    Family mealtimes and adolescent psychopathology

    This thesis examines aspects of adolescent family mealtimes and psychopathology among both non-clinical (adolescents and mothers of adolescents) and clinical (adolescents with anorexia nervosa (AN)) samples. It contains seven studies employing quantitative methodology, which address three broad aims. First, to examine relationships between aspects of family mealtimes and psychopathology among adolescents. Second, to examine links between family mealtime emotions and psychopathology among mothers of adolescents. Finally, to examine associations between specific parental mealtime interactions and adolescent outcomes during a therapeutic meal session for adolescents with AN. Self-report questionnaires were completed by non-clinical adolescents and mothers of adolescents to examine associations between characteristics of family mealtimes (mealtime environment, mealtime emotions and parental feeding practices) and eating psychopathology, anxiety and depression. In addition, observational analyses were conducted on recordings of the family meal session (session two) of Family-Based Treatment (FBT) for adolescent AN. [Continues.

    Family mealtimes and eating psychopathology : the role of anxiety and depression among adolescent girls and boys

    Characteristics of family mealtimes are associated with disordered eating behaviours. However, little is known about the relationships between characteristics of family mealtimes and disordered eating attitudes, or how symptoms of anxiety or depression may contribute to these relationships. This study therefore aimed to examine differences between adolescent girls and boys in the relationship between family mealtime characteristics and eating psychopathology, and to explore the influence of anxiety and depression on this relationship. Adolescents (N=535; 286 girls and 249 boys) aged 14-18 years completed self-report measures of family mealtime characteristics, eating psychopathology, anxiety and depression. Reports of more frequent family mealtimes, a more positive mealtime atmosphere and a high level of priority placed on mealtimes were all associated with significantly lower levels of eating-disordered attitudes among girls only. For boys, all four mealtime measures (higher mealtime frequency, more positive mealtime atmosphere, greater priority of mealtimes and higher levels of mealtime structure) were associated with lower levels of depression. Among girls, several of the family mealtime and eating psychopathology relationships were partially or fully mediated by either anxiety or depression. While these findings require longitudinal replication, family mealtimes are likely to be important in promoting psychological well-being among both girls and boys. Families should be encouraged to think beyond the frequency of mealtimes and to foster a positive mealtime environment which may help to promote adolescent psychological well being, and might even protect young females against the development of eating psychopathology

    The global impact of management on soil nematode abundances

Management effects on soil nematode abundance differ among functional groups and land-use types at a global scale. Journal of Animal Ecology, https://doi.org/10.1111/1365-2656.13744. Despite the well-documented decline of aboveground species abundances as a result of land-use intensification, there has been little attention on the effects of human activities on belowground species abundances. Li et al. analyse nematode data, the most abundant animal on the planet, from across the globe to determine whether their abundances vary between managed and unmanaged habitats. The authors show that, unlike aboveground biodiversity, nematode abundance is higher in managed than unmanaged primary and secondary habitats. Furthermore, responses to land management vary between trophic groups and they do not appear to follow the general hypothesis that higher trophic levels are more vulnerable to human activity than those further down the food chain, except in urban habitats. Finally, Li et al. show that the relationships between environmental predictors and species abundance were weakened (and sometimes reversed) in managed habitats. Together, their results reveal how land-use management is impacting the trophic composition of soil nematode communities and their relationships with the environment, which has implications for ecosystem functioning

    Eating disorder examination questionnaire : factor structure for adolescent girls and boys

