    Fire truck relocation during major incidents

    The effectiveness of a fire department is largely determined by its ability to respond to incidents in a timely manner. To do so, fire departments typically have fire stations spread evenly across the region, and dispatch the closest truck(s) whenever a new incident occurs. However, large gaps in coverage may arise in the case of a major incident that requires many nearby fire trucks over a long period of time, substantially increasing response times for emergencies that occur subsequently. We propose a heuristic for relocating idle trucks during a major incident in order to retain good coverage. This is done by solving a mathematical program that takes into account the location of the available fire trucks and the historic spatial distribution of incidents. This heuristic allows the user to balance the coverage and the number of truck movements. Using extensive simulation experiments we test the heuristic for the operations of the Fire Department of Amsterdam‐Amstelland, and compare it against three other benchmark strategies in a simulation fitted using 10 years of historical data. We demonstrate substantial improvement over the current relocation policy, and show that not relocating during major incidents may lead to a significant decrease in performance

    ПЕРСПЕКТИВЫ И ТЕНДЕНЦИИ РАЗВИТИЯ ТЕРАГЕРЦОВЫХ ТЕХНОЛОГИЙ: ПАТЕНТНЫЙ ЛАНДШАФТ

    The article is dedicated to the analysis of areas where terahertz radiation is applicable with the hwlp of a patent papers review, as well as to obtaining knowledge about the invention activity changes in this area over the past 35 years.Orbit patent databases’ capabilities were used to complete this analysis. Data search was conducted by keywords “terahertz” or “THz.” The searches were neither limited by submission dates, by priorities, nor by the country. A comprehensive analysis will allow to determine a more than 7389 patent-analog families’ aplications submitted in the period between 1980 and 2017 years. Statistical processing of obtained documents has been completed with the assistance of program software, indicated in the database. It was revealed that terahertz technologies have significant prospects to be applied in various areas; a fact, that is confirmed by an identified range of fields, where electromagnetic radiation of terahertz spectrum is applicable and is constantly growing. The dynamics of patenting is characterized by annual stable positive growth in the number of applications for inventions in the researched area. The highest number of inventions was identified in the field of instrumentation technologies, optics, telecommunications, semiconductor technologies, and medicine. Additionally, there was analyzed the state of patenting over the course of last five years. Examples of inventions were given. There were identified leading countries and companies in the researched area.Статья посвящена анализу областей применения терагерцового излучения на основе обзора патентной документации, а также получению сведений об изменении изобретательской активности в этой области за последние 35 лет. Для анализа использовались возможности патентной базы данных Орбит. Поиск осуществлялся по ключевым словам “terahertz” или “THz”. Поиски не были ограничены ни датой подачи или приоритета, ни страной. Комплексный анализ позволил выявить более 7389 семейств патентов-аналогов, заявки на которые были поданы в период с 1980 по 2017 гг. Статистическая обработка найденных документов осуществлялась с помощью программного обеспечения указанной БД. Выявлено, что терагерцовые технологии имеют чрезвычайно широкие перспективы применения в различных отраслях, о чем свидетельствует выявленный спектр областей использования электромагнитного излучения терагерцового диапазона, который постоянно расширяется. Динамика патентования характеризуется ежегодным стабильным положительным приростом количества заявок на изобретения в исследуемой области. Наибольшее количество изобретений было выявлено в области измерительной техники, оптики, телекоммуникациях, полупроводниковой техники и медицине. Дополнительно проанализирована патентная ситуация в указанных областях за последние пять лет. Приведены примеры изобретений. Выявлены ведущие страны и компании в исследуемой области

