A36 The TP53 mutations in the Russian patients with de novo DLBCL

BackgroundTP53 dysfunction is implicated in lymphomagenesis and disease progression. Information about the frequency and spectrum of TP53 mutations in the Russian pathients with diffuse large B-cell lymphoma (DLBCL) in the current version of the IARC TP53 Mutation Database R17 is not represented. The goal of this work was to study the frequency, spectrum and functional significance of TP53 mutations in Russian patients with DLBCL.Material and methodsAt the present time the pilot group of 14 patients were included in the study. Diagnosis was assessed according to the criteria of the WHO classification system. Genomic DNA was isolated from formalin-fixed, paraffin embedded tissue blocks. Direct sequence analysis of gene TP53 was performed according to the IARC protocol, 2010 update.ResultsIn two patients were identified single nucleotide substitutions that are not described in the current version of the PubMed database. All of mutations occurred in the DNA-binding domain of p53. The nonsense mutation Arg196Ter was detected in one patient. Previously it was shown that formation of this premature stop codon might activate the nonsense-mediated RNA decay pathway. The second patient had two missense mutations – Leu130Phe and Arg156Cys. The first of them leads to p53 inactivation according to the analysis of the functional importance of amino acid substitutions using service PolyPhen-2.ConclusionWe detected TP53 mutation in 14% cases. The mutational rate in our study is in good agreement with other studies where the frequency of the TP53 mutations in patients with DLBCL ranged mostly from 13% to 23%

Citoprotection of endometrium in women with syndrome of loss of pregnancy of early terms by infectionally-inflammatory genesis: lectin-histochemical research

Aim of this reaserch is to give structural characteristic of mucoprotective apparatus of endometrium at female which were suffering from the loss of pregnancy early term with infection-inflammation genesis. Lectinohistochemical research of aspiration biopsies of endometrium from 84 patients in age 18-35 years which were suffering from the loss of pregnancy early term with infection-inflammation genesis was perfomed. Control is biopsies of endometrium from 15 somatically and gynecologically healthy women of the reproductive age, at wich research and had established selective lectine markers of some components of mucociliary apparatus of normal endometrium. All females of basic group have structural change of endometrium, wich characteristic for chronic endometritis. At the loss of pregnancy associated with chronic endometritis are noted increase level of connecting fucomucins and decrease level of sialic acid (depression of bactericidal property of uterine mucus). This can be promote stagnation of mucus in cavity of uteri, persistence of microorganisms, cause development of inflammatory, dystrophy and disregeneratory processes in endometrium, and also have significance in formation local immunopathologic reaction, wich complicate process of implantation of fetal ovum in endometrium.Цель исследования — дать структурную характеристику мукопротективного аппарата слизистой оболочки матки у женщин с невынашиванием беременности ранних сроков инфекционно- воспалительного генеза. Проведено лектиногистохимическое исследование аспирационных биоптатов эндометрия от 84 пациенток в возрасте 18-35 лет с невынашиванием беременности (НБ) ранних сроков инфекционно-воспалительного генеза. Контроль — биоптаты эндометрия от 15 соматически и гинекологически здоровых женщин репродуктивного возраста, при исследовании которых установлены селективные лектиновые маркеры некоторых компонентов мукоцилиарного аппарата неизмененного эндометрия. У всех женщин основной группы имелись структурные изменения слизистой оболочки матки, характерные для хронического эндометрита (ХЭ). При НБ, ассоциированном с ХЭ, отмечено повышение уровня связывания фукомуцинов и падение уровня сиаловых кислот (угнетение бактерицидных свойств маточной слизи). Это может способствовать застою слизи в полости матки, персистенции микроорганизмов, обусловливать развитие воспалительных, дистрофических и дисрегенераторных процессов в эндометрии, а также иметь значение в формировании местных иммунопатологических реакций, затрудняющих процесс имплантации плодного яйца в эндометрий