    Objective: To examine the factor structure of the EDE-Q among a sample of adolescents. Method: A community-based sample of 917 adolescents (522 girls and 395 boys) aged 14-18 years completed the EDE-Q version 6.0 as part of a larger study. Participants were randomly assigned to one of two subsamples to enable separate analyses. Results: A confirmatory factor analysis on the original four factor model of the EDE-Q produced an inadmissible model with a poor fit. Exploratory factor analysis using principal axis factoring produced an alternative three factor model of the EDE-Q among adolescents. The Shape and Weight Concerns, Restriction and Preoccupation and Eating Concern subscales accounted for 65% of the total variance. Subscale and global scores were significantly higher for girls than for boys. A high proportion of both girls (53.6%) and boys (30.5%) reported participating in at least one key eating disordered behaviour during the previous 28 days. Discussion: The results of this study present three new subscales (Shape and Weight Concerns, Restriction and Preoccupation and Eating Concern) which are suggested for use in future research which uses the EDE-Q with community samples of adolescents

    Initial ecological recovery post‐weir removal amidst catchment‐wide improvements, in a groundwater‐dominated chalk stream

    Physical habitat modification is one of the main pressures affecting river environments, impacting their ecosystem health and compromising their ability to adapt to the effects of climate change. Addressing the impacts of physical modification through reinvigorating natural processes has become a globally established river restoration technique. Here, we appraised such an approach by assessing ecological responses to a weir removal project on an English groundwater‐dominated ‘chalk’ stream. Using a Before‐After‐Control‐Impact (BACI) approach, we found that 3 years post‐restoration the macroinvertebrate communities are moving towards those of the target community both in terms of structural complexity (e.g., taxonomic composition) and functional integrity (e.g., trait composition). The progress is ongoing and has occurred alongside wider catchment improvements. Our results indicate that ecological responses to passive restoration undertaken on low energy streams, such as chalk streams, may be gradual, and thus longer‐term assessment is needed to fully appraise ecological recovery. We highlight the importance of a BACI approach to understand the local responses to restoration in a catchment context. Our findings also provide further evidence highlighting complementary ecological information provided by assessing taxonomic and functional properties concurrently in post‐project appraisals. A better understanding of ecological recovery times should be incorporated into future restoration planning. Such evidence would help develop robust assessments over appropriate timescales, increasing the likelihood of accurately and effectively appraising restoration project success, and helping to build support to increase the scale and pace of restoration actions needed to address biodiversity loss

    NuSTAR hard X-ray observation of a sub-A class solar flare

    We report a NuSTAR observation of a solar microflare, SOL2015-09-01T04. Although it was too faint to be observed by the GOES X-ray Sensor, we estimate the event to be an A0.1 class flare in brightness. This microflare, with only 5 counts per second per detector observed by RHESSI, is fainter than any hard X-ray (HXR) flare in the existing literature. The microflare occurred during a solar pointing by the highly sensitive NuSTAR astrophysical observatory, which used its direct focusing optics to produce detailed HXR microflare spectra and images. The microflare exhibits HXR properties commonly observed in larger flares, including a fast rise and more gradual decay, earlier peak time with higher energy, spatial dimensions similar to the RHESSI microflares, and a high-energy excess beyond an isothermal spectral component during the impulsive phase. The microflare is small in emission measure, temperature, and energy, though not in physical size; observations are consistent with an origin via the interaction of at least two magnetic loops. We estimate the increase in thermal energy at the time of the microflare to be 2.4x10^27 ergs. The observation suggests that flares do indeed scale down to extremely small energies and retain what we customarily think of as "flarelike" properties.Comment: Status: Accepted by the Astrophysical Journal, 2017 July 1

    Microflare Heating of a Solar Active Region Observed with NuSTAR, Hinode/XRT, and SDO/AIA