    Altermagnetic lifting of Kramers spin degeneracy

    Lifted Kramers spin-degeneracy has been among the central topics of condensed-matter physics since the dawn of the band theory of solids. It underpins established practical applications as well as current frontier research, ranging from magnetic-memory technology to topological quantum matter. Traditionally, lifted Kramers spin-degeneracy has been considered to originate from two possible internal symmetry-breaking mechanisms. The first one refers to time-reversal symmetry breaking by magnetization of ferromagnets, and tends to be strong due to the non-relativistic exchange-coupling origin. The second mechanism applies to crystals with broken inversion symmetry, and tends to be comparatively weaker as it originates from the relativistic spin-orbit coupling. A recent theory work based on spin-symmetry classification has identified an unconventional magnetic phase, dubbed altermagnetic, that allows for lifting the Kramers spin degeneracy without net magnetization and inversion-symmetry breaking. Here we provide the confirmation using photoemission spectroscopy and ab initio calculations. We identify two distinct unconventional mechanisms of lifted Kramers spin degeneracy generated by the altermagnetic phase of centrosymmetric MnTe with vanishing net magnetization. Our observation of the altermagnetic lifting of the Kramers spin degeneracy can have broad consequences in magnetism. It motivates exploration and exploitation of the unconventional nature of this magnetic phase in an extended family of materials, ranging from insulators and semiconductors to metals and superconductors, that have been either identified recently or perceived for many decades as conventional antiferromagnets

    In order to identify potential inhibitors and modulators of thromboxane synthase, its molecular cloning, heterologous expression, isolation, and purification have been carried out. The recombinant protein is isolated in the homogenous state and possesses the functional activity. Peptide with an amino acids sequence SGVYKVLYDWQHGGF is identified using the random dodecapeptides phage library. It is revealed that this peptide has a great similarity to thrombopoietin peptidomimetic. This allows one to suppose the role of this hormone in the thromboxane synthase regulation.С целью поиска ингибиторов и модуляторов активности тромбоксан синтазы проведена гетерологическая экспрессия, выделение и очистка тромбоксан синтазы человека. Рекомбинантный белок получен в гомогенном состоянии и обладает функциональной активностью. С использованием фаговой библиотеки рандомизированных додекапептидов идентифицирован пептид с аминокислотной последовательностью SGVYKVLYDWQHGGF, обладающий высоким сродством к белку. Показано высокое сходство его последовательности с последовательностью пептидомиметика тромбопоэтина, что позволяет предположить роль этого гормона в регуляции активности тромбоксан синтазы

    ФАГОВЫЙ ДИСПЛЕЙ В КОНСТРУИРОВАНИИ АНТИТЕЛ С ЗАДАННЫМИ СВОЙСТВАМИ

    Antibodies are an essential tool being a part of immunodiagnostics, therapeutics and life science research instruments. In this paper we analyze the recent developments in recombinant antibodies generation by means of phage display technology. We also provide our own results dedicated to antibodies development process. Analysis of antibody-antigen complexes with resolved structures allow to create synthetic phage display library of Fab antibody fragments with the diversity of 1010 independent clones. Utilizing a negative selection approach, we succeeded in generation and characterization of specific single-domain antibodies that has substantially different binding kinetics to CYP11B2 and CYP11B1 isoenzymes – proteins with 93% sequence identity. We also generated specific binders to a few high-molecular weight human antigens – erytropoetin, growth hormone and thyroperoxidase.Уникальная особенность моноклональных антител высокоспецифично взаимодействовать с молекулярными мишенями позволила им занять ведущее положение в терапии онкологических и аутоиммунных заболеваний, стать незаменимым инструментом протеомных исследований и компонентом диагностических систем. Представлен обзор новейших литературных данных по использованию метода фагового дисплея для получения рекомбинантных антител, которые другими методами получить невозможно, а также собственные оригинальные результаты в этой области. На основании бионформационного анализа структур депонированных комплексов антител с антигенами нами создана комбинаторная библиотеки Fab фрагментов антител человека, обладающая разнообразием более 1010 независимых клонов и способная служить источником рекомбинантных антител. С использованием негативной селекции нами получены и характеризованы высокоспецифичные однодоменные антитела к альдостерон-синтазе (цитохром Р45011В2, CYP11B2), не обладающие в ИФА кросс-реактивностью с ее гомологом CYP11B1 (93% идентичности последовательностей), а также антитела к таким высокомолекулярным мишеням, как эритропоэтин, соматотропный гормон и тиреопероксидаза человека.
