbreaks with carbapenemase-producing Enterobacterales (CPE) have been described. Aim: To investigate the effect of an IPC intervention (introduction of IPC protocols, IPC lectures, hand hygiene campaign) in four companion animal clinics. Methods: IPC practices, environmental and hand contamination with antimicrobial-resistant micro-organisms (ARM) and hand hygiene (HH) were assessed at baseline, and 1 and 5 months after the intervention. Results: Median IPC scores (% maximum score) improved from 57.8% (range 48.0-59.8%) to 82.9% (range 81.4-86.3%) at 1-month follow-up. Median cleaning frequency assessed by fluorescent tagging increased from 16.7% (range 8.9-18.9%) to 30.6% (range 27.8-52.2%) at 1-month follow-up and 32.8% (range 32.2-33.3%) at 5-month follow-up. ARM contamination was low in three clinics at baseline and undetectable after the intervention. One clinic showed extensive contamination with ARM including CPE before and after the intervention (7.5-16.0% ARM-positive samples and 5.0-11.5% CPE-positive samples). Mean HH compliance improved from 20.9% [95% confidence interval (CI) 19.2-22.8%] to 42.5% (95% CI 40.4-44.7%) at 1-month follow-up and 38.7% (95% CI 35.7-41.7%) at 5-month follow-up. Compliance was lowest in the pre-operative preparation area at baseline (11.8%, 95% CI 9.3-14.8%) and in the intensive care unit after the intervention (28.8%, 95% CI 23.3-35.1%). HH compliance was similar in veterinarians (21.5%, 95% CI 19.0-24.3%) and nurses (20.2%, 95% CI 17.9-22.7%) at baseline, but was higher in veterinarians (46.0%, 95% CI 42.9-49.1%) than nurses (39.0%, 95% CI 36.0-42.1%) at 1-month follow-up. Conclusion: The IPC intervention improved IPC scores, cleaning frequency and HH compliance in all clinics. Adapted approaches may be needed in outbreak situations

    Protective Immunity against Infection with <i>Mycoplasma haemofelis</i>

    Hemoplasmas are potentially zoonotic mycoplasmal pathogens, which are not consistently cleared by antibiotic therapy. Mycoplasma haemofelis is the most pathogenic feline hemoplasma species. The aim of this study was to determine how cats previously infected with M. haemofelis that had recovered reacted when rechallenged with M. haemofelis and to characterize the immune response following de novo M. haemofelis infection and rechallenge. Five specific-pathogen-free (SPF)-derived naive cats (group A) and five cats that had recovered from M. haemofelis infection (group B) were inoculated subcutaneously with M. haemofelis. Blood M. haemofelis loads were measured by quantitative PCR (qPCR), antibody response to heat shock protein 70 (DnaK) by enzyme-linked immunosorbent assay (ELISA), blood lymphocyte cell subtypes by flow cytometry, and cytokine mRNA levels by quantitative reverse transcriptase PCR. Group A cats all became infected with high bacterial loads and seroconverted, while group B cats were protected from reinfection, thus providing the unique opportunity to study the immunological parameters associated with this protective immune response against M. haemofelis. First, a strong humoral response to DnaK was only observed in group A, demonstrating that an antibody response to DnaK is not important for protective immunity. Second, proinflammatory cytokine interleukin-6 (IL-6) mRNA levels appeared to increase rapidly postinoculation in group B, indicating a possible role in protective immunity. Third, an increase in IL-12p35 and -p40 mRNA and decrease in the Th2/Th1 ratio observed in group A suggest that a Th1-type response is important in primary infection. This is the first study to demonstrate protective immunity against M. haemofelis reinfection, and it provides important information for potential future hemoplasma vaccine design

    Simulations and plans for possible DLA experiments at SINBAD

    In this work we present the outlines of possible experiments for dielectric laser acceleration (DLA) of ultra-short relativistic electron bunches produced by the ARES linac, currently under construction at the SINBAD facility (DESY Hamburg). The experiments are to be performed as part of the Accelerator on a Chip International Program (ACHIP), funded by the Gordon and Betty Moore Foundation. At SINBAD we plan to test the acceleration of already pre-accelerated relativistic electron bunches in laser-illuminated dielectric grating structures. We present outlines of both the acceleration of ultra-short single bunches, as well as the option to accelerate phase-synchronous sub-fs microbunch trains. Here the electron bunch is conditioned prior to the injection by interaction with an external laser field in an undulator. This generates a sinusoidal energy modulation that is transformed into periodic microbunches in a subsequent chicane. The phase synchronization is achieved by driving both the modulation process and the DLA with the same laser pulse. In addition to the conceptual layouts and plans of the experiments we present start-to-end simulation results for different ARES working points.Comment: EAAC'17 conference proceeding

    Green Facades – How they Matter for Working Environments, Public Spaces and the Livability of a City