    NuSTAR is a highly sensitive focusing hard X-ray (HXR) telescope and has observed several small microflares in its initial solar pointings. In this paper, we present the first joint observation of a microflare with NuSTAR and Hinode/XRT on 2015 April 29 at ~11:29 UT. This microflare shows heating of material to several million Kelvin, observed in Soft X-rays (SXRs) with Hinode/XRT, and was faintly visible in Extreme Ultraviolet (EUV) with SDO/AIA. For three of the four NuSTAR observations of this region (pre-, decay, and post phases) the spectrum is well fitted by a single thermal model of 3.2-3.5 MK, but the spectrum during the impulsive phase shows additional emission up to 10 MK, emission equivalent to A0.1 GOES class. We recover the differential emission measure (DEM) using SDO/AIA, Hinode/XRT, and NuSTAR, giving unprecedented coverage in temperature. We find the pre-flare DEM peaks at ~3 MK and falls off sharply by 5 MK; but during the microflare's impulsive phase the emission above 3 MK is brighter and extends to 10 MK, giving a heating rate of about 2.5×10252.5 \times 10^{25} erg s1^{-1}. As the NuSTAR spectrum is purely thermal we determined upper-limits on the possible non-thermal bremsstrahlung emission. We find that for the accelerated electrons to be the source of the heating requires a power-law spectrum of δ7\delta \ge 7 with a low energy cut-off Ec7E_{c} \lesssim 7 keV.     Further Evidence of Early Development of Attention to Dynamic Facial Emotions: Reply to Grossmann and Jessen

    Adults exhibit enhanced attention to negative emotions like fear, which is thought to be an adaptive reaction to emotional information. Previous research, mostly conducted with static faces, suggests that infants exhibit an attentional bias toward fearful faces only at around 7 months of age. In a recent study (Journal of Experimental Child Psychology, 2016, Vol. 147, pp. 100–110), we found that 5-month-olds also exhibit heightened attention to fear when tested with dynamic face videos. This indication of an earlier development of an attention bias to fear raises questions about developmental mechanisms that have been proposed to underlie this function. However, Grossmann and Jessen (Journal of Experimental Child Psychology, 2016, Vol. 153, pp. 149–154) argued that this result may have been due to differences in the amount of movement in the videos rather than a response toemotional information. To examine this possibility, we tested a new sample of 5-month-olds exactly as in the original study (Heck, Hock, White, Jubran, & Bhatt, 2016) but with inverted faces. We found that the fear bias seen in our study was no longer apparent with inverted faces. Therefore, it is likely that infants’ enhanced attention to fear in our study was indeed a response to emotions rather than a reaction to arbitrary low-level stimulus features. This finding indicates enhanced attention to fear at 5 months and underscores the need to find mechanisms that engender the development of emotion knowledge early in life

    Visual Scanning of Males and Females in Infancy

    This study addressed the development of attention to information that is socially relevant to adults by examining infants\u27 (n = 64) scanning patterns of male and female bodies. Infants exhibited systematic attention to regions associated with sex-related scanning by adults, with 3.5-and 6.5-month-olds looking longer at the torso of females than males and longer at the legs of males than females. However, this pattern of looking was not found when infants were tested on headless bodies in Experiment 2, which suggests that infants\u27 differential gaze pattern in Experiment 1 was not due to low-level stimulus features, such as clothing, and also indicates that facial/head information is necessary for infants to exhibit sex-specific scanning. We discuss implications for models of face and body knowledge development

    CRISPR-Mediated Protein Tagging with Nanoluciferase to Investigate Native Chemokine Receptor Function and Conformational Changes

    © 2020 The Authors G protein-coupled receptors are a major class of membrane receptors that mediate physiological and pathophysiological cellular signaling. Many aspects of receptor activation and signaling can be investigated using genetically encoded luminescent fusion proteins. However, the use of these biosensors in live cell systems requires the exogenous expression of the tagged protein of interest. To maintain the normal cellular context here we use CRISPR/Cas9-mediated homology-directed repair to insert luminescent tags into the endogenous genome. Using NanoLuc and bioluminescence resonance energy transfer we demonstrate fluorescent ligand binding at genome-edited chemokine receptors. We also demonstrate that split-NanoLuc complementation can be used to investigate conformational changes and internalization of CXCR4 and that recruitment of β-arrestin2 to CXCR4 can be monitored when both proteins are natively expressed. These results show that genetically encoded luminescent biosensors can be used to investigate numerous aspects of receptor function at native expression levels