Spontaneous abortion in aspect of the doctrine about pathomorphosis

It is carried out the analysis of case records of 342 women with the sponta-neous abortion treated in 1996-1998 and 871 women with the spontaneous abortion, treated in 2006-2008. It is established, that at the present stage at everyone the fourth women, suffering from losing of the pregnancy, the first pregnancy comes to an end spontaneous abortion. There is a tendency to reduction of frequency habitual losing of the pregnancy. The share of not developing pregnancy in structure of spontaneous abortion has considerably increased and has made 42,3 %. At everyone the second patient spontaneous abortion occurs on term of pregnancy of 5-8 weeks, at everyone the tenth patient has late spontaneous premature baby. In the mechanism of spontaneous interruption of pregnancy at the present stage prevails infectionally-inflammatory genesis. Probably, it is connected to increase of frequency of chronic infectionally-inflammatory diseases in genital organs.Проведен анализ историй болезни 342 женщин с самопроизвольным абортом, лечившихся в 1996-1998 г.г., и 871 пациентки, лечившейся в 2006-2008 г.г. Установлено, что на современном этапе у каждой четвертой пациентки, страдающей невынашиванием беременности, первая беременность заканчивается самопроизвольным абортом. Имеется тенденция к уменьшению частоты привычного невынашивания беременности. Значительно возросла доля неразвивающейся беременности в структуре самопроизвольного аборта и составила 42,3%. У каждой второй пациентки самопроизвольный аборт происходит на сроке беременности 5-8 недель, у каждой десятой наблюдаются поздние самопроизвольные выкидыши. В механизме самопроизвольного прерывания беременности на современном этапе преобладает инфекционно-воспалительный генез, что, возможно, связано с увеличением частоты хронических инфекционно-воспалительных заболеваний гениталий

Parasitofauna of pike Esox lucius of the Lower Tobol (Russia)

One of the predators of the ichthyofauna of the Lower Tobol is Esox lucius (Linnaeus, 1758) (Esociformes, Esocidae). The purpose of this work is to study the current state of the E. lucius parasite fauna in the lower reaches of the Tobol River. In the present study 47 individuals of the northern pike of different sex aged from 2+ to 8+ years were examined by the method of complete parasitological dissection. As a result of the research, 23 types of parasites were found in the pike. The largest number of species of parasitic organisms – 20 – were found in May, 12 were found in December and 11 in September. Of these, 10 species are specific to northern pike: Haemogregarina esoci (Nawrotzky, 1914), Chloromyxum esocinum (Dogiel, 1934), Myxidium lieberkuhni (Biitschli, 1882), Myxosoma anurum (Cohn, 1895), Henneguya psorospermica (Thelohan, 1995), Tetraonchus monenteron (Wagener, 1857), Gyrodactylus lucii (Kulakowskaja, 1951), Triaenophorus nodulosus (Pallas, 1781), Azygia lucii (Miiller, 1776), Raphidascaris acus (Bloch, 1779). For the first time an intraerythrocyte parasite – H. esoci – was found in pike in the Lower Tobol, the extensiveness of invasion was 18.7% in December, and 6.2% in May. Epistylis sp. and Trichodinella epizzotica parasitized on the surface of the body of fish only in May, whereas members of the genus Trichodina were found in winter and spring. In all seasons, parasitization by M. anurum and H. psorospermica was established on the gills. The greatest occurrence of spores of M. lieberkuhni and C. esocinum was observed in the spring. Myxosporidia parasitized in the kidneys and T. monenteron parasitized on the gills during all periods of the study (the latter with an extensivity of invasion of 100.0%). All examined fish are infected with T. nodulosus 100.0% of cases. In May, other types of cestodes were found: P. esocis and Dibothriocephalus latus. Three pike were infected with proteocephalis. The larval stage of diphyllobothriid was found in one fish in the liver and gonads in an amount of 139 individuals. In the pikes’ stomach, during all periods of the study, A. lucii was found in all the fish examined. In the autumn period of the study immature R. campanula were found in in the intestine of 4 fish specimens. The nematode R. acus was found in pike only in spring. This nematode was found in fish aged 4+–8+. In May, glochidia with a high intensity of invasion were found on fins, gill covers and gills of fish. E. sieboldi copepods were found on the gills; in May, one specimen was found in one pike and in September in 6 with AI of 0.7. Tetraonchus monenteron was a dominant species, in May its degree of dominance decreased with the greatest uniformity of species in this study period. Having considered the age dynamics of pike infection by various types of parasites, it was found that in the age group 4+–5+ the number of species of parasites was greatest (20), while in groups 2+–3+ and 6+–8+ it was 14. In the pike, the core parasitic fauna were M. anurum, H. psorospermica, T. monenteron, T. nodulosus, A. lucii. Seasonality has virtually no effect on the degree of infection with specific parasites. The age of fish largely determines the qualitative and quantitative composition of the parasite fauna

