    Community core detection in transportation networks

    This work analyses methods for the identification and the stability under perturbation of a territorial community structure with specific reference to transportation networks. We considered networks of commuters for a city and an insular region. In both cases, we have studied the distribution of commuters' trips (i.e., home-to-work trips and viceversa). The identification and stability of the communities' cores are linked to the land-use distribution within the zone system, and therefore their proper definition may be useful to transport planners.Comment: 8 pages, 13 figure

    Objective: We sought to assess and compare the effects of myo-inositol combined with monacolin k (Group A) versus inositol only (Group B) and metformin (Group C), on the clinical, hormonal, metabolic and lipid profiles of insulin-resistant PCOS patients. Study design: 60 insulin resistant PCOS patients were randomly assigned into three groups. All groups were treated for 6 months with either myo-inositol and monacolin k, inositol only or metformin. Patients clinical, hormonal and metabolic profiles were compared between all groups, and within each group, prior and following the aforementioned treatment. Results: All treatment arms improved patients' clinical, hormonal and metabolic profiles, with a tendency toward better results in patients using the combination of myo-inositol and monacolin k. Conclusion: The combined treatment with the natural products: monacolin K and myo-inositol represents a valid and well tolerated alternative to the common medical therapy, improving the PCOS related symptoms, with minimal side effects

    Sperm quality improvement after natural anti-oxidant treatment of asthenoteratospermic men with leukocytospermia

    Aim: To study the immune-modulating and anti-oxidant effects of beta-glucan, papaya, lactoferrin, and vitamins C and E on sperm characteristics of patients with asthenoteratozoospermia associated with leucocytosis. Methods: Fifty-one patients referred to our Sterility Center for semen analysis were selected. Sperm parameters were assessed before and after patient's treatment with beta-glucan, lactoferrin, papaya, and vitamins C and E. DNA damage was assessed by the acridine orange test and sperm structural characteristics were evaluated by transmission electron microscopy. Results: After 90 days of treatment, an increase in the percentage of morphologically normal sperm (17.0 ± 5.2 vs. 29.8 ± 6.5) and total progressive motility (19.0 ± 7.8 vs. 34.8 ± 6.8) were detected. Structural sperm characteristics as well as chromatin integrity were also improved after treatment. In terms of leukocyte concentration in seminal fluid, a significant reduction was recorded (2.2 ± 0.9 vs. 0.9 ± 0.2). Conclusion: The treatment of an inflammatory process by the synergic action of immune modulators and anti-oxidants could protect sperm during maturation and migration, leading to improved sperm function

    An In-Silico Pipeline for Rapid Screening of DNA Aptamers against Mycotoxins: The Case-Study of Fumonisin B1, Aflatoxin B1 and Ochratoxin A

    Aptamers are single-stranded oligonucleotides selected by SELEX (Systematic Evolution of Ligands by EXponential Enrichment) able to discriminate target molecules with high affinity and specificity, even in the case of very closely related structures. Aptamers have been produced for several targets including small molecules like mycotoxins; however, the high affinity for their respective target molecules is a critical requirement. In the last decade, the screening through computational methods of aptamers for their affinity against specific targets has greatly increased and is becoming a commonly used procedure due to its convenience and low costs. This paper describes an in-silico approach for rapid screening of ten ssDNA aptamer sequences against fumonisin B1 (FB1, n = 3), aflatoxin B1 (AFB1, n = 2) and ochratoxin A (OTA, n = 5). Theoretical results were compared with those obtained by testing the same aptamers by fluorescent microscale thermophoresis and by magnetic beads assay for their binding affinity (KD) revealing a good agreement

    Characterization of the endophytic bacterial community of Bituminaria bituminosa plant grown in vitro and its interaction with the plant extract

    IntroductionBituminaria bituminosa is a medicinal plant recognized for its phytochemicals, such as furanocoumarins, pterocarpans, and flavonoids. Since the secondary metabolism is influenced by the plant-endophyte interactions, the endophytic bacterial community of B. bituminosa was explored and the possible interactions with the plant were described.Materials and methodsDifferent bacterial strains were isolated from different organs of in vitro plants as shoots, roots, and seeds. The bacterial strains were identified and phenotypically characterized for different traits; strains were also exposed to different concentrations of B. bituminosa plant extract showing different susceptibility, probably determined by different secondary metabolites produced by the plant in the different organs (i.e. aerial parts and roots).Results and discussionBacterial strains showed different phenotypic characteristics; the 6 detected haplotypes were dominated by a single species related to Stenotrophomonas rhizophila. Endophytes isolated from the aerial parts produced a higher indole-3-acetic acid (IAA) amount than those of the roots, while all strains were unable to produce biosurfactants and antagonistic activity toward the other strains. The research opens new perspectives for future analysis addressed to test the susceptibility of the endophytic bacterial community of B. bituminosa toward the pure compounds extracted from the plants, and to investigate the role of these compounds on the distribution of endophytes within the different plant tissues