    Sustainable urban development is the focus of many research initiatives, especially due to increasing urbanization and climate change. Buildings and their renovation are central to the European "Green Deal". In the new Austrian climate and energy program, the topic of buildings comes first with a target renovation rate of 3%. Current climate change adaptation strategies call for an increase in greening of existing buildings and on facades. Public spaces are shaped by the surrounding buildings. The facades and roofs of these buildings can have a high potential to mitigate urban heat island effects. Social change and innovation in working cultures result in reshaping working environments and the need for public space. Large-scale glass buildings are widely considered architectural highlights, but pose problematic challenges to urban spaces. Glass has a significant impact on the microclimate inside the building and in the immediate outdoor environment: a concentration of radiant energy and high indoor temperatures put a strain on the energy balance and the well-being of the occupants. The retrofit greening of glass facades is a gap in building expertise and there is a lack of standard applications for the retrofit shading and insulation of glass buildings to obtain associated microclimatic benefits. The project GLASGrĂźn aims to develop, implement, test and monitor modular vertical greening standards for active external shading by deciduous plants on commercial buildings with large glazed facades. Transferable modular-based designs are to be developed. Additionally, sociological surveys on acceptance and perception will be implemented. GLASGrĂźn generates quantitative data on energy, temperature and microclimate balance as well as qualitative data on the perception of the building situation before and after greening interventions and on public awareness. New findings on the acceptance and well-being of employees and customers, on purchasing behavior and market-economic parameters will be available. GLASGrĂźn is developing guidelines for constructive solutions, submission processes and care and maintenance management plans for the systems under consideration and for the vertical green standards tested, which are scalable and transferable and form an economic basis for future adaptations of further buildings as well as for their maintenance. A socio-ecological transformation faces the challenge of how integrated solutions can be developed in dialogue with the users and to what extent these produce the desired effects such as greenhouse gas reduction or better indoor climate. On the other hand, the best solutions in the technical sense can also fail due to social barriers: the acceptance of decision-makers, a lack of willingness to cooperate on the part of employees, or a loss of image in the neighborhood, to name just a few examples. Acceptance depends amongst other factors on both the concrete technical implementation and the process of introduction. Thus, acceptance is not a static variable, but is in a relationship with the technical solution options themselves. Public spaces are key to the discussion on sustainable urban development in their function against urban heat islands. Their diversity of uses and users allows for both a broad discussion and start of discourses and the testing of innovative sustainable measures, in this case greening of facades on buildings perceived in public space. In this paper we will present 2 case studies in Austria with the first results of interviews with employees and users of glass facade buildings and the users of public space

    Embryonic Synthesis of the Inner Limiting Membrane and Vitreous Body

    PURPOSE. The inner limiting membrane (ILM) and the vitreous body (VB) are major parts of the extracellular matrix of the eye. The present study was undertaken to investigate the synthesis and turnover of the ILM and VB in chick and human embryonic and postembryonic eye development. METHODS. The abundance of ILM and VB proteins was determined by Western blot analysis using samples from chick and human VB of different ages. The mRNA expression of the ILM proteins in lens was determined by in situ hybridization and RT-PCR. RESULTS. Based on the abundance of mRNA expression, the prominent sources of ILM and VB proteins in chick eyes are the lens and ciliary body. In chick, ILM and VB matrix proteins were most abundant in embryonic VB, and their concentration declined precipitously after hatching. Most ILM and VB proteins were no longer detectable in the adult VB. In humans, a similar developmentally regulated expression of ILM and VB proteins in VB was detected: The highest concentrations of ILM and VB proteins were detected in fetal VB, the lowest in the adult VB. The decline in ILM and VB protein synthesis occurred within the first 2 years of life. CONCLUSIONS. The abundance of ILM and VB proteins in the embryonic VB, their sharp decline at postembryonic stages, and their very low abundance in the adult VB show that ILM and VB are assembled during embryogenesis and are maintained throughout life with minimum turnover

    Chronic "Candidatus Mycoplasma turicensis" infection

    "Candidatus Mycoplasma turicensis" infects felids. The pathogenesis of "Candidatus M. turicensis" chronic infection is poorly understood. The goals of the present study were to (1) induce reactivation of the infection in chronic carrier cats by attempted immunosuppression, (2) identify potential tissue sequestration using real-time TaqManÂŽ PCR and (3) monitor the humoral immune response by DnaK enzyme-linked immunosorbent assay (ELISA). Ten specified pathogen-free cats that had ostensibly recovered from experimental "Candidatus M. turicensis" infection were used: five cats (group 1) received high dose methylprednisolone (attempted immunosuppression), while five cats served as untreated controls (group 2). Besides weekly blood samples, tissue samples were collected from bone marrow, kidney, liver and salivary glands at selected time points. The cats in group 1 had significantly lower lymphocyte counts and higher blood glucose levels after methylprednisolone administration than the controls. After methylprednisolone administration one blood and three tissue samples from cats in group 1 tested PCR-positive; before the administration, only one sample was positive. All other samples tested PCR-negative. All cats stayed seropositive; the antibody levels of the cats in group 1 showed a significant transient decrease after methylprednisolone administration. This is the first study to report the presence of "Candidatus M. turicensis" in tissues of chronically infected cats and the persistence of anti-feline hemoplasma antibodies in the absence of detectable bacteremia. Methylprednisolone administration did not lead to a significant reactivation of the infection. Our results enhance the knowledge of "Candidatus M. turicensis" infection pathogenesis and are clinically relevant to the prognosis of hemoplasma-infected cats

    Horizontal gene transfer contributed to the evolution of extracellular surface structures

    The single-cell layered ectoderm of the fresh water polyp Hydra fulfills the function of an epidermis by protecting the animals from the surrounding medium. Its outer surface is covered by a fibrous structure termed the cuticle layer, with similarity to the extracellular surface coats of mammalian epithelia. In this paper we have identified molecular components of the cuticle. We show that its outermost layer contains glycoproteins and glycosaminoglycans and we have identified chondroitin and chondroitin-6-sulfate chains. In a search for proteins that could be involved in organising this structure we found PPOD proteins and several members of a protein family containing only SWT (sweet tooth) domains. Structural analyses indicate that PPODs consist of two tandem β-trefoil domains with similarity to carbohydrate-binding sites found in lectins. Experimental evidence confirmed that PPODs can bind sulfated glycans and are secreted into the cuticle layer from granules localized under the apical surface of the ectodermal epithelial cells. PPODs are taxon-specific proteins which appear to have entered the Hydra genome by horizontal gene transfer from bacteria. Their acquisition at the time Hydra evolved from a marine ancestor may have been critical for the transition to the freshwater environment