Free radical processes in patients suffering from noncarrying of pregnancy

There have been studied lipid peroxidation and blood serum protein carbonylization results in 30 patients with noncanying of pregnancy and in 30 women with normal reproductive function. In patients with noncarrying pregnancy it was established peroxidation of polar lipids and proteins functioning in water phase activation. At the same time non-polar lipids extracted by heptane in fact do not subject to free-radical impairment. It makes expedient to use antioxidants acting in water phase in course of female rehabilitation after spontaneous miscarriage.Изучены показатели перекисного окисления липидов и карбонилирования белков сыворотки крови у 30 пациенток с невынашиванием беременности и 30 женщин с ненарушенной репродуктивной функцией. У пациенток с невынашиванием беременности установлена активация перекисного окисления полярных липидов и белков, функционирующих в водной фазе. В то же время неполярные липиды, экстрагируемые гептаном, фактически не подвергаются свободнорадикальному повреждению. Это обусловливает целесообразность применения антиоксидантов, действующих в водной фазе, в программе реабилитации женщин после самопроизвольного выкидыша

The practice of using the project method in online foreign language lessons and its role in the formation of critical thinking of adolescents

Целью данного исследования является анализ и обобщение практического опыта применения метода проектов при обучении подростков иностранному языку (английский) и результаты наблюдения за уровнем развития их критического мышления посредством организации проектной деятельности. Приведенные результаты наблюдения показывают, что проектная деятельность является своеобразным катализатором развития данного мягкого навыка у подростков, благоприятно влияет на мотивацию при изучении иностранного языка и рекомендован авторами в качестве одного из перспективных инструментов организации обучения.The purpose of this study is to analyze and summarize the practical experience of using the project method in teaching teenagers a foreign language (English) and the results of monitoring the level of development of their critical thinking through the organization of project activities. The above observation results show that project activity is a kind of catalyst for the development of this soft skill in adolescents, has a positive effect on motivation when learning a foreign language and is recommended by the authors as one of the promising tools for organizing learning

Morphofunctional characteristic of remodeling of endometrium on the syndrome of loss of pregnancy by infectionally-inflammatory genesis

It was carried out clinico-laboratory inspection of 30 women, which were suffering from the loss of pregnancy and from chronic endometritis. It was spent immunihistochemical research of endometrium biopsies with the help monoclonic antibodies to gelatinase (ММР-2, MMP-9) and their inhibitor T1MP-1 for an estimation of intensity and an orientation of remodeling processes in endometrium. As the control served biopsies, which were taken from a mucous membrane of body in uterus from 15 somatically and gynecologically healthy women of the reproductive age who has addressed for inspection before planning of pregnancy or contraception. Results of this experience show that the main features are reduction of MMP-9 and MMP-2 intensity expression and also increasing of values T1MP-1 as original «counterbalance» to system of gelatinases. It serves as the reason of the remodeling in endomatrium with progressing of collagenogenesis and atrophy of the glandular device on a background against persistation activators of an infection. It can be one of factors in the loss of pregnancy genesis. The marked direct strong correlation between violations in the system «proteases-antiproteases» and the degree of activity of an inflammation in a mucous membrane of uterus allows to plan approaches to purposive medicamentous influence on some pathogenetic parts of loss of pregnancy, associated with chronic endometritis.Для оценки интенсивности и направленности процессов ремоделирования слизистой оболочки матки проведено иммуногистохимическое исследование биоптатов эндометрия с помощью моноклональных антител к желатиназам (ММР-2, ММР-9) и их ингибитору TIMP-1 у 30 женщин с хроническим эндометритом и невынашиванием беременности. Контролем служили биоптаты слизистой оболочки тела матки от 15 соматически и гинекологически здоровых женщин репродуктивного возраста, обратившихся для обследования перед планированием беременности или контрацепции. Результаты проведенного исследования эндометрия при невынашивании беременности, ассоциированном с хроническим эндометритом, свидетельствуют о снижении уровня экспрессии ММР-9 и ММР-2, а чакже повышении значений TIMP-1 как своеобразного «противовеса» системе желатиназ. Это служит причиной ремоделирования слизистой оболочки матки с прогрессированием коллагеногенеза и атрофии железистого аппарата на фоне персистенции возбудителей инфекции, что может явиться одним из факторов в генезе невынашивания беременности. Отмеченная прямая сильная корреляционная связь дисбаланса в системе «протеазы-антипротеазы» со степенью активности воспаления в слизистой оболочке матки позволяет наметить подходы к таргетному медикаментозному воздействию на некоторые патогенетические звенья невынашивания беременности, ассоциированного с хроническим эндометритом