    PCOS Physiopathology and Vitamin D Deficiency: Biological Insights and Perspectives for Treatment

    Recent literature has stressed the importance of vitamin D (VD) in polycystic ovary syndrome (PCOS). Women with PCOS are deficient in VD, particularly those with a higher weight. Hypovitaminosis is a risk factor for glucose intolerance, and reduced levels of VD is associated with insulin resistance and increased diabetes risk. Since women with PCOS and hirsutism seem to have lower levels of VD than women with PCOS without hirsutism, a correlation between VD deficiency and hyperandrogenism may be suggested. Interestingly, VD is crucial for many human physiological functions, including to counteract inflammation and oxidative stress. Some studies evaluated effects of VD supplementation on glucose homeostasis variables, hormonal status, lipid concentrations, and biomarkers of inflammation and oxidative stress among VD-deficient women. Moreover, VD has been shown to play a role in egg quality and fertility. This review aims to show the relationship between VD and the endocrine and metabolic profile of PCOS patients, as well as its implications for their fertility. The supplement of VD to the common therapy can lead to an improvement of the insulin resistance and lipid metabolism, a reduction of circulating androgens, as well as a better response to the induction of ovulation in PCOS women

    Impact of asymptomatic genital tract infections on in vitro Fertilization (IVF) outcome

    BACKGROUND: Infertility is estimated to affect approximately 9-30% of reproductive-aged couples. Several conditions involving one or both partners may contribute to infertility. The aim of this study is to evaluate the role of asymptomatic genital tract infections in the outcome of In Vitro Fertilization (IVF) in couples with infertility. METHODS: A total of 285 infertile couples were enrolled in the study. Vaginal/endocervical swabs and semen samples were collected and subjected to microbiological analysis. Spermiograms were carried out on semen specimens, and lactobacilli were quantified in vaginal swabs. Data were associated with IVF results and analysed by using non parametric tests and multivariate analysis. RESULTS: Microbiological analysis showed that 46.3% of couples presented with an asymptomatic genital tract infection. Spermiogram results showed a significantly diminished motility of sperm cells in samples positive to microbiological testing compared to negative specimens. Enterococcus faecalis was the most prevalent species (11.6%) in positive semen samples and was found to negatively affect both sperm morphology (p = 0.026) and motility (p = 0.003). Analysis of genital swabs from females showed that the presence of E. faecalis (p<0.0001), Escherichia coli (p = 0.0123), Streptococcus agalactiae (p<0.0001), and Gardnerella vaginalis (p = 0.0003) was significantly associated to reduced levels of vaginal lactobacilli. Association of microbiological data with IVF outcome showed that 85.7% of IVF+ couples was microbiologically negative, while IVF was successful in just 7.5% of couples infected with E. faecalis and/or U. urealyticum and/or M. hominis (p = 0.02). CONCLUSIONS: The results show the negative impact of E. faecalis on sperm quality and the association of definite bacterial pathogens with reduced levels of vaginal lactobacilli. The presence of E. faecalis and/or U. urealyticum and/or M. hominis in genital samples of infertile couples is predictive for a negative outcome of IVF

    Psychological adjustment and heart rate variability in ovarian cancer survivors

    Introduction: Body image, posttraumatic growth, quality of life, coping, and social support are relevant concepts to ovarian cancer survivors. This study aimed to examine the associations among these concepts as well as their relationships with heart rate variability (HRV), which is an index of vagal tone. Methods: an exploratory and correlational study was conducted on 25 ovarian cancer survivors. We used self-report measures to assess psychological variables. HRV parameters recorded for this study were analyzed in the time domain and in the frequency domain. Spearman correlations were performed. Results: Positive attitude coping strategy was associated with psychological and physical distress related to problems of appearance (Rho = -.57, p &lt; .01), emotional functioning (Rho = .53, p &lt; .01), and global health (Rho = .47, p &lt; .05). Problem solving coping strategy was correlated with a higher posttraumatic growth, namely greater personal strength (Rho = .44, p &lt; .05) and better relationships with others (Rho = .40, p &lt; .05). Seeking social support was associated with growth in relationships with others (Rho = .40, p &lt; .05). Higher HRV parameters were associated with higher physical functioning (SDNN: Rho = .59, p &lt; .01; RMSSD: Rho = .54; p &lt; .01; pNN50: Rho = .56, p &lt; .01; HF: Rho = .58, p &lt; .01). The ratio of low-frequency to high-frequency power (LF/HF) was negatively associated with posttraumatic growth (i.e., personal strength: Rho = .51, p &lt; .05; new possibilities: Rho = -.54, p &lt; .01). Discussion: Positive attitude and problem solving coping strategies may facilitate psychological adjustment to ovarian cancer. The strong association between markers of vagal tone and physical functioning offers insights on the possible role of vagus nerve in ovarian cancer survivors. These findings should be further investigated by future studies with larger samples and longitudinal designs
