    Low-volume jet injection for intradermal immunization in rabbits

    BACKGROUND: This study tested a low-volume (20–30 μl/20–30 μg DNA) jet injection method for intradermal delivery of a DNA vaccine. Jet injection offers the advantages of a needle-less system, low-cost, rapid preparation of the injected DNA solution, and a simple delivery system. More than one construct can be injected simultaneously and the method may be combined with adjuvants. RESULTS: Low-volume jet injection targeted delivery of a DNA solution exclusively to the dermis and epidermis of rabbits. A three injection series of plasmid DNA, encoding the Hepatitis B Surface Antigen stimulated a humoral immune response in 2/5 rabbits. One rabbit developed a significant rise in antibody titer after 1 injection and one following 2 injections. There were no significant differences between jet injection and particle bombardment in the maximal antibody titers or number of injections before response. A three injection series of the same plasmid DNA by particle bombardment elicited a significant rise in antibody titer in 3/5 rabbits. One rabbit developed antibody after 1 injection and two after 3 injections. In contrast, 0/5 rabbits receiving DNA by needle and syringe injection responded. In the jet injection and particle bombardment groups, gene expression levels in the skin did not predict response. While immune responses were similar, luciferase gene expression levels in the skin following particle bombardment were 10–100 times higher than jet injection. CONCLUSION: Low-volume jet injection is a simple, effective methodology for intradermal DNA immunization

    An integrated optical Bragg grating refractometer for volatile organic compound detection

    We report an integrated optical Bragg grating detector, fabricated using a direct UV-writing approach, that when coated with a thin-film of a hydrophobic siloxane co-polymer can perform as an all-optically accessed detector for hydrocarbon vapour. Upon exposure to a series of organic solvent vapours, both negative and positive Bragg wavelength shifts of differing magnitudes were measured. This was attributed to a combination of swelling and/or hydrocarbon solvent filling the free volume within the polymer film. A quantitative structural property relationship (QSPR) approach was utilised to create a multiple variable linear regression model, built from parameters that chemically described the hydrocarbons and the intermolecular interactions present between the co-polymer and hydrocarbon molecules. The resulting linear regression model indicated that the degree of swelling of the polysiloxane thin film when exposed to vapours of different hydrocarbons was due to the physico-chemical properties of the hydrocarbons and that this was the main causative factor of the measured Bragg wavelength shifts. Furthermore, this linear regression model allows for the prediction of the Bragg wavelength shift that would be measured upon exposure to vapours of another defined hydrocarbon. This detector is intrinsically safe in flammable environments. It includes on-chip thermal compensation, operates at telecoms wavelengths and has a predictable response to a variety of hydrocarbons making it ideal for detection of flammable hydrocarbon vapours in industrial and domestic processes

    Feasibility Study of the World Health Organization Health Care Facility-Based Antimicrobial Stewardship Toolkit for Low- and Middle-Income Countries

    Antimicrobial stewardship (AMS) has emerged as a systematic approach to optimize antimicrobial use and reduce antimicrobial resistance. To support the implementation of AMS programs, the World Health Organization developed a draft toolkit for health care facility AMS programs in low- and middle-income countries. A feasibility study was conducted in Bhutan, the Federated States of Micronesia, Malawi, and Nepal to obtain local input on toolkit content and implementation of AMS programs. This descriptive qualitative study included semi-structured interviews with national- and facility-level stakeholders. Respondents identified AMS as a priority and perceived the draft toolkit as a much-needed document to further AMS program implementation. Facilitators for implementing AMS included strong national and facility leadership and clinical staff engagement. Barriers included lack of human and financial resources, inadequate regulations for prescription antibiotic sales, and insufficient AMS training. Action items for AMS implementation included improved laboratory surveillance, establishment of a stepwise approach for implementation, and mechanisms for reporting and feedback. Recommendations to improve the AMS toolkit\u27s content included additional guidance on defining the responsibilities of the committees and how to prioritize AMS programming based on local context. The AMS toolkit was perceived to be an important asset as countries and health care facilities move forward to implement AMS programs
